The phytochrome photoreceptors play important roles in the photoperiodic
control of vegetative bud set, growth cessation, dormancy induction, and
cold-hardiness in trees. Interestingly, ecotypic differences in
photoperiodic responses are observed in many temperate- zone tree species.
Northern and southern ecotypes of black cottonwood (Populus trichocarpa
Torr. & Gray), for example, exhibit marked differences in the timing of
short-day-induced bud set and growth cessation, and these responses are
controlled by phytochrome. Therefore, as a first step toward determining
the molecular genetic basis of photoperiodic ecotypes in trees, we
characterized the phytochrome gene (PHY) family in black cottonwood. We
recovered fragments of one PHYA and two PHYB using PCR-based cloning and by
screening a genomic library. Results from Southern analyses confirmed that
black cottonwood has one PHYA locus and two PHYB loci, which we arbitrarily
designated PHYB1 and PHYB2. Phylogenetic analyses which included PHY from
black cottonwood, Arabidopsis thaliana and tomato (Solanum lycopersicum)
suggest that the PHYB/D duplications in these species occurred
independently. When Southern blots were probed with PHYC, PHYE, and PHYE
heterologous probes, the strongest bands that we detected were those of
black cottonwood PHYA and/or PHYB. These results suggest that black
cottonwood lacks members of the PHYC/F and PHYE subfamilies. Although black
cottonwood could contain additional PHY that are distantly related to known
angiosperm PHY, our results imply that the PHY family of black cottonwood
is less complex than that of other well-characterized dicot species such as
Arabidopsis and tomato. Based on Southern analyses of five black cottonwood
genotypes representing three photoperiodic ecotypes, substantial
polymorphism was detected for at least one of the PHYB loci but not for the
PHYA locus. The novel character of the PHY family in black cottonwood, as
well as the differences in polymorphism we observed between the PHYA and
PHYB subfamilies, indicates that a number of fundamental macro- and
microevolutionary questions remain to be answered about the PHY family in
dicots.

Ganoderma lucidum powder using hot water and methanol extraction methods indicated a twofold more active cytotoxic activity with IC50 of 44 ± 3.8 μg/ml in the latter method. The representative dose-response curves of the G. lucidum crude extracts on J558 cell-lines revealed that there were great similarities between the curves which reflected rapid killing
activities. The percentage viability of the J558 cell exposed to these crude extracts was dose dependent only up to 150 μg/ml.
After which, there was no significant reduction when the dose was increased to 200 or 400 μg/ml. The morphological alterations
induced by the crude extract were examined under the phase contrast, fluorescent and electron microscopy. When J558 cells
were treated with doses higher than 50 μg/ml of the crude extract, obvious morphological changes and apoptosis occurred after
72 h. At 400 μg/ml, most of the cells showed necrosis characterized as small fragments with uniformly stained red nuclei.
The apoptotic and necrotic cells increased by 16.5 and 29.1%, respectively whereas the viable cells decreased by as much as
45.6. The mode of cell death via apoptosis was 3.6% higher than necrosis. However, these morphological changes were not observed
in the case of 3T3 cells. Results obtained from scanning electron microscopy and transmission electron microscopy further
confirmed the occurrence of various apoptotic and necrotic features. 相似文献

Production of aflatoxins on both natural (rice and corn) and semisynthetic (YES) media was conducted using an identified toxin-producing strain ofAspergillus flavus. TheA flavus strain was able to produce 4 types of aflatoxins, namely B1, B2, G1, and G2 on rice, corn, and YES media. Quantitative data showed that the concentrations of aflatoxins B1 and G1 produced were 52, 40.3, and 39.6; and 64.7, 45.0, and 58.0jug for 50g of rice, corn, and YES media, respectively. In comparison, the yielded amounts of aflatoxins B2 and G2 were much lower: 11.5, 17.9, and 17.5; and 28.S, 40.3, and 39.5 μg for 50 g of rice, corn, and YES media, respectively. A bioassay was conducted using the following 5 standard bacterial strains:Bacillus megaterium. Bacillus subtilis, Streptococcus faecal is, Staphylococcus epidermidis, andParacoccus denitrificans as well as a field strain of Candida albicans. All strains exceptP denitrificans showed varied degrees of inhibition when applied with crude aflatoxins at 5 to 40μg/mL. The minimum concentration of crude aflatoxins needed to inhibitP denitrificans was 10μg/mL. Moreover,Candida albicans was not inhibited at any concentration of aflatoxins applied in this work. Both undiluted and diluted (1/10, 1/100, and 1/1000) bacterial broth cultures showed a direct relationship between the diameter of inhibition zones and the concentrations of crude aflatoxins. Mean diameters of (7.0–20.5), (5–14), (4.5–13.0), (3.0–12.0), and (1.5–11.0) mm were observed when various concentrations of aflatoxins were applied usingB megaterium, S epidermidis, S faecal is, B subtilis, andP denitrificans, respectively. Field trials were applied to testify the validity of our data. A 1/100 dilution was prepared from each strain of 4 different species to estimate aflatoxins in samples of contaminated corn. Both chemical and biological assays were carried out at the same time. Data revealed that the most sensitive organism inhibited by as low as 7.5μg aflatoxins/mL wasB megaterium giving an inhibition zone of 10.5 mm, followed byS epidermidis with an inhibition zone of 7.5mm. In relation, the other 2 organisms were less sensitive to crude aflatoxins. Similarly, the biological assay was applied to detect aflatoxins in some samples of wheat, corn, peanut, rice, and poultry rations. The anti-algal activity of five macrophyte extracts on the cyanobacterium Microcystis aeruginosa in Egypt was investigated in 2013. Extract activity varied according to plant type, extracting solvent and its concentration. The highest inhibitory activity was achieved with ethanol extract at a concentration of 80 mg l?1, followed by chloroformic extracts, at 60 mg l?1. Methanolic extracts of Eichhornia crassipes and Polygonum tomentosum inhibited growth of Microcystis aeruginosa at all concentrations. Acetonic extracts inhibited algal growth at 60 mg l?1, except for the extract of Ceratophyllum subdemersum, which showed stimulation of M. aeruginosa growth. Eichhornia crassipes ethanolic extract exerted the most powerful inhibition by more than five-fold, 570.17%, followed by those of P. tomentosum, Saccharum spontaneum, Ceratophyllum demersum and C. subdemersum, 559.48, 553.99, 544.11 and 366.51%, respectively. Phytochemical screening for the tested plant extracts revealed the presence of biologically active substances of different concentrations, with P. tomentosum having the highest polyphenols, 1.95% of dry weight. 相似文献

