The insulin-like growth factor-1 receptor (IGF-1R) is a tyrosine kinase receptor of central importance in cell proliferation. A fragment (residues 1-462) comprising the L1-cysteine rich-L2 domains of the human IGF-1R ectodomain has been overexpressed in glycosylation-deficient Lec8 cells and has been affinity-purified via a c-myc tag followed by gel filtration. The fragment was recognized by two anti-IGF-1R monoclonal antibodies, 24-31 and 24-60, but showed no detectable binding of IGF-1 or IGF-2. Isocratic elution of IGF-1R/462 on anion-exchange chromatography reduced sample heterogeneity, permitting the production of crystals that diffracted to 2.6 A resolution with cell dimensions a = 77.0 A, b = 99.5 A, c = 120.1 A, and space group P2(1)2(1)2(1).  相似文献

Fixation of cellulose-acetate membranes with either glutaraldehyde-osmium tetroxide or glutaraldehyde-ruthenium tetroxide resulted in extensive electron beam damage. Beam damage was eliminated and the bacterial surface structure was preserved, however, when cellulose-acetate membranes were fixed with glutaraldehyderuthenium tetroxide and treated successively with thiocarbohydrazide and osmium tetroxide.  相似文献

This study investigates the effects of captivity and testosterone treatment on the volumes of brain regions involved in processing visual and spatial information in adult dark‐eyed juncos (Junco hyemalis). We treated captive and free‐living male juncos with either testosterone‐filled or empty implants. Captive juncos had a smaller hippocampal formation (HF) (both in absolute volume and relative to telencephalon) than free‐living birds, regardless of hormone treatment. Testosterone‐treated males (both captive and free‐living) had a smaller telencephalon and nucleus rotundus, but not a smaller HF or ectostriatum, than controls. We found that free‐living testosterone‐treated males had larger home ranges than free‐living controls in agreement with earlier experiments, but we found no corresponding difference in HF volume. Sensitivity of tissues to the acute toxicity of cadmium is reviewed. It is concluded that the initial effect of acute cadmium administration is on the integrity and permeability of the vascular endothelium; other necrotic changes occur secondarily to this effect. In a sensitive tissue, not all of the endothelial cells are susceptible to cadmium. Furthermore, after necrosis of the sensitive cells, the resistant cells proliferate and result in regeneration of the vasculature and subsequent acquired resistance to the metal. It is found that sex hormones are probably important in determining susceptibility and response of tissues to cadmium. The role of metallothionein in these phenomena remains to be elucidated.  相似文献

Neurologic disease caused by human immunodeficiency virus type 1 (HIV-1) is ultimately refractory to highly active antiretroviral therapy (HAART) because of failure of complete virus eradication in the central nervous system (CNS), and disruption of normal neural signaling events by virally induced chronic neuroinflammation. We have previously reported that HIV-1 Tat can induce mitochondrial hyperpolarization in cortical neurons, thus compromising the ability of the neuron to buffer calcium and sustain energy production for normal synaptic communication. In this report, we demonstrate that Tat induces rapid loss of ER calcium mediated by the ryanodine receptor (RyR), followed by the unfolded protein response (UPR) and pathologic dilatation of the ER in cortical neurons in vitro. RyR antagonism attenuated both Tat-mediated mitochondrial hyperpolarization and UPR induction. Delivery of Tat to murine CNS in vivo also leads to long-lasting pathologic ER dilatation and mitochondrial morphologic abnormalities. Finally, we performed ultrastructural studies that demonstrated mitochondria with abnormal morphology and dilated endoplasmic reticulum (ER) in brain tissue of patients with HIV-1 inflammation and neurodegeneration. Collectively, these data suggest that abnormal RyR signaling mediates the neuronal UPR with failure of mitochondrial energy metabolism, and is a critical locus for the neuropathogenesis of HIV-1 in the CNS.  相似文献

Thermotoga sp. strain RQ2 is probably a strain of Thermotoga maritima. Its complete genome sequence allows for an examination of the extent and consequences of gene flow within Thermotoga species and strains. Thermotoga sp. RQ2 differs from T. maritima in its genes involved in myo-inositol metabolism. Its genome also encodes an apparent fructose phosphotransferase system (PTS) sugar transporter. This operon is also found in Thermotoga naphthophila strain RKU-10 but no other Thermotogales. These are the first reported PTS transporters in the Thermotogales.  相似文献

The appearance of the spionid polychaete Marenzelleria spp. in the Tay estuary (Scotland) was first reported in 1984. Since estuaries are known as environments where abiotic conditions fluctuate widely, a laboratory study was conducted aimed to elucidate how this introduced species deals with low oxygen concentrations and exposure to hydrogen sulphide. Experimental evidence reveals that during severe hypoxia Marenzelleria cf. wireni switched to anaerobic pathways known from other marine invertebrates. Under the influence of sulphide (1 mmol l-1) accumulation of anaerobic end products is more pronounced. It is assumed that in the presence of sulphide, due to its inhibition of aerobic respiration, the worms have to switch to anaerobic energy production directly after the onset of hypoxia. Seasonal differences in the metabolic reaction were found. It was shown that in specimens that were filled with gametes (winter), accumulation of end products of anaerobic metabolism was more pronounced than in immature worms (summer). The amount of succinate, an indicator for anaerobic energy production for instance, was nearly twice as high in the winter specimens. Since the amount of thiosulphate (a typical sulphide detoxification product) in the tissues is relatively low, it is suggested that Marenzelleria cf. wireni also accumulates other detoxification products. Sulphite, however, also known as a detoxification product, was found only in traces. Field measurements provide evidence that the potential of Marenzelleria cf. wireni to survive low oxygen and high sulphide concentrations is clearly higher than that normally needed in the Tay estuary.  相似文献

Genetic variation for quantitative traits is often greater than that expected to be maintained by mutation in the face of purifying natural selection. One possible explanation for this observed variation is the action of heterogeneous natural selection in the wild. Here we report that selection on quantitative trait loci (QTL) for fitness traits in the model plant species Arabidopsis thaliana differs among natural ecological settings and genetic backgrounds. At one QTL, the allele that enhanced the viability of fall-germinating seedlings in North Carolina reduced the fecundity of spring-germinating seedlings in Rhode Island. Several other QTL experienced strong directional selection, but only in one site and seasonal cohort. Thus, different loci were exposed to selection in different natural environments. Selection on allelic variation also depended upon the genetic background. The allelic fitness effects of two QTL reversed direction depending on the genotype at the other locus. Moreover, alternative alleles at each of these loci caused reversals in the allelic fitness effects of a QTL closely linked to TFL1, a candidate developmental gene displaying nucleotide sequence polymorphism consistent with balancing selection. Thus, both environmental heterogeneity and epistatic selection may maintain genetic variation for fitness in wild plant species.  相似文献

Background  

Streptomyces coelicolor is a bacterium with a vast repertoire of metabolic functions and complex systems of cellular development. Its genome sequence is rich in genes that encode regulatory proteins to control these processes in response to its changing environment. We wished to apply a recently published bioinformatic method for identifying novel regulatory sequence signals to gain new insights into regulation in S. coelicolor.  相似文献
