Two NADPH-dependent alpha-keto ester reductases (Streptomyces thermocyaneoviolaceus keto ester reductase, STKER-II and -III) were purified from S. thermocyaneoviolaceus IFO 14271, one of thermophilic actinomycetes. The molecular masses of native STKER-II and -III were estimated to be 60 kDa and 70 kDa by gel filtration chromatography, respectively. These enzymes were both homodimers, with 29-kDa and 30-kDa subunit molecular masses based on SDS polyacrylamide gel electrophoresis. STKER-II and -III were stable from pH 7.0 to 10.0 and pH 5.5 to 9.0, respectively. Ethyl 3-methyl-2-oxobutanoate was reduced by both enzymes isolated to the corresponding (R)-hydroxy ester with excellent enantiomeric excess. STKER-III showed high stereoselectivity for the reduction of bulky substrates, while the selectivity of the STKER-II-catalyzed reduction was low except for ethyl 3-methyl-2-hydrox-ybutanoate. The spectral properties of the eyes of 3 species of diurnal and 8 species of nocturnal Japanese fireflies, in many cases males and females, were determined by an electroretinographic method. With the exception of Hotaria parvula males, which had a λmax of 580 nm, almost all species studied possessed a maximum around 500–540 nm. The eyes of diurnal and nocturnal species did not differ significantly in their sensitivity maxima. As in North American species of fireflies (Lall, 1981a,b) congruency existed between visual sensitivity peaks and light emission maxima in Luciola cruciata, L. lateralis and Hotaria parvula. In agreement with Seliger et al. (1982a,b) we conclude that an adaptation of the visual sensitivity to the light produced need not have occurred and that evolutionary adaptation of light emission to an existing ancestral green-sensitivity of the eye is the more likely course of events. 相似文献

Movement, nictation, and infectivity of Steinernema carpocapsae strain All were compared for ensheathed (EnJ) and desheathed (DeJ) infective juveniles exposed to the insecticides acephate, dichlorvos, methomyl, oxamyl, or permethrin. Nematode response to various solutions included normal sinusoidal movement, uncoordinated motion, twitching, convulsion or formation of a pretzel shape, an inactive "S" posture with fine twitching, or a quiescent straight posture. The DeJ displayed these movements at lower concentrations of each insecticide than did EnJ. In petri dish bioassays, insecticide-treated EnJ caused generally lower mortality in the common cutworm, Spodoptera litura, than did EnJ alone but caused greater insect mortality than did insecticides alone. Nematode response to chemicals was more clearly demonstrated by nictating behavior than by the movement bioassay. Nictation of DeJ was suppressed by the test chemicals at low concentrations, except for acephate and permethrin. Nictating EnJ or DeJ, regardless of chemical treatment, killed host insects faster than did non-nictating juveniles. Insecticides that enhance nictating behavior at certain concentrations may be used for mixed applications with nematodes. 相似文献

We studied the photosynthetic electron transfer system of membrane-bound and soluble cytochromec inChlorobium tepidum, a thermophilic green sulfur bacterium, using whole cells and membrane preparations. Sulfide and thiosulfate, physiological electron donors, enhanced flash-induced photo-oxidation ofc-type cytochromes in whole cells. In membranes,c-553 cytochromes with two (or three) heme groups served as immediate electron donors for photo-oxidized bacteriochlorophyll (P840) in the reaction center, and appeared to be closely associated with the reaction center complex. m-value of 180 mV. When isolated soluble cytochromec-553, which has an apparent molecular weight of 10 kDa and seems to correspond to the cytochromec-555 inChlorobium limicola andChlorobium vibrioforme, was added to a membrane suspension, rapid photo-oxidation of both soluble and membrane-bound cytochromesc-553 was observed. The oxidation of soluble cytochromec-553 was inhibited by high salt concentrations. In whole cells, photo-oxidation was observed in the absence of exogenous electron donors and re-reduction was inhibited by stigmatellin, an inhibitor of the cytochromebc complex. These results suggest that the role of membrane-bound and soluble cytochromec inC. tepidum is similar to the role of cytochromec in the photosynthetic electron transfer system of purple bacteria. 相似文献

