Constructed tide tanks were used to examine the accumulation and distribution of heavy metals in various components of a simulated mangrove ecosystem. Young Kandelia candel plants grown in mangrove soils were irrigated with wastewater of various strengths twice a week for a period of one year. The amounts of heavy metals released via tidal water and leaf litter were monitored at regular time intervals. The quantities of heavy metals retained in mangrove soil and various plant parts were also determined. Results show that most heavy metals from wastewater were retained in soils with little being uptake by plants or released into tidal seawater. However, the amounts of metals retained in plants on a per unit dry weight base were higher than those in soils as the biomass production from the young mangrove plants was much smaller when compared to the vast quantity of soils used in this study. A significantly higher heavy metal content was found in roots than in the aerial parts of the mangrove plant,indicating that the roots act as a barrier for metal translocation and protect the sensitive parts of the plant from metal contamination. In both soil and plant, concentrations of Zn, Cd, Pb and Ni increased with the strengths of wastewater, although the bioaccumulation factors for these metals decreased when wastewater strengths increased. These results suggest that the mangrove soil component has a large capacity to retain heavy metals, and the role of mangrove plants in retaining metals will depend on plant age and their biomass production. Group II introns are large autocatalytic RNAs found in organellar genomes of plants and lower eukaryotes, as well as in some bacterial genomes. Interestingly, these ribozymes share characteristic traits with both spliceosomal introns and non-LTR retrotransposons and may have a common evolutionary ancestor. Furthermore, group II intron features such as structure, folding and catalytic mechanism differ considerably from those of other large ribozymes, making group II introns an attractive model system to gain novel insights into RNA biology and biochemistry. This review explores recent advances in the structural and mechanistic characterization of group II intron architecture and self-splicing.  相似文献

Herbivore species sharing a host plant often compete. In this study, we show that host plant-mediated interaction between two insect herbivores-a generalist and a specialist-results in a sex ratio shift of the specialist's offspring. We studied demographic parameters of the specialist Tupiocoris notatus(Hemiptera: Miridae)when co-infesting the host plant Nicotiana attenuata(Solanaceae) with the generalist leafhopper Empoasca sp.(Hemiptera: Cicadellidae). We show that the usually female-biased sex ratio of T. notatus shifts toward a higher male proportion in the offspring on plants coinfested by Empoasca sp. This sex ratio change did not occur after oviposition, nor is it due differential mortality of female and male nymphs. Based on pyrosequencing and PCR of bacterial 16 S rRNA amplicons, we concluded that sex ratio shifts were unlikely to be due to infection with Wolbachia or other known sex ratio-distorting endosymbionts. Finally, we used transgenic lines of N.attenuata to evaluate if the sex ratio shift could be mediated by changes in general or specialized host plant metabolites. We found that the sex ratio shift occurred on plants deficient in two cytokinin receptors(irCHK2/3).Thus, cytokinin-regulated traits can alter the offspring sex ratio of the specialist T.notatus.  相似文献

Trypanosoma cruzi (T. cruzi), the etiological agent of Chagas' disease, causes cardiac alterations in the host. Although the main clinical manifestations arise during the chronic stage, the mechanisms leading to heart damage develop early during infection. In fact, an intense inflammatory response is observed from acute stage of infection. Recently, peroxisome proliferator-activated receptors (PPARs) have attracted research interest due to their participation in the modulation of inflammation. In this work we addressed the role of 15-Deoxy-?12,14 ProstaglandinJ2 (15dPGJ2), a PPARγ natural ligand in the regulation of inflammatory mediators, in acute and chronic experimental mouse models of Chagas' disease with the RA and K98 T. cruzi strains, respectively. This work demonstrates that 15dPGJ2 treatment inhibits the expression and activity of inducible nitric oxide synthase (NOS2) as well as TNF-α and IL-6 mRNA levels. Also, expression and activity of metalloproteinases 2 (MMP-2) and 9 (MMP9) were inhibited by 15dPGJ2. Moreover GW9662, a specific PPARγ antagonist, revealed the participation of other signaling pathways since, in GW9662 presence, 15dPJG2 had a partial effect on the inhibition of inflammatory parameters in the acute model of infection. Accordingly, NF-κB activation was demonstrated, assessing p65 nuclear translocation in the hearts of infected mice with both T. cruzi strains. Such effect was inhibited after 15dPGJ2 treatment. Our findings support the concept that in vivo PPARγ and NF-κB pathways are implicated in the inhibitory effects of 15dPGJ2 on inflammatory mediators at different times depending on whether the infection is caused by the lethal or non-lethal T. cruzi strain.  相似文献

Epigenetic mechanisms, including DNA methylation, are important determinants in development and disease. There is a need for technologies capable of detecting small variations in methylation levels in an accurate and reproducible manner, even if only limited amounts of DNA are available (which is the case in many studies in humans). Quantitative methylation analysis of minute DNA amounts after whole bisulfitome amplification (qMAMBA) has been proposed as an alternative, but this technique has not been adequately standardized and no comparative study against conventional methods has been performed, that includes a wide range of methylation percentages and different target assays. We designed an experiment to compare the performance of qMAMBA and bisulfite-treated genomic (non-amplified) DNA pyrosequencing. Reactions were performed in duplicate for each technique in eight different target genes, using nine artificially constructed DNA samples with methylation levels ranging between 0% and 100% with intervals of 12.5%. Cubic polynomial curves were plotted from the experimental results and the real methylation values and the resulting equation was used to estimate new corrected data points. The use of the cubic regression-based correction benefits the accuracy and the power of discrimination in methylation studies. Additionally, dispersion of the new estimated data around a y = x line (R2) served to fix a cutoff that can discriminate, with a single 9-point curve experiment, whether whole bisulfitome amplification and subsequent qMAMBA can produce accurate methylation results. Finally, even with an optimized reagent kit, DNA samples subjected to whole bisulfitome amplification enhance the preferential amplification of unmethylated alleles, and subtle changes in methylation levels cannot be detected confidently.  相似文献

This paper analyzes increasingly individualized herding behavior after the implementation of a grazing ban policy in northern China based on empirical research in 12 pastoralist villages. The findings reveal that de-collectivization of pastureland has not necessarily led to direct changes in individual land use strategies. Instead, a wider institutional context influenced by the implementation of a grazing ban has led to more individualized herding and increased short-term considerations of profit maximization in the study area, both of which are seen to undermine the sustainable use of pastureland. Based on our observations of herder responses to privatization, the grazing ban and a short experiment with lifting the grazing ban, we propose that the special characteristics of grassland and pastoralism call for institutions that facilitate locally originated pasture land use practice (e.g., co-operative herding, self-organized management) instead of exclusive reliance on a rigidly defined private property regime focused on fixed property boundaries.  相似文献
