Effects of the rad 2-20, rad 9-4, r1s, and the corresponding wild type RAD alleles in haploid and homozygous diploid Saccharomyces strains on UV induced mutation rates from adenine, lysine and histidine dependence to independence are reported. The UV induced mutation rates were similar for the RAD, r1s, and rad 9-4 haploids, whereas the rad 2-20 mutation causes a marked increase in the UV induced mutation rates. The diploid rad 2-20 strain also exhibits a marked increase in the UV induced mutation rates, whereas the rad 9-4 diploid has reduced mutation rates when compared to the wildtype. The UV induced mutation rates of haploid and diploid RAD strains are almost identical. For the rad 2-20 and rad 9-4 diploids, however, these rates are smaller than in the corresponding haploid strains. Differential effects of the rad genes on the ratio of locus to suppressor mutations were found. The implications of these findings on possible repair processes in yeasts are discussed. 相似文献

The morphological and molecular differentiation of the Micrasterias truncata (Corda) ex Bréb species complex was investigated. In total, 17 strains traditionally assigned to M. truncata were isolated from different European localities (Czech Republic, southwest France, Ireland), and obtained from public culture collections. In addition, strains of the morphologically similar species, M. decemdentata (Nägeli) W. Archer and M. zeylanica F. E. Fritsch, were also included. Molecular phylogenetic analysis based on trnGucc intron sequences revealed five well supported clades. Two Australian strains assigned to M. truncata var. pusilla G. S. West formed a lineage sister to M. zeylanica. This was evident from a concatenated phylogeny based on small subunit rDNA and trnGucc intron sequences. The isolated position of these strains was also illustrated by parallel landmark‐based geometric morphometric analysis of cell shapes. The strains NIES 783 and NIES 784 probably represent a separate species. Particular analysis, including additional strains, is needed to resolve the relationship inside this lineage. The second phylogenetic lineage, containing two strains of M. truncata var. semiradiata (Kützing) Wolle, was also different from other strains on the basis of morphometric data. We suggest recognizing this variety as a separate species, Micrasterias semiradiata L.A. Brébisson ex F. T. Kützing. The remaining three clades formed a firmly supported group of the ‘core’M. truncata recognized by both molecular markers. However, neither any morphological, morphometric, nor geographical pattern was detected among members of these three clades. This pattern could be caused by a relatively recent origin of these lineages that may represent a sympatric, truly cryptic species. The present study focuses on temporal and microscale spatial variation of the community structure and richness of subaerial microalgae growing on the bark of European beech (Fagus sylvatica) trees in temperate deciduous forests. Subaerial phototrophic biofilms present common and conspicuous microalgal communities growing on a variety of natural and man‐made substrata. However, in comparison with other major microalgal communities such as phytoplankton and microphytobenthos, basic patterns of their spatio‐temporal variation remain largely unknown. The bark samples were collected six times each spring and autumn in a period of 3 years (2010–2013) and were cultured on agar plates, and then individual clonal strains were identified by light microscopy. A total of 55 morphotypes (considered as operational taxonomic units for subsequent analyses) were recognized, which mainly belong to the classes Trebouxiophyceae and Chlorophyceae. Interestingly, temporal variation explained the largest proportion of variation in the community structure. This variation was primarily related to seasonal fluctuations, and although the communities recorded in spring and autumn showed many overlapping taxa, a clear distinction in species composition and abundance was observed. However, the microhabitat characteristics such as bark roughness also significantly structured the microalgal community. Conversely, spatial factors such as the height of the samples above ground or distance of the samples on a trunk seemed to be of lesser importance on this scale. Thus, we concluded that the previously unrecognized seasonal changes, resulting from variation in temperature, humidity, and irradiance, as well as the non‐seasonal temporal changes, possibly resulting from local colonization or extinction of individual taxa, should be considered as one of the important factors in structuring aerial microalgal communities. 相似文献

