The lignin-carbohydrate complex (LCC-W), isolated from the milled-wood, lignin fraction of Pinus densiflora Sieb. et Zucc., comprised three fractions (W-1,2,3) by gel filtration on Sepharose 4B. W-1 was eluted at the void volume, whereas W-2 and W-3 were included in the gel and had apparent weight-average molecular weights of 5.0 × 105 and 5.0 × 103, respectively. W-2 and W-3 were homogeneous in ultracentrifugal and electrophoretic analyses. The sedimentation coefficients of W-2 and W-3 were 25.7 and 0.4S, respectively. The chemical composition of W-2 was 38.0% of neutral sugar, 6.2% of uronic acid, 51.5% of lignin, and the corresponding values for W-3 were 73.1, 11.0, and 22.2%. The neutral carbohydrate residues of W-2 and W-3 were l-arabinose, d-xylose, d-mannose, d-galactose, and d-glucose in the ratios 15.8:16.2:37.3:16.7:14.0 and 27.6:16.5:26.1:19.3:10.5, respectively. Based on the results of methylation and Smith-degradation analyses, the carbohydrate moiety of the LCC-W fractions was found to be multiply branched. The major backbone structure was composed of (1→4)-linked d-mannopyranosyl residues. By hydrophobicinteraction chromatography on Phenyl- and Octyl-Sepharsoe CL-4B gels, it is concluded that the LCC-W fractions have a hydrophobic property that is exclusively ascribed to the lignin moiety.  相似文献

Cat eye syndrome (CES) is associated with a duplication of a segment of human chromosome 22q11.2. Only one gene,ATP6E, has been previously mapped to this duplicated region. We now report the mapping of the human homologue of the apoptotic agonistBidto human chromosome 22 near locus D22S57 in the CES region. Dosage analysis demonstrated thatBIDis located just distal to the CES region critical for the majority of malformations associated with the syndrome (CESCR), as previously defined by a single patient with an unusual supernumerary chromosome. However,BIDremains a good candidate for involvement in CES-related mental impairment, and its overexpression may subtly add to the phenotype of CES patients. Our mapping of murineBidconfirms that the synteny of the CESCR and the 22q11 deletion syndrome critical region immediately telomeric on human chromosome 22 is not conserved in mice.Bidand adjacent geneAtp6ewere found to map to mousechromosome 6, while the region homologous to the DGSCR is known to map to mouse chromosome 16.  相似文献

Heparin-binding epidermal growth factor-like growth factor (HB-EGF) is known to induce cell growth in various cell types via transactivation of epidermal growth factor receptor (EGFR). To investigate the involvement of HB-EGF and EGFR in cardiac remodeling after myocardial infarction (MI), we examined the expressions of mRNA and protein in rat hearts 6 weeks after MI-induction. Where increased expressions of HB-EGF mRNA and protein were observed, infarcted myocardium was replaced by extracellular matrix and interstitial fibroblasts. EGFR mRNA and protein expression did not show significant changes in sham-operated heart tissues, non-infarcted region, and infarcted region. In vitro study demonstrated that HB-EGF mRNA was expressed mainly in cultured fibroblasts rather than in myocytes. We suggest that the interaction between HB-EGF and EGFR transactivation is closely related to the proliferation of cardiac fibroblasts and cardiac remodeling after MI in an autocrine, paracrine, and juxtacrine manner.  相似文献

Stomatal movement is regulated by changes in the volume of guardcells, thought to be mainly controlled by an osmo-regulatorysystem. In the present study, we examined the additional involvementof cytoskeletal events in the regulation of stomatal movement.Microtubules (MTs) in guard cells of Viciafaba L., grown undersunlight, were observed during the day and night by immunofluorescencemicroscopy. Cortical MTs began to be organized in a radial arrayat dawn and increased in numbers in the morning following theincrease in the stomatal aperture size. Thereafter, MTs becamelocalized near the nucleus and began to be destroyed from theevening to midnight, following the decrease in stomatal aperturesize. These diurnal changes in MT organization were observedeven two days after transfer from natural light condition tototal darkness, and were accompanied by corresponding changesin stomatal aperture. The increase in stomatal aperture sizein the early morning was inhibited by 50 µM propyzaraide,which destroys cortical MTs in guard cells, whereas the decreasein aperture size in the evening was suppressed by 10 µMtax-ol, which stabilizes cortical MTs. Pup ultrasonic vocalizations (USVs) are emitted from maternally separated pups and are thought to be a trigger for eliciting maternal behavior in mice. We investigated the effects of social experience and gonadectomy on the retrieving behavior of mice and their responses to pup USVs produced by a nanocrystalline silicon thermo-acoustic emitter. In each experiment, virgin, gonadectomized, sham-operated, sexually experienced, and parenting mice of both sexes were used, and the effects of these manipulations were compared in each sex. The retrieving behavior of both sexes increased with social experience or gonadectomy. In particular, mothers showed the highest retrieving activity among female groups, while castrated male mice showed the highest retrieving activity among male groups. All groups of female mice responded to pup USVs, with the responsiveness of sexually experienced female mice being the most enhanced. Unlike the females, virgin male mice did not respond to pup USVs, although socially experienced or castrated males showed this response; fathers exhibited the highest responsiveness. These results suggest that not only parenting experience, but also mating experience, may enhance retrieving activity and response to pup USVs in mice of both sexes. Nevertheless, the degree to which parenting experience contributed to the enhancement of both activities differed between the sexes. Furthermore, gonadectomy enhanced both activities in both sexes, although its effect was more prominent in males. Overall, our findings suggest that alteration in responsiveness of mice to pup USVs might be one of the changes in parental behavior caused by social experiences or gonadal hormones.  相似文献

