It is shown how a multicompartmental infectious disease model can be systematically examined for reduction of structural complexity. For steadystate situations, four basic rules are proposed for eliminating components of flow-lines, whole flow-lines, and compartments, plus combining compartments. An application to a typhoid fever model allows calculations to be done on a pocket calculator. The approach could be particularly important in developing countries.  相似文献

Aleurone tissue from undried immature developing wheat grains (Triticum aestivum L. cv. Sappo), normally insensitive to gibberellic acid, can be made to respond to the hormone by a series of temperature treatments. Incubation of the de-embryoed grains at temperatures above 27° C for at least 8 h causes the tissue to become sensitive. Prolonged incubation at temperatures below 27° C does not effect a change in sensitivity. In addition to the requirement for exposure to an elevated temperature for a period of several hours the tissue must also subsequently be subjected to a period at a lower temperature for just a few seconds for the response to be observed. Once sensitized, the tissue remains responsive to gibberellic acid for substantial periods of time. Exposure of the tissue to temperatures which induce sensitivity to gibberellic acid also results in an increased leakage of amino acids. It is suggested that the increase in sensitivity to gibberellin requires two separate processes to take place. One could be a homeoviscous adaptation of the cell membranes in response to elevated temperature, the other a subsequent, permanent change in conformation of membrane components.  相似文献

Benzodiazepines are known to facilitate GABA-ergic transmission at synaptic sites, while sodium valproate is an anticonvulsant drug which is reported to elevate GABA levels in the brain. In order to determine whether these two drugs interact functionally at GABA receptor sites, graded doses of chlordiazepoxide (CDZ) and sodium valproate were injected bilaterally into the nucleus accumbens and their effect on the dopamine (DA)-induced stimulation of motor activity was studied. Both of these compounds, as well as GABA, produced an inhibition of the hyperactivity induced by the bilateral injection of DA into the nucleus accumbens. Bicuculline, the GABA receptor antagonist, blocked the effect of CDZ on the DA-induced hyperactivity. A low dose of CDZ (2 μg), which by itself did not significantly inhibit the effect of DA, potentiated the inhibition of the hyperactivity produced by valproate. These results suggest that CDZ and sodium valproate can interact functionally at GABA-ergic sites in the central nervous system.  相似文献

Summary UV- or -irradiated G2 phase cells of rad + Schizosaccharomyces pombe show increased inactivation if incubated post-irradiation, in liquid growth medium containing caffeine, before being plated on normal agar medium. The following, however, do not show such caffeine-induced lethality: G1 phase rad + cells; ascospores of a rad + strain; either G2 or G1 phase cells of the recombination-deficient rad1 strain; unirradiated rad + cells. Of the above, only the G2 phase rad + cells possess, at the time of radiation exposure, the capability for recombination. These results indicate that a recombinational process is responsible for caffeine-induced lethality after exposure to UV or ionizing radiation.Similarly, the negative liquid holding effect (a progressive inactivation seen if UV- or -irradiated cells are incubated in non-nutritive medium such as buffer before being plated) is manifested only in G2 phase rad + cells, and not in either G1 phase rad + cells or rad1 cell (whether G2 or G1 phase). Both the negative liquid holding effect and caffeine-induced lethality therefore are seen only in cells which fulfill all of the following conditions: (a) they must be genetically recombination-proficient; (b) they must possess at the time of irradiation the necessary two DNA copies with which to perform recombinational repair (for a haploid cell, this means they must be in G2 phase); (c) their DNA must be damaged, such as by UV or -ray exposure, thus requiring that recombinational repair capability be exercised in order to maintain viability; and (d) they must be incubated under conditions that fail to support the normal progress of recombinational repair. The exercising of recombinational repair capability has been shown to require an incubation medium capable of supporting growth. The incubation conditions that give rise to further inactivation of irradiated cells (non-nutritive liquid holding medium in the case of the negative liquid holding effect and exposure to caffeine in the case of caffeine-induced lethality) have been demonstrated not to support recombinational repair.AECL Reference No. 7182  相似文献

A passive hemagglutination assay was developed to measure Trypanosoma musculi-specific antibody in mice. Indicator-erythrocyte donor mice received 550 rad 60Co 24 hr before intraperitoneal injection of 3 × 104T. musculi. T. musculi antigen-coated erythrocytes were obtained from these mice on Day 9 postinfection. T. musculi antigen-coated erythrocytes obtained in this manner were used as indicator erythrocytes in a passive hemagglutination procedure. Serum from hyperimmunized mice (three consecutive infections at 21-day intervals) gave titers as high as 1:1024. Titers of 1:256 and 1:512 were obtained from singly infected mice on Days 18 and 28 postinfection, respectively. In marked contrast, nude mice infected with T. musculi did not produce a detectable agglutinating antibody response. Erythrocytes obtained from either irradiated (550 rad 60Co) uninfected mice, nonirradiated infected mice, or normal mice did not agglutinate when combined with any of the sera tested. These data suggest the usefulness of this passive hemagglutination assay for the measurement of antibody to T. musculi in the serum of infected mice.  相似文献

Neonatal rats injected with either thyroxine (T4) or vehicle (NaOH) on postnatal Days 1, 2, and 3 were given classical-conditioning pairings of an odor with footshock when 7, 9, or 11 days of age. In accord with the conventional acceleration of other indices of maturation following the T4 treatment, 24-hr retention of the conditioned odor aversion was substantially enhanced among the 11 day-old rats given the earlier T4 treatment. This effect was marginally significant among 9-day olds but not among 7-day olds.  相似文献

The role of four forms of cytochrome P-450 from rabbit liver in the metabolic activation of two suspected carcinogens, 2-aminoanthracene and benz[α]pyrene, was investigated with a S. typhimurium tester strain, TA 98. Each of the forms, 2,3,4 and 6 was reconstituted with NADPH-cytochrome P-450 reductase and lipid, and assay conditions were established such that the cytochrome P-450 concentration was rate-limiting. Under these conditions, cytochrome P-450 form 4, but not the other forms, converted 2-aminoanthracene into a potent mutagen. In contrast, form 6 was the only form which metabolized benz[α]pyrene to a mutagen. These results indicate that specific cytochrome P-450 forms preferentially activate particular mutagens.  相似文献

Utilizing skin grafting techniques, melanocytes were incorporated into adult steel (SlSld) and dominant spotting (WWv) hair follicles, where they survived and pigmented a number of hair generations. Previous investigations have indicated that the steel alleles act on the embryonic skin to prevent the survival of melanocytes. The present study suggests that this gene action mechanism may be temporary and does not continue into adult life. In addition, the study demonstrates that some hair bulb melanocytes can pigment more than one hair generation.  相似文献