Sporopollenin layers in the cell wall of coccal green algae are responsible for the resistance of cell walls to destructive processes during fossilization as well as during chemical preparation of samples for pollen-analysis. Pollen slides of samples from limnic sediments thus also contain some algal cell walls. Although some pollen-analysts tried to stress this fact, the finds of algae in pollen slides have not been paid systematic attention yet, despite their potential use for a more accurate palaeoecological reconstruction. The article summarizes the results of palaeoecological studies showing how the algae can be used in palaeoecological reconstruction of past environments. The possibility of utilizing the indicative value of algal finds is demonstrated on examples of algal communities from fossil, subrecent and recent sediments from different longitudes, latitudes, and altitudes. The identification and indicative values of species and varieties ofPediastrum are included in a special review (Komárek &; Jankovská, Biblioth. Phycol., in press). The contemporary knowledge of ecological requirements of the given taxa, completed by information from their fossil finds, makes possible the reconstruction of trophic and temperature conditions and of the purity of the water environment of the past water biotopes. 相似文献

Removal of inorganic nitrogen compounds from wastewaters can be accomplished by a combination of the biological processes of nitrification and denitrification. The information on the microbiota present in denitrifying reactors is still scarce. In the present work the evaluation of the denitrifying microbiota of different reactor sludges was performed by specific activity measurements and MPN count of denitrifiers. We also present the isolation and physiological and phylogenetic characterisation of denitrifying bacteria from the anoxic reactor of a combined system treating landfill leachate. Specific denitrifying activity measurements were faster to perform and more reliable than MPN enumerations. 16S rDNA characterisation of the isolates showed that they belonged to the genera Thauera, Acidovorax and Alcaligenes and were closely related to microorganisms retrieved from ecosystems rich in recalcitrant compounds. Two of the isolates could grow on aromatic compounds as sole carbon source. 相似文献

Geckos of the genus Trigonodactylus are widely distributed in the sand deserts of the Arabian Peninsula. Three species of this genus are currently recognized, with a fourth one, Stenodactylus pulcher, which placement within Trigonodactylus has been tentatively suggested, but not yet confirmed. We present a phylogenetic analysis of the genus Trigonodactylus with new specimens collected in central Saudi Arabia and southern Jordan. New genetic data has been generated from three mitochondrial markers to investigate the phylogenetic relationships of all species of the genus and to assess the putative generic assignment of S. pulcher. Our results confirm that S. pulcher indeed belongs within Trigonodactylus, branching as a sister lineage to all other species of the genus. The new samples cluster within Trigonodactylus arabicus, thus confirming the genetic homogeneity of the species across its large and seemingly inhospitable range. The new specimen collected in southern Jordan represents the first record for the country and a considerable range extension to the northwest from all previously reported localities. Our findings and discovery of a new species for Jordan highlight the need of more field surveys to be carried out in the underexplored parts of Jordan and northern Saudi Arabia, as these places still hold a potential for new discoveries and are crucial for understating the biogeography of the Arabian herpetofauna. 相似文献

Finds of fossil wood with bivalve wood borings (Teredolites clavatus and T. longissimus) occur in various facies and presumed sedimentary settings of the platform, shallow-marine Bohemian Crectaceous Basin. The basin comprises areas with sandy-dominated sediments, with marl and clay-dominated sediments, areas with predominat sandy-marly rocks, and finally areas dominated by calcareous nearshore sediments. Teredolites clavatus is common in fossil wood of sandstones, originating in beach or deltaic settings; marl and clay-dominated rock frequently bear wood fragments densely bored by Teredolites longissimus. When accompanied by evidence of marine environments as body fossils, glauconite or typical trace fossils, most of the wood fragments are bored. The presence/absence of borings in wood fragments can be considered the most reliable and easily useable criterion of distinction of marine settings in sandy sediments of the margin of the Bohemian Cretaceous Basin. 相似文献

A series of new monophosphates of 1-[2-(phosphonomethoxy)alkyl]thymines, such as PMPTp, 3-MeO-PMPTp, HPMPTp, and FPMPTp, were synthesized and tested for their ability to inhibit human thymidine phosphorylase. Kinetic measurements of enzyme activity were performed using thymidine and inorganic phosphate as the substrates. The data show that some monophosphates provide a considerable increase of the multisubstrate inhibitory effect. The highest inhibitory potency was found with (R)-FPMPTp 4c (K i dT = 4.09 ± 0.47 μM, K i(Pi) = 2.13 ± 0.29 μM) and (R) 3-MeO-PMPTp 4d (K i dT = 5.78 ± 0.71 μM, K i(Pi) = 2.71 ± 0.37 μM). 相似文献
