Response of Steinernema carpocapsae Infective Juveniles to the Plasma of Three Insect Species

Exsheathed infective juveniles of Steinernema carpocapsae All strain were attracted to the plasma of three species of insects in agar plate bioassays. Plasma of Pieris rapae crucivora, Spodoptera litura, and Agrotis segetum attracted 88.6%, 80.4%, and 64.4%, respectively, of Steinernema carpocapsae juveniles added to plates. Autoclaved plasma of S. litura larvae attracted more juveniles than saline controls, but less than nonautoclaved plasma. The active agent passed through a 14,000 MW dialysis membrane.     

Wheat germ agglutinin, concanavalin A, and lens culinalis agglutinin block the inhibitory effect of nerve growth factor on cell-free phosphorylation of Nsp100 in PC12h cells

It has been shown that in PC12 and its subclone PC12h treatment of the cells with nerve growth factor (NGF) induces a selective decrease in the incorporation of radioactive phosphate into a 100,000-dalton protein, designated in an earlier study as Nsp100, in the subsequent phosphorylation of soluble extracts from cells with (gamma-32P)ATP. In the present study, we show that plant lectins, wheat germ agglutinin (WGA), concanavalin A (Con A), and lens culinaris agglutinin (LCA), inhibit the action of NGF on Nsp100 phosphorylation in PC12h cells. Treatment of the cells with WGA, which binds to N-acetylglucosamine and sialic acid residues on glycoproteins, strongly blocked the inhibitory action of NGF on the protein phosphorylation. Con A and LCA, both of which recognize the same specific sugars (mannose, glucose), displayed only a moderate blocking effect. Unlike the native lectin, succinylated WGA, which has the ability to bind to N-acetylglucosamine but not to sialic acid residues, and other lectins examined in this study did not inhibit the action of NGF on Nsp100. WGA-mediated inhibition of NGF action was reversed by the addition of N-acetylglucosamine and by the addition of a much lower concentration of a sialoglycoprotein, mucin, into the culture. Since the binding of succinylated WGA to N-acetylglucosamine residues of cell-surface glycoconjugates is not sufficient to prevent the action of NGF, WGA might act on sialic acid residues of the NGF receptor molecule to effect the inhibition of biological actions of NGF.     

Effect of estrogen on postovulatory LH surge in baboons.

In normally cycling female baboons, an LH surge appeared prior to ovulation, in addition, another LH surge (postovulatory LH surge) was observed within two days after ovulation. An attempt was then made to determine the effect of postovulatory LH on the luteinization of corpus luteum in baboons. Injections of 300 micrograms estradiol benzoate were given at 09.00 and 16.00 hr daily for 5 days following ovulation; the plasma level of LH was increased, but plasma progestin was suppressed. These results infer that the injected estrogen (estradiol benzoate) may inhibit the luteotrophic effect of postovulatory LH on the corpus luteum, therefore, plasma progestin remains lower even though postovulatory LH is elevated.     

Studies on inoculation of aerial algae. I. Semi-mass culture and quantification of algal color of Klebsormidium flaccidum (Ulotrichales, Chlorophyceae) sprayed on to concrete board

This paper describes methods of semi-mass culture and the quantification of algal color sprayed on a concrete board of the alga Klebsormidium flaccidum (Küitzing) S. Mattox et Blackwell. The alga was cultured in 100 L tanks normally used for hatching brine shrimp, with aeration from the deepest parts of the slanting sides. Initially mean algal density increased rapidly and attained 9.1 times the original density after 4 weeks. Subsequently, increase of the algal density was slow, and the density became 16.1 times the original level after 8 weeks. The alga cultured in this study was sprayed on to a concrete board using an agricultural sprayer. The relationship between the algal density on the board and its color, expressed as L*a*b* color space, was quantified. The chromaticity was maximum as the algal density on the board was about 5 g/m², and the color was most vivid. When the algal density was more than 20 g/m², both the lightness and chromaticity changed slightly, and the color became dark green.     

Prevalence of Genotypes of Orientia tsutsugamushi in Patients with Scrub Typhus in Miyazaki Prefecture

The genotypes of Orientia tsutsugamushi in patients with scrub typhus in Miyazaki Prefecture were examined by polymerase chain reaction (PCR) and the restriction fragment length polymorphism method. Specific patterns for genotypes Irie, Hirano, Tazume and Yoshimura were detected in 26, 6, 5 and 2 of 39 DNA samples obtained from peripheral blood mononuclear cells, respectively. DNA sequences of the PCR products from the Tazume strain were genetically very close to the Hirano strain and the Yoshimura strain was also very close to the Karp strain. Furthermore, the DNA sequences from the Irie and Tazume strains were completely homologous to the reported sequences of the Kawasaki and Kuroki strains, respectively.     

Immunochemical Difference of Wound-Induced 1-Aminocyclopropane-1-carboxylate Synthase from the Auxin-Induced Enzyme

Immunochemical cross-reactivity of wound- and auxin-induced1-aminocyclopropane-1-carboxylate (ACC) synthase was examinedwith the antibody against wound-induced ACC synthase purifiedfrom mesocarp of winter squash (Cucurbita maxima Duch.). Theantibody recognized ACC synthase from wounded hypocotyls ofwinter squash and from wounded pericarp of tomato fruits, butnot the enzyme from IAA-treated hypocotyls of winter squash,tomato and mung bean. These results indicate that the primarystructure of the wound-induced enzyme is different from thatof the auxin-induced enzyme in the same species, and impliesthat there are two different genes for ACC synthase, one forwound induction and the other for auxin induction.
