High biodiversity is known to increase many ecosystem functions, but studies investigating biodiversity effects have more rarely looked at multi‐trophic interactions. We studied a tri‐trophic system composed of Centaurea jacea (brown knapweed), its flower head‐infesting tephritid fruit flies and their hymenopteran parasitoids, in a grassland biodiversity experiment. We aimed to disentangle the importance of direct effects of plant diversity (through changes in apparency and resource availability) from indirect effects (mediated by host plant quality and performance). To do this, we compared insect communities in C. jacea transplants, whose growth was influenced by the surrounding plant communities (and where direct and indirect effects can occur), with potted C. jacea plants, which do not compete with the surrounding plant community (and where only direct effects are possible). Tephritid infestation rate and insect load, mainly of the dominant species Chaetorellia jaceae, decreased with increasing plant species and functional group richness. These effects were not seen in the potted plants and are therefore likely to be mediated by changes in host plant performance and quality. Parasitism rates, mainly of the abundant chalcid wasps Eurytoma compressa and Pteromalus albipennis, increased with plant species or functional group richness in both transplants and potted plants, suggesting that direct effects of plant diversity are most important. The differential effects in transplants and potted plants emphasize the importance of plant‐mediated direct and indirect effects for trophic interactions at the community level. The findings also show how plant–plant interactions critically affect results obtained using transplants. More generally, our results indicate that plant biodiversity affects the abundance of higher trophic levels through a variety of different mechanisms. 相似文献

We show in this paper that a complex constituted by phospholipids and LHI and LHII α polypeptides was inserted in a heavy
membrane fraction in a nonextractable form, indicating a transmembrane localization. The best accepting membranes originated
from aerobically grown cells. Addition of ATP during the insertion inhibited this reaction 25 to 30% in heavy membranes isolated
from aerobically grown cells (HMaer) and a higher inhibition (60 to 65%) was detected when using heavy membranes isolated from photosynthetically grown cells
(HMpho). Purification by gel filtration of a crude Na2CO3 extract yielded three phosphate-labeled fractions. Two of them contained protein and phospholipids in a stable association.
However, only fractions containing phosphatidylethanolamine were shown to be reconstituted. The third radioactive fraction
contained labeled ATP and protein, but no phospholipids and could not be reassociated to the heavy membranes of any origin.
A model for the insertion of the LH polypeptides is presented in which the recently synthesized polypeptides are phosphorylated
and become associated to anionic phospholipids. The interaction of this complex to the membrane spontaneously leads to stable
insertion.

The retroviral integrase (IN) is required for the integration of viral DNA into the host genome. The N terminus of IN contains an HHCC zinc finger-like motif, which is conserved among all retroviruses. To study the function of the HHCC domain of Moloney murine leukemia virus IN, the first N-terminal 105 residues were expressed independently. This HHCC domain protein is found to complement a completely nonoverlapping construct lacking the HHCC domain for strand transfer, 3′ processing and coordinated disintegration reactions, revealing trans interactions among IN domains. The HHCC domain protein binds zinc at a 1:1 ratio and changes its conformation upon binding to zinc. The presence of zinc within the HHCC domain stimulates selective integration processes. Zinc promotes the dimerization of the HHCC domain and protects it from N-ethylmaleimide modification. These studies dissect and define the requirement for the HHCC domain, the exact function of which remains unknown. 相似文献

An important feature of glucose homeostasis is the effective release of glucagon from the pancreatic alpha cell. The molecular mechanisms regulating glucagon secretion are still poorly understood. We now demonstrate that human alpha cells express ionotropic glutamate receptors (iGluRs) that are essential for glucagon release. A lowering in glucose concentration results in the release of glutamate from the alpha cell. Glutamate then acts on iGluRs of the AMPA/kainate type, resulting in membrane depolarization, opening of voltage-gated Ca(2+) channels, increase in cytoplasmic free Ca(2+) concentration, and enhanced glucagon release. In vivo blockade of iGluRs reduces glucagon secretion and exacerbates insulin-induced hypoglycemia in mice. Hence, the glutamate autocrine feedback loop endows the alpha cell with the ability to effectively potentiate its own secretory activity. This is a prerequisite to guarantee adequate glucagon release despite relatively modest changes in blood glucose concentration under physiological conditions. 相似文献

