Investigation on the substrate specificity of human plasmin using tripeptidyl-p-nitroanilide substrates

The hydrolysis of 35 tripeptidyl-p-nitroanilides was studied with human plasmin and the kinetic parameters were determined. The individual contribution of the various side chains to the kinetic parameters was calculated by regression analysis. Considering Km, substrates having Z-D-Ile-Phe-Lys as well as H-D-Ile-Phe-Lys sequences were found to be the best, while Bz-Ile-Leu-Lys and pGlu-Leu-Lys sequences are the best for kcat. The Km values of substrates protected at N-terminus are lower, their kcat values are higher than those of the unprotected ones with the same sequence.     

Cyclic AMP stimulates dephosphorylation of specific proteins in intact S49 mouse lymphoma cells

Two-dimensional gel electrophoresis of proteins labeled with ³²P₁ in S49 mouse lymphoma cells revealed five phosphoproteins that were rapidly and reversibly dephosphorylated in response to elevation of cyclic AMP (cAMP). Under basal conditions, labeling of at least two of these proteins was limited by slow turnover of protein-bound phosphate. The rapid cAMP-mediated dephosphorylation of these species was attributable, therefore, to stimulation of a specific phosphoprotein phosphatase.     

Immunoelectron microscopic studies on the localization of peptidoglycan peptide subunit pentapeptides in bacterial cell walls

The peptide subunit pentapeptide H-L-Ala-D-Glu(L-Lys-D-Ala-D-Ala-OH)-NH₂ of peptidoglycan was localized in the cell walls of several Gram-positive bacteria employing the indirect immunoferritin technique. Specific antibodies to the D-alanyl-D-alanine moiety of non-crosslinked peptide subunit pentapeptide were raised in rabbits by immunization with synthetic immunogen albumin-(CH₂CO-Gly-L-Ala-L-Ala-D-Ala-D-Ala-OH)₃₉. Specificity of these antibodies for the peptide subunit pentapeptide and not for the peptide subunit tetrapeptide was corroborated in a Farr-type radio-active hapten binding assay. Specificity of labelling with ferritin was established by immunoelectron microscopic controls, and by the excellent correlation between specific labelling of cells with ferritin and the particular peptidoglycan primary structure of bacterial strains investigated. Cells of Lactobacillus gasseri, Streptococcus pyogenes and Staphylococcus aureus revealing non-crosslinked peptide subunit pentapeptides in their peptidoglycans could specifically be labelled. Covalent immobilization of the multienzyme enniatin synthetase

Summary The multienzyme enniatin synthetase was covalently immobilized to N-hydroxysuccinimide activated agarose. The stability of the immobilized enzyme at 25°C was enhanced compared to the soluble enzyme. Immobilization experiments also indicated that the enniatins are synthesized by a single molecule and thus do not require interactions of several enzyme molecules.     

Anaerobic degradation of 3,4,5-trimethoxybenzoate by a defined mixed culture of Acetobacterium woodii, Pelobacter acidigallici, and Desulfobacter postgatei

Abstract Acetobacterium woodii was continuously grown on 3,4,5-trimethoxybenzoate as pure culture or in commensalistic combination with Pelobacter acidigallici and Desulfobacter postgatei . Under pure culture conditions the following growth parameters were determined: μ _max= 0.112 h⁻¹, K _s= 1.07 mM, Y _max= 35 g/mol, and m = 0.22 mmol·g⁻¹·h⁻¹. In coculture with P. acidigallici the affinity for the substrate increased and the K _s value was found to be 135 μM. Under batch culture conditions mixed populations of A. woodii, P. acidigallici , and D. postgatei completely mineralized 3,4,5-trimethoxybenzoate to CO₂, whereas under continuous culture conditions more than 3 mM acetate remained unused.

