Accumulation of abnormal proteins occurs in many neurodegenerative diseases including Huntington's disease (HD). However, the precise role of protein aggregation in neuronal cell death remains unclear. We show here that the expression of N-terminal huntingtin proteins with expanded polyglutamine (polyQ) repeats causes cell death in neuronal PC6.3 cell that involves endoplasmic reticulum (ER) stress. These mutant huntingtin fragment proteins elevated Bip, an ER chaperone, and increased Chop and the phosphorylation of c-Jun-N-terminal kinase (JNK) that are involved in cell death regulation. Caspase-12, residing in the ER, was cleaved in mutant huntingtin expressing cells, as was caspase-3 mediating cell death. In contrast, cytochrome-c or apoptosis inducing factor (AIF) was not released from mitochondria after the expression of these proteins. Treatment with salubrinal that inhibits ER stress counteracted cell death and reduced protein aggregations in the PC6.3 cells caused by the mutant huntingtin fragment proteins. Salubrinal upregulated Bip, reduced cleavage of caspase-12 and increased the phosphorylation of eukaryotic translation initiation factor-2 subunit-alpha (eIF2alpha) that are neuroprotective. These results show that N-terminal mutant huntingtin proteins activate cellular pathways linked to ER stress, and that inhibition of ER stress by salubrinal increases cell survival. The data suggests that compounds targeting ER stress may be considered in designing novel approaches for treatment of HD and possibly other polyQ diseases.  相似文献

We aimed to study whether the varying changes in predation pressure by perch (Perca fluviatilis) reflect the biomass, density, and community structure of the benthic macroinvertebrates. Prey preference is size-dependent, and overall predation pressure is density dependent, and thus the size structure of the P. fluviatilis population should affect the structure of the macroinvertebrate community, and the population density of P. fluviatilis should reflect the overall density of benthic macroinvertebrates. We sampled the littoral benthic community in a boreal lake that had been divided into two parts that were subjected to two different fishing procedures during 2007–2012 period and analyzed the macroinvertebrate diet of fish. The benthic macroinvertebrate community reflected the predation pressure. Total macroinvertebrate biomass increased during the study period in the lake division with a non-size-selective fishing procedure (NSF), i.e., all invertivorous perch size-classes targeted, but decreased in the section with negatively size-selective fishing procedure (SSF), i.e., large invertivorous individuals ≥ 16 cm were not targeted. This difference was a result of the increase in large-sized species, such as Odonata, for the NSF procedure and decrease in the SSF procedure. In contrast to total biomass, total macroinvertebrate density did not show a response to predator size structure but rather total macroinvertebrate density decreased with increasing fish density. The study demonstrates the effect of predation pressure of P. fluviatilis on benthic communities, thus highlighting the keystone predator role of the species in boreal lakes and gives more insight on the multiple effects of fish predation on littoral benthic communities.  相似文献

The coexistence of competing species relies on niche partitioning. Competitive exclusion is likely inevitable at high niche overlap, but such divide between competitors may be bridged if environmental circumstances displace competitor niches to enhance partitioning. Foraging‐niche dimension can be influenced by environmental characteristics, and if competitors react differently to such conditions, coexistence can be facilitated. We here experimentally approach the partitioning effects of environmental conditions by evaluating the influence of water turbulence on foraging‐niche responses in two competing fish species, Eurasian perch Perca fluviatilis and roach Rutilus rutilus, selecting from planktonic and benthic prey. In the absence of turbulence, both fish species showed high selectivity for benthic chironomid larvae. R. rutilus fed almost exclusively on zoobenthos, whereas P. fluviatilis complemented the benthic diet with zooplankton (mainly copepods). In turbulent water, on the other hand, the foraging‐niche widths of both R. rutilus and P. fluviatilis increased, while their diet overlap simultaneously decreased, caused by 20% of the R. rutilus individuals turning to planktonic (mainly bosminids) prey, and by P. fluviatilis increasing foraging on littoral/benthic food sources. We show that moderate physical disturbance of environments, such as turbulence, can enhance niche partitioning and thereby coexistence of competing foragers. Turbulence affects prey but not fish swimming capacities, with consequences for prey‐specific distributions and encounter rates with fish of different foraging strategies (pause‐travel P. fluviatilis and cruise R. rutilus). Water turbulence and prey community structure should hereby affect competitive interaction strengths among fish species, with consequences for coexistence probability as well as community and system compositions.  相似文献

Resting‐state functional magnetic resonance imaging (rs‐fMRI) has been successfully used to probe the intrinsic functional organization of the brain and to study brain development. Here, we implemented a combination of individual and group independent component analysis (ICA) of FSL on a 6‐min resting‐state data set acquired from 21 naturally sleeping term‐born (age 26 ± 6.7 d), healthy neonates to investigate the emerging functional resting‐state networks (RSNs). In line with the previous literature, we found evidence of sensorimotor, auditory/language, visual, cerebellar, thalmic, parietal, prefrontal, anterior cingulate as well as dorsal and ventral aspects of the default‐mode‐network. Additionally, we identified RSNs in frontal, parietal, and temporal regions that have not been previously described in this age group and correspond to the canonical RSNs established in adults. Importantly, we found that careful ICA‐based denoising of fMRI data increased the number of networks identified with group‐ICA, whereas the degree of spatial smoothing did not change the number of identified networks. Our results show that the infant brain has an established set of RSNs soon after birth.  相似文献

Bifidobacterium longum NRRL B-41409 l-arabinose isomerase (l-AI) was overexpressed in Lactococcus lactis using a phosphate depletion inducible expression system. The resting L. lactis cells harboring the B. longum l-AI were used for production of d-tagatose from d-galactose in the presence of borate buffer. Multivariable analysis suggested that high pH, temperature and borate concentration favoured the conversion of d-galactose to d-tagatose. Almost quantitative conversion (92 %) was achieved at 20 g L?1 substrate and at 37.5 °C after 5 days. The d-tagatose production rate of 185 g L?1 day?1 was obtained at 300 g L?1 galactose, at 1.15 M borate, and at 41 °C during 10 days when the production medium was changed every 24 h. There was no significant loss in productivity during ten sequential 24 h batches. The initial d-tagatose production rate was 290 g L?1 day?1 under these conditions.  相似文献