Exercise enhances insulin-stimulated glucose transport (GT) in skeletal muscle. Evidence suggests that 5' AMP-activated protein kinase (AMPK) and glycogen may be important for enhanced insulin sensitivity. Our goals were to investigate the effect of various in situ muscle contraction protocols on insulin-stimulated GT and assess the relationship of contraction-induced changes in AMPK and glycogen with postcontraction improvement in insulin-stimulated GT. Rats were anesthetized, both ulnar nerves were exposed, and one nerve was electrically stimulated to contract forelimb muscles. We performed a series of five experiments, sequentially varying only one contraction parameter (train duration, train rate, pulse frequency, number of 5-min bouts, or pulse duration) while holding the others constant. Both epitrochlearis muscles were dissected out and incubated for 3.5 h before measurement of GT. For each contraction parameter studied, we identified an apparent threshold value that did not induce a significant increase in insulin-stimulated GT and an apparent peak value, above which there was a plateau or decline in insulin-stimulated GT. Using other rats, we evaluated muscle AMPK phosphorylation and glycogen concentration immediately postcontraction. AMPK phosphorylation and reduction in glycogen were increased compared with resting controls in each protocol, which had previously been shown to increase insulin-stimulated GT, as well as in several protocols that did not significantly increase insulin-stimulated GT. These data suggest that contraction-induced AMPK phosphorylation and decrease in glycogen may be necessary but are not sufficient for the postcontraction increase in insulin-stimulated GT in rat skeletal muscle. 相似文献

Nineteen plants from the Republic of Panama were selected by their traditional uses in the treatment of hypertension, cardiovascular, mental and feeding disorders and 149 extracts were screened using radioligand-receptor-binding assays. The methanol:dicloromethane extracts of the bark and leaves of Anacardium occidentale L., the leaves of Begonia urophylla Hook., the roots of Bocconia frutescens L., the stems and leaves of Cecropia cf.obtusifolia Bertol., the branches of Clusia coclensis Standl., the bark of Cochlospermum vitifolium (Willd.)Spreng., the roots of Dimerocostus strobilaceus Kuntze, the bark of Guazuma ulmifolia Lam., the leaves of Persea americana Mill. and the branches of Witheringia solanaceae L'Her. inhibited the [3H]-AT II binding (angiotensin II AT1 receptor) more than 50%. Only extracts of the roots of Dimerocostus strobilaceus Kuntze and the stems of Psychotria elata (Sw.) Hammel were potent inhibitors of the [3H] NPY binding (neuropeptide Y Y1 receptor) more than 50% and the ethanolic extracts of the leaves of Cecropia cf. obtusifolia Bertol., the leaves of Hedyosmum bonplandianum H.B.K., the roots of Bocconia frutescens L., the stem of Cecropia cf. obtusifolia Bertol. and the branches of Psychotria elata (Sw.) Hammel showed high inhibition of the [3H] BQ-123 binding (endothelin-1 ET(A) receptor) in a preliminary screening. The distribution pattern of C3 and C4 grasses was studied in eight sites located between 350 m and 2100 m along an altitudinal gradient in Central Argentina. Of 139 taxa fifty-nine are C3 and eighty C4 . Species of the C3 tribes (Stipeae, Poeae, Meliceae, Aveneae, Bromeae and Triticeae) and C3 Paniceae species increase in number at higher elevations; only one C3 species was found below 650 m. C4 Aristideae, Pappophoreae, Eragrostideae, Cynodonteae, Andropogoneae and Paniceae increase at lower altitudes. The floristic crossover point is at about 1500 m; the ground cover cross-over point is at about 1000 m. Analysis of the relationships between % C4 species along the gradient and nine climatic and environmental variables showed the highest correlation with July mean temperature, but all temperature variables show highly significant correlations with % C4 . Correlation with annual rainfall is lower but also significant. These results are consistent with previous research showing the relative importance of C4 grasses as temperature increases. C3 species make a high contribution to relative grass coverage below the C3 /C4 floristic crossover point but are rare below 1000 m. 相似文献
