Procedures are deseribed for the efficient isolation of protoplasts from a variety of oil palm (Elaeis guineensis Jacq.) tissues. Various factors including donor source, composition of enzyme mixture and culture medium affected the yield and viability of the protoplasts Polyembryogenic cultures of oil palm were the most suitable starting material in terms of yield, viability and metabolic competence. Pectolyase Y-23 in association with cellulase and hemicellulase was required for the efficient release of protoplasts from the oil palm tissues. Summary The occurrence of cells resembling mammalian Langerhans cells in the avian epidermis was studied by ATPase histochemistry, Ia immunoreactivity and electron microscopy. The existence of MHC class II antigen-(Ia) expressing, ATPase-positive dendritic cells, which are ultrastructurally similar to mammalian Langerhans cells except for the absence of Birbeck granules, was demonstrated. Ensuring food security for the rapidly increasing population and changing climatic scenarios are requisites for
exploiting the genetic divergence of food crops. A study was undertaken to sort out an early maturing chickpea
variety for fitting easily between rice-rice cropping systems in the Eastern Indo-Gangetic Plain of Bangladesh. The
trial was comprised of eight elite lines of chickpea and executed at various localities in Bangladesh from 2014–
15 to 2017–18. The result explored the chickpea genotype, BARI Chola-11 remained superior to the rest of
the elite genotypes for having a short maturity period (100–106 days), and lesser days to 50% flowering (47–
55 days). The same genotype was recorded to have robust vegetative and reproductive yield attributes including
plant height (49–57 cm), podsplant−1 (37–50), and optimum 100 seed weight (19.5–20.6 g). Owing to better yield
attributes, BARI Chola-11 resulted in the maximum seed yield (1200–1500 kg ha−1
) of chickpea and might be

Many plants have been known to be contaminated and accumulate plasticizers from the environment, including water sources, soil, and atmosphere. Plasticizers are used to confer elasticity and flexibility to various fiber and plastic products. Consumption of plasticizers can lead to many adverse effects on human health, including reproductive and developmental toxicity, endocrine disruption, and cancer. Herein, we report for the first time that two plasticizers, bis(2-ethylhexyl) terephthalate (DEHT) and bis(2-ethylhexyl) phthalate (DEHP), have been isolated from the leaves of Capparis spinosa L. (the caper bush), a plant that is widely used in food seasonings and traditional medicine. 297 mg/kg of DEHT and 48 mg/kg of DEHP were isolated from dried and grounded C. spinosa L. leaves using column chromatography and semi-preparative high-performance liquid chromatography. Our study adds to the increase in the detection of plasticizers in our food and medicinal plants and to the alarming concern about their potential adverse effects on human health. 相似文献

