An in vitro biological screening of Malaysian plants allowed the selection of several species with a significant binding affinity for the antiapoptotic protein Bcl-xL. The chemical investigation of Endiandra kingiana led to the isolation of a series of polyketides named kingianins A-N, having a pentacyclic carbon skeleton described for the first time in nature. Fourteen compounds were isolated as racemic mixtures, and characterized by mass spectrometryand extensive one- and two-dimensional NMR spectroscopy. The (-) and (+) enantiomers of kingianins A and G-L were separated using chiral HPLC, and the absolute configuration of four of them was clearly established by CD analysis. The levorotatory enantiomers showed the more potent binding affinity for Bcl-xL with Ki ranging from 1.0 to 12μM.  相似文献

The essential oils from the leaves and rhizomes of Alpinia pahangensis Ridl ., collected from Pahang, Peninsular Malaysia, were obtained by hydrodistillation, and their chemical compositions were determined by GC and GC/MS analyses. The major components of the rhizome oil were γ‐selinene (11.60%), β‐pinene (10.87%), (E,E)‐farnesyl acetate (8.65%), and α‐terpineol (6.38%), while those of the leaf oil were β‐pinene (39.61%), α‐pinene (7.55%), and limonene (4.89%). The investigation of the antimicrobial activity of the essential oils using the broth microdilution technique revealed that the rhizome oil of A. pahangensis inhibited five Staphylococcus aureus strains with minimum inhibitory concentration (MIC) values between 0.08 and 0.31 μg/μl, and four selected fungi with MIC values between 1.25 and 2.50 μg/μl.  相似文献

Epinephelus fuscoguttatus is a commercially important marine fish species in southeast Asia. Due to overfishing and water pollution, this species has been declared as near-threatened. Thus, to provide information to help maintain and preserve the species, microsatellites were developed, using an enriched genomic library method. Thirty individuals were collected from the hatchery of the Fishery Research Institute, Terengganu, Malaysia. These individuals, from four to six years old, originated from Sabah and are maintained in captive culture as broodstock. Genomic DNA was extracted from the fins of selected individuals that weighed 3-8 kg. Ten microsatellite loci were found to be polymorphic in this population, with 5 to 21 alleles per locus. Observed and expected heterozygosities ranged from 0.53 to 0.97 and 0.59 to 0.95, respectively. Only one locus deviated significantly from Hardy-Weinberg equilibrium and no significant linkage disequilibrium was found among the pairs of loci. These polymorphic microsatellite loci will be used by the Malaysian Fishery Research Institute for investigating genetic diversity and for developing breeding strategies.  相似文献

Chisocheton is one of the genera of the family Meliaceae and consists of ca. 53 species; the distribution of most of those are confined to the Indo‐Malay region. Species of broader geographic distribution have undergone extensive phytochemical investigations. Previous phytochemical investigations of this genus resulted in the isolation of mainly limonoids, apotirucallane, tirucallane, and dammarane triterpenes. Reported bioactivities of the isolated compounds include cytotoxic, anti‐inflammatory, antifungal, antimalarial, antimycobacterial, antifeedant, and lipid droplet inhibitory activities. Aside from chemistry and biological activities, this review also deals briefly with botany, distribution, and uses of various species of this genus.  相似文献

Nucleocapsid (N) protein of Nipah virus (NiV) is a potential serological marker used in the diagnosis of NiV infections. In this study, a rapid and efficient purification system, HisTrap? 6 Fast Flow packed bed column was applied to purify recombinant histidine-tagged N protein of NiV from clarified feedstock. The optimizations of binding and elution conditions of N protein of NiV onto and from Nickel Sepharose? 6 Fast Flow were investigated. The optimal binding was achieved at pH 7.5, superficial velocity of 1.25 cm/min. The bound N protein was successfully recovered by a stepwise elution with different concentration of imidazole (50, 150, 300 and 500 mM). The N protein of NiV was captured and eluted from an inlet N protein concentration of 0.4 mg/ml in a scale-up immobilized metal affinity chromatography (IMAC) packed bed column of Nickel Sepharose? 6 Fast Flow with the optimized condition obtained from the method scouting. The purification of histidine-tagged N protein using IMAC packed bed column has resulted a 68.3% yield and a purification factor of 7.94.  相似文献