Zusammenfassung Süßwasserschwämme (Spongilliden) lassen sich in 120-1-Aquarien mit Herkunftswasser der Schwämme kultivieren. Das gut durchlüftete Wasser soll nicht wärmer als 17°C sowie mit den Zusätzen Aquatonic und Schwarztorf versehen sein. Anstelle des Teichbzw. Flußwassers kann auch Leitungswasser oder eine Mischung von Leitungs- und vollentsalztem Wasser verwendet werden. Das Wasser der Aquarien ist alle vier Monate zu erneuern.Als Kulturgefäße eignen sich Glas- und Kunststoffschalen. Diese werden mittels spezieller Halter in die Aquarien eingebracht.Langzeitkulturen bieten beste Voraussetzungen für die Schwammforschung.

Permanent cultures of freshwater sponges (Porifera, Spongillidae) in the laboratory

Summary Freshwater sponges (Spongillidae) can be cultivated in 120-1 aquariums with water from the natural sponge habitats. The well-aired water should not be warmer than 17°C and should contain Aqua-tonic and black peat. Instead of habitat water, tap water or a mixture of tap and distilled water may be used. The water in the aquariums should be renewed every 4 months.For the culture we use glass or plastic vessels. These are mounted in the aquariums by special holders.Permanent cultures offer ideal conditions for sponge research.

     

Detection of platelet-associated IgG in chronic immune thrombocytopenic purpura using antibody-coated polyacrylamide beads

Platelet-associated IgG (PAIgG) was detected by means of anti-human IgG coated polyacrylamide beads ("Immunobeads") technique in 32 patients with chronic ITP. Both a direct test (with in vivo sensitized platelets) and an indirect test (with in vitro loaded platelets) were carried out. The percent of rosette forming beads was both in the direct test (41.2%) and in the indirect test (32.6%) significantly higher in the cases of chronic ITP patients than in the controls (2.5% and 3.2%, respectively). These results confirm the diagnostic value of this new, relatively simple and rapid method in routine clinical practice.     

Comparative Characterization of Glutamate Decarboxylase in Crude Homogenates of Oviduct, Ovary, and Hypothalamus

Some biochemical characteristics of L-glutamate decarboxylase (GAD) were compared using crude homogenates of the rat oviduct, ovary, and hypothalamus. As estimated by the measurement of CO2 production, the specific activities of oviductal and ovarian GAD were about 10 and 3% of the hypothalamic value, respectively. Stoichiometric formation of gamma-aminobutyric acid (GABA) and CO2 from L-glutamate could be observed in oviduct and hypothalamus, while in ovarian homogenates the production of CO2 was more than nine times that of GABA. Hypothalamic and tubal GAD showed similar time course, temperature dependence, and pH dependence. The dependence on protein concentration and on exogenous cofactor supply was also similar in these two tissues. The kinetic constant for L-glutamate as a substrate was nearly the same in oviduct (1.30 mM) and hypothalamus (1.64 mM). The responsiveness of tubal and hypothalamic GAD to various inhibitors was also similar. The present findings indicate that the oviductal and the hypothalamic GAD may be identical, and they suggest a possible GABAergic innervation of the Fallopian tube.     

From icons to symbols: Some speculations on the origins of language

This paper is divided into three sections. In the first section we offer a retooling of some traditional concepts, namely icons and symbols, which allows us to describe an evolutionary continuum of communication systems. The second section consists of an argument from theoretical biology. In it we explore the advantages and disadvantages of phenotypic plasticity. We argue that a range of the conditions that selectively favor phenotypic plasticity also favor a nongenetic transmission system that would allow for the inheritance of acquired characters. The first two sections are independent, the third depends on both of them. Cyclic nucleotide phosphodiesterases in larval brain of wild type and dunce mutant strains of Drosophila melanogaster: isoenzyme pattern and activation by Ca2+/calmodulin

Like adult heads and whole flies, larval brains of wild type Drosophila melanogaster contain two major soluble cyclic nucleotide phosphodiesterases, forms I and II. Larval brains of the learning-defective mutant strain, dunceM11, contain only the form I enzyme. In both wild type and dunce strains the form I enzyme is activated by Ca2+/calmodulin. A time-dependent loss of this Ca2+ activation was observed.