A direct continuous UV-Vis spectrophotometric assay has been developed for VanX, a D-alanyl-D-alanine aminodipeptidase necessary for vancomycin resistance. This method is based on the hydrolysis of the alternative substrate D-alanyl-alpha-(R)-phenylthio-glycine D-Ala-D-Gly(S-Ph)-OH (H-DAla-DPsg-OH, 5a). Spontaneous decomposition of the released phenylthioglycine generates thiophenol, which is quantified using Ellman's reagent. The dipeptide behaved as an excellent substrate of VanX, exhibiting Michaelis-Menten kinetics with a kcat of 76 +/- 5/s and a KM of 0.83 +/- 0.08 mm (kcat = 46 +/- 3/s and KM = 0.11 +/- 0.01 mm for D-Ala-D-Ala). Determination of the kinetic parameters of the previously reported mechanism-based inhibitor D-Ala-D-Gly(SPhip-CHF2)-OH (H-D-Ala-DPfg-OH, 5c) [Araoz, R., Anhalt, E., René, L., Badet-Denisot, M.-A., Courvalin, P. & Badet, B. (2000) Biochemistry 39, 15971-15979] using the substrate reported in the present study yielded values of Kirr of 22 +/- 1 microM and kinact of 9.3 +/- 0.4/min in good agreement with values previously obtained in our laboratory (Kirr = 30 +/- 1 mm; kinact = 7.3 +/- 0.3/min). In addition, inhibition by the competing substrate D-Ala-D-Ala resulted in determination of a Ki = 70 +/- 6 microM close to the previously reported KM value. To determine how the cost of reproduction varies with brood size, a population of blue tits (Parus caeruleus) breeding in Wytham Wood, England, was manipulated over a three year period. Two hundred sixteen pairs were randomly assigned 3, 6, 9, 12, or 15 nestlings; nestlings were exchanged soon after hatching. Survival of adult females (as measured by the proportion recaptured in the following winter and/or spring) declined significantly with increasing brood size in two out of three years; there was significant year-to-year variation in the relationship of recapture rate to brood size. Mean female recapture rates (averaged over the three years) declined in a linear fashion (P < 0.01). There was no significant linear or curvilinear relationship between male-recapture rate and brood size in any of the three years nor was there a significant linear or curvilinear relationship for the data averaged over the three years. Nevertheless, recapture proportions for males differed significantly with respect to brood size (χ2 test, P < 0.05). The possibility that experimental brood size influences subsequent dispersal (and therefore biases measures of survival based on recapture rates to differing degrees) was examined by comparing distances moved by breeding adults from one year to the next. There was no relationship between brood size and dispersal distance within the study area for either sex, except that females given broods of three were significantly more likely to move more than 300 m than were those given broods of 6–15 young. Both males and females showed evidence of a cost with respect to future fecundity: as brood size increased, the number of surviving offspring produced in the following year decreased from 1.5–1.6 (for adults that had reared 3–6 young) to 0.4 (for those that had reared 15 young). The relationship of future reproductive success to experimental brood size did not differ among years or between the sexes. The number of eggs laid and number of young hatched in year n + 1 did not differ significantly with respect to brood size in year n; rather, differences in future fecundity reflected differences in the survival prospects of young reared in year n + 1. 相似文献

Alkali extracted mycelial biomass from Aspergillus niger, referred to as Biosorb, was found to sequester metal ions (Cd2+, Cu2+, Zn2+, Ni2+ and Co2+) efficiently both from dilute and concentrated solutions upto 10% of its weight (w/w). Sequestration of metal ions from a mixture was also efficient but with attendant antagonisms. The kinetics of metal binding by Biosorb indicated that it is a rapid process and about 70–80% of the metal is removed from solution in 5 min followed by a slower rate. The mechanism of metal binding is shown to be due to exchange of calcium and magnesium ions of the Biosorb during which equimolar concentrations of both the ions were released into the medium. Following this an efficient procedure for the regeneration and reuse of Biosorb was standardized by washing the biosorbent with calcium and magnesium solution (0.1 m). Biosorbents prepared from Neurospora, Fusarium and Penicillium also exhibited similar mechanisms for metal ion binding, though they had a lower metal binding capacity when compared with Biosorb. Chemical modification of carboxylic acid functional groups of the Biosorb resulted in loss of 90% of metal binding capacity which could not be restored even on regeneration. The significance of this finding on the metal sequestration mechanisms of microbial biosorbents is discussed. 相似文献

In model experiments with Bonus barley,where the growth of a fraction of the seedlings is retarded by a raised thiol content, obtained through treatment with 2-mercaptoethanol, stimulation of growth is reproducibly induced by low doses of γ-rays. As illustrated by a probit representation of seedling heights, and by the decrease of coefficients of variation, the radiostimulation is restricted to that fraction of seedlings which exhibits a suppressd growth. The validity of the model to natural situations was sustained in studies of the variety Impala, which was shown by Fendrik and Bors to respond regularly to stimulating doses of X-rays. Azospirillum brasilense was attracted to capillaries containing either phosphate buffer, distilled water, or saline. The number of bacteria in these capillaries was 3–4×104, after 1 h of incubation. In the presence of phosphate buffer + attractants, the number of cells accumulated in the capillary increased only to 5×104–1.1×105 cells. It was not possible, therefore, to measure chemotaxis inA. brasilense as distinct from aerotaxis by the capillary method. Chemotaxis was observed in semi-solid agar plates and was determined by a growth band oriented towards the attractant.