In aseasonal tropics, timing of leaf emergence and leaf fall may differ between the shoots of different crown parts within a tree. This is important for the efficient development of crowns because leaves should be produced as soon as enough carbohydrates are accumulated. This hypothesis was tested by investigating leaf demography over a 44-mo period for 17 Malaysian trees and comparing the timings of leaf emergence and fall between the upper and lower crowns. The timings of leaf emergence were synchronized between the upper and lower crowns, but those of leaf fall were less synchronized in most trees. Greater rates of leaf production in the upper than in the lower crowns were attributable to the differences in the number of leaves that emerged per leaf emergence event, rather than differences in frequency of leaf emergence per year. Timings of leaf emergence and leaf fall were mainly simultaneous in the upper and lower crowns, but unsynchronized leaf production and leaf fall also occurred. Such limited plasticity of leaf demography within crowns may be the result of physiological integration of branches or the compromise between the advantages of satiating herbivores and effective crown development in the trees of aseasonal tropics.  相似文献

The esterification of palm-based 9,10-dihydroxystearic acid (DHSA) and 1-octanol in hexane as catalyzed by lipase from Rhizomucor meihei (Lipozyme IM) followed Michaelis-Menten kinetics. The esterification reaction follows a Ping-Pong, Bi-Bi mechanism. The maximum rate was estimated to be 1 micromol min(-1) mg(-1) catalyst in hexane at 50 degrees C, and the Michaelis-Menten constants for DHSA and 1-octanol were 1.3 M and 0.7 M, respectively.  相似文献

Exposure to toxic metals and excessive amount of trace elements is a risk factor of cognitive decline. Continuous monitoring of these elements by the use of metabolically inactive tissues such as fingernails may help in taking preventive measures to delay the cognitive decline process. In this study, the cognitive function of 54 elderlies (60–72 years old) from FELDA Sungai Tengi, Selangor, was evaluated using the Malay version of Mini-Mental State Examination (MMSE) and Montreal Cognitive Assessment (MoCA). The levels of arsenic (As), cadmium (Cd), copper (Cu), manganese (Mn), lead (Pb), and zinc (Zn) in fingernail were detected using Inductively Coupled Plasma-Mass Spectrometry. Results showed that (92.6 %) our population was cognitively impaired based on the MoCA with mean score of 18.07?±?5.11. The mean level of elements in fingernails for As, Cd, Cu, Mn, Pb, and Zn were 16.68?±?3.52, 4.26?±?0.91, 4.81?±?1.16, 1.00?±?0.23, 1.83?±?0.47, and 40.86?±?10.81 μg/g, respectively. Significant inverse correlations were observed between MoCA with age (r?=??0.543, p?<?0.01), Cu level (r?=??0.330, p?<?0.05), and Mn level (r?=??0.496, p?<?0.01). MMSE was correlated to age (r?=??0.346, p?<?0.05) only. In conclusion, besides aging, the progression of cognitive decline among the elderly FELDA population was associated with increased levels of copper and manganese.  相似文献

This report highlights the first successful cryopreservation protocol for shoot tips of Garcinia mangostana L. achieved by using vitrification technique. We investigated the effects of different temperatures and exposure periods to a plant vitrification solution 2 (PVS2), sucrose concentrations and preculture periods, and unloading treatments in steps of the vitrification protocol on the survival of G. mangostana shoot tips after cryopreservation. Exposure to PVS2 for 25 min gave beneficial effects with 10.4 ± 1.8 % survival at 0 °C with average water content of 1.1 ± 0.3 g g?1 dry mass. Survival was 13.7 ± 5.5 % when using preculture medium with full-strength Murashige and Skoog (MS) medium supplemented with 0.6 M sucrose for 2 days. A significant difference was observed in survival of shoot tips when treated with various sucrose concentrations in preculture which strengthens their importance towards enhancing survival of shoot tips after cryopreservation. MS with 0.4 M sucrose and 2 M glycerol applied as an unloading solution increased the survival of shoot tips to 44.1 ± 6.5 %. Experiments on the effect of ascorbic acid were also conducted for each step of vitrification. Our results showed higher survival of 45.8 ± 3.8 % but there were no significant effects compared with the control (without ascorbic acid). Further study on the recovery dark/light period was conducted. Survival of shoot tips significantly increased to 50.0 ± 16.7 % when subjected to 7 days in the dark before transferring to 16 h/8 h light/dark photoperiod. These studies strengthen suggestions that cryopreservation through vitrification is possible for ex situ conservation of germplasm of this tropical recalcitrant species.  相似文献