]is the key enzyme regulating ethylene biosynthesis in higherplants. A complementary DNA encoding wound-induced ACC synthasefrom mesocarp of winter squash (Cucurbita maxima Duch.) fruitswas cloned, and its complete nucleotide sequence determined.The cloned cDNA contained an open reading frame of 1479 basepairs encoding a sequence of 493 amino acids. Identificationof the cDNA was accomplished by expression of active enzymein Escherichia coli harboring the cDNA and by the presence ofa partial amino acid sequence identical to that found in thepurified enzyme. A putative pyridoxal phosphate binding siteof the enzyme is suggested. Northern blot analysis showed thatthe ACC synthase gene was activated by tissue wounding, andits expression was repressed by ethylene. Background
Calcifying nested stromal epithelial tumor (CNSET) is a primary neoplasm of the liver, characterized by well-demarcated nests consisting of spindle and epithelioid cells with calcification and bone formation. An association of Cushing syndrome with CNSET has drawn attention, but the origin of CNSET has not been clarified.Case presentation
We report here the case of a 20-year-old male with Klinefelter syndrome who underwent liver resection for an increasing liver tumor that was pathologically diagnosed with CNSET. He was postoperatively followed up and received several examinations, and recurrences and extrahepatic lymph node metastases were detected on the 64th day after surgery. Chemoembolization and chemotherapy were not effective, leading to tumor progression with development of progressive liver failure, and the patient finally died 164 days after hepatectomy.Conclusions
This case suggests that an imbalance of hormones affects the genesis and progression of CNSET, and indicates the importance of closely following patients with CNSET by imaging with attention to hepatic recurrence and extrahepatic metastases.449.
We have purified a unique enzyme, α-amino--caprolactam racemase 945-fold from an extract of Achromobacter obae by Octyl—Sepharose CL-4B and Thiopropyl—Sepharose 6B and some other chromatographies. The purified enzyme was found homogeneous by sodium dodecyl sulfate—polyacrylamide gel electrophoresis and analytical ultracentrifugation. The enzyme has a monomeric structure with Mr 50 000 and a sedimentation coefficient (s20,w) of 4.28 S. The enzyme contains pyridoxal 5'-phosphate as a coenzyme. The pH optimum for the enzyme activity is 9.0. D- and L-α-amino--caprolactams are the only substrates. The Km values for the D- and L-isomers are, 8 and 6 mM, respectively. 相似文献

In order to investigate changes in levels of monoamines and their related substances together with those of other neurotransmitters (acetylcholine and GABA), choline and substances related to energy metabolism (ATP, lactate and glucose) accompanying incomplete cerebral ischemia, a bilateral common carotid artery occlusion model of spontaneously hypertensive rats (SHR) was utilized. Animals were subjected to 1 or 2 h ischemia. Then the concentrations of substances were measured in the cerebral cortex, hippocampus and striatum and compared with control values. Due to the incomplete ischemia, ATP showed a moderate decrease, while lactate and choline increased remarkably, and GABA underwent a moderate increase. With regard to monoamines, both noradrenaline and serotonin levels were reduced in the cerebral cortex and hippocampus, whereas dopamine levels increased in the hippocampus. All monoamine metabolites, i.e. metabolites by monoamine oxidase (MAO), metabolites by catechol-O-methyltransferase (COMT), and metabolites by both MAO and COMT, underwent increases. The 3-methoxytyramine level in particular showed marked increases. Furthermore levels of precursor amino acids as well as 5-hydroxytryptophan rose. Acetylcholine decreased moderately only in the cerebral cortex. Among these changes, sustained increases in all the monoamine metabolites were characteristic in the incompletely ischemic brain, suggesting that both COMT and MAO retain their activities in the incompletely ischemic brain. 相似文献