Coronary stenting (stent implantation) has evolved over the last 5 years with changes in stent design, stent material and the implantation technique. The use of high-pressure balloon inflation (HP), intravascular ultrasound (IVUS) and appropriate antiplatelet therapy have contributed to the abolishment of the need for subsequent anticoagulation, allowing extended stent applications. We compared results in three groups of patients having stent implantation throughout the period of evolution: group A: no IVUS, no HP, with subsequent anticoagulation treatment (n 3 434); group B: no IVUS, yes HP, without subsequent anticoagulation treatment (n 3 192); and group C: yes IVUS, yes HP, without subsequent anticoagulation treatment (n 3 588). The primary success rates were comparable in all groups. There was a clear change in indications for stenting in groups B and C compared with group A (elective stenting: group A 3 32%; group B 3 66%; group C 3 69%; P < 0.0001), in reference vessel size (group A 3 3.22 3 0.37 mm; group B 3 2.92 3 0.56 mm; group C 3 2.98 3 0.57 mm; P < 0.0001), and for presence of type B2 and C lesions (group A 3 57%; group B 3 72%; group C 3 74%; P < 0.001). The complication rate significantly decreased in group C (group A 3 3.6%; group B 3 4.1%; group C 3 1.2%; P < 0.001) and the mean patient hospital stay decreased to 2 days in groups B and C due to the abolition of the need for anticoagulant treatment. The angiographic restenosis rate increased in groups B and C (group A 3 20%; group B 3 34%; group C 3 32%; P < 0.001). The need for a repeat procedure increased as stenting of more complex lesions and smaller vessels was attempted: target lesion revascularization (TLR) was performed in 16% of patients in group A (73/434), in 18% of group B (35/192) and in 22% of group C (129/588) (P 3 0.04 for A versus C). Major cardiac events (MACE) occurred in 142 patients in group A (33%), 60 patients in group B (31%) and in 181 patients in group C (30%). The evolving technique of coronary stenting has expanded the spectrum of indications and range of coronary vessels attempted, and decreased the complication rates and hospital stay. However, in less-favorable subsets, additional improvements are needed to affect the long-term outcome.  相似文献

A small freshwater fish medaka (Oryzias latipes) has been one of the most attractive experimental systems for research in genetics and developmental biology. We have formed an international consortium Medaka Genome Initiative (MGI) to collect and share various information and resources on medaka. The MGI has set an ambitious goal aiming at the complete sequencing of the medaka genome and as a feasibility study we have begun sequencing one particular chromosome, linkage group 22 (LG22) of 22 Mb in size. Initial sequence analysis revealed unique features of the medaka genome in comparison to fugu genome.  相似文献

The Cgl1427 gene was previously found to be relevant to the microaerobic growth of Corynebacterium glutamicum (Ikeda et al. Biosci Biotechnol Biochem 73:2806–2808, 2009). In the present work, Cgl1427 was identified as a cytidylate kinase gene (cmk) by homology analysis of its deduced amino acid sequence with that of other bacterial cytidylate kinases (CMP kinases) and on the basis of findings that deletion of Cgl1427 results in loss of CMP kinase activity. Deletion of the cmk gene significantly impaired the growth of C. glutamicum in oxygen-limiting static culture, and the impaired growth was restored by introducing a plasmid containing the cmk gene, suggesting that this gene plays an important role in the microaerobic growth of C. glutamicum. On the other hand, in the main culture with aerobic shaking, a prolonged lag phase was observed in the cmk disruptant, despite an unchanged growth rate, compared to the behavior of the wild-type strain. The prolongation was observed when using seed culture grown to later growth stages in which oxygen limitation occurred, but it was not observed when using seed culture grown to an earlier growth stage in which oxygen remained relatively plentiful. Since nucleotide biosynthesis in C. glutamicum requires oxygen, we hypothesized that the ability of the cmk disruptant to synthesize nucleotides was influenced by oxygen limitation in the later growth stages of the seed culture, which caused the prolongation of the lag phase in the following shaken culture. To verify this hypothesis, a plasmid containing genes encoding all components of a homologous ribonucleotide reductase, a key enzyme for nucleotide synthesis that requires oxygen for its reaction, was introduced into the cmk disruptant, which significantly ameliorated the lag phase prolongation. Furthermore, this experimental setup almost completely restored the growth of the cmk disruptant in the oxygen-limiting static culture. These results indicate that CMP kinase plays an important role in normal nucleotide biosynthesis under an oxygen-limiting environment.  相似文献

Summary By transfection of an expression vector of human cytochrome P450 2E1 (CYP2E1) into a human hepatoma cell line (HLE), a new cell line (HLE/2E1) that stably expresses activity of CYP2E1 has been established. The HLE/2E1 cell line expressed a higher level of CYP2E1 messenger ribonucleic acid than did the mother HLE cell line. CYP2E1 enzyme activity determined by ap-nitrophenol oxidation assay was also higher in HLE/2E1 cells than in HLE cells. In addition, the enzyme activity of the HLE/2E1 cells was increased by ethanol treatment. Exposure to acetaminophen (APAP) or buthionine sulfoximine (BSO) caused a greater decrease in viability of the HLE/2E1 cells than that of the HLE cells, as determined by the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay. The cytotoxicity of APAP or BSO to HLE/2E1 cells was inhibited by the addition of ethanol or vitamin E. However, the cytotoxicity of both APAP and BSO was enhanced by 24-h preincubation of HLE/2E1 cells with ethanol. These results show that this cell line provides a useful model for studying catalytic properties of CYP2E1 and cytotoxic mechanisms of chemicals metabolized by CYP2E1.  相似文献