Due to the recent advances of atovaquone, a naphthoquinone, through clinical trials
as treatment for malarial infection, 19 quinone derivatives with previously reported
structures were also evaluated for blood schizonticide activity against the malaria
parasite Plasmodium falciparum. These compounds include 2-hydroxy-3-methylamino
naphthoquinones (2-9), lapachol (10), nor-lapachol (11), iso-lapachol (12), phthiocol
(13) and phenazines (12-20). Their cytotoxicities were also evaluated against human
hepatoma and normal monkey kidney cell lines. Compounds 2 and 5 showed the highest
activity against P. falciparum chloroquine-resistant blood-stage parasites (clone
W2), indicated by their low inhibitory concentration for 50% (IC50) of
parasite growth. The therapeutic potential of the active compounds was evaluated
according to the selectivity index, which is a ratio of the cytotoxicity minimum
lethal dose which eliminates 50% of cells and the in vitro IC50.
Naphthoquinones 2 and 5, with activities similar to the reference antimalarial
chloroquine, were also active against malaria in mice and suppressed parasitaemia by
more than 60% in contrast to compound 11 which was inactive. Based on their in vitro
and in vivo activities, compounds 2 and 5 are considered promising molecules for

At present, no definitive agreement on either the correct branching order
or differential rates of evolution among the higher primates exists,
despite the accumulated integration of decades of morphological,
immunological, protein and nucleic acid sequence data, and numerous
reasonable theoretical models for the analysis, interpretation, and
understanding of those data. Of the three distinct unrooted phylogenetic
trees, that joining human with chimpanzee and the gorilla with the
orangutan is currently favored, but the two alternatives that group humans
with either gorillas or the orangutan rather than with chimpanzees also
have support. This paper is a synthetic and critical review of the
methodological literature and isolates some 20 specific reasons why
uncertainty in the evolutionary understanding of our closest living
relatives persists. Many of the difficulties are eliminated or ameliorated
by Lake's new methods of phylogenetic invariants and operator metrics. In
the companion paper these new methods are used to analyze both the nuclear
and mitochondrial DNA of the higher primates.

The aim of the present study was to enhance the dissolution rate of meloxicam (MLX), a practically water-insoluble drug by
preparation of solid dispersion using a hydrophilic polymer, poloxamer 188 (PXM). The kneading technique was used to prepare
solid dispersions. A 32 full factorial design approach was used for optimization wherein the drug, polymer ratio (X
1), and the kneading time (X
2) were selected as independent variables and the dissolution efficiency at 60 min (%DE60) and yield percent were selected as the dependent variable. Multiple linear regression analysis revealed that for obtaining
higher dissolution of MLX from PXM solid dispersions, a high level of X
1 and a high level of X
2 were suitable. The use of a factorial design approach helped in optimization of the preparation and formulation of solid
dispersion. The optimized formula was characterized by solubility studies, angle of repose, and contact angle; Fourier transform
infrared spectroscopy, differential scanning calorimetry, x-ray diffraction studies, and scanning electron microscopy demonstrated
that enhanced dissolution of MLX from solid dispersion might be due to a decrease in the crystallinity of MLX and PXM. Analysis
of dissolution data of optimized formula indicated the best fitting with Korsemeyer–Peppas model and the drug release kinetics
as Fickian diffusion. In conclusion, dissolution enhancement of MLX was obtained by preparing its solid dispersion with PXM
