beta cells sense glucose through its metabolism and the resulting increase in ATP, which subsequently stimulates insulin secretion. Uncoupling protein-2 (UCP2) mediates mitochondrial proton leak, decreasing ATP production. In the present study, we assessed UCP2's role in regulating insulin secretion. UCP2-deficient mice had higher islet ATP levels and increased glucose-stimulated insulin secretion, establishing that UCP2 negatively regulates insulin secretion. Of pathophysiologic significance, UCP2 was markedly upregulated in islets of ob/ob mice, a model of obesity-induced diabetes. Importantly, ob/ob mice lacking UCP2 had restored first-phase insulin secretion, increased serum insulin levels, and greatly decreased levels of glycemia. These results establish UCP2 as a key component of beta cell glucose sensing, and as a critical link between obesity, beta cell dysfunction, and type 2 diabetes.  相似文献

Acca sellowiana has commercial potential due to the quality and the unique flavor of its fruit. Conservation of natural populations and management of breeding programmes would benefit from the availability of molecular markers that could be used to characterize levels and distribution of genetic variability. Thus, 13 microsatellite markers were developed from an enriched genomic library of A. sellowiana. They were characterized using 40 samples. The expected and observed heterozygosities ranged from 0.513 to 0.913 and from 0.200 to 0.889, respectively. These are the first microsatellite loci characterized from A. sellowiana that will contribute to improve researches on its genetic conservation, characterization and breeding.  相似文献

Adipose tissue plays an important role in glucose homeostasis and affects insulin sensitivity in other tissues. In obesity and type 2 diabetes, glucose transporter 4 (GLUT4) is downregulated in adipose tissue, and glucose transport is also impaired in muscle. To determine whether overexpression of GLUT4 selectively in adipose tissue could prevent insulin resistance when glucose transport is impaired in muscle, we bred muscle GLUT4 knockout (MG4KO) mice to mice overexpressing GLUT4 in adipose tissue (AG4Tg). Overexpression of GLUT4 in fat not only normalized the fasting hyperglycemia and glucose intolerance in MG4KO mice, but it reduced these parameters to below normal levels. Glucose infusion rate during a euglycemic clamp study was reduced 46% in MG4KO compared with controls and was restored to control levels in AG4Tg-MG4KO. Similarly, insulin action to suppress hepatic glucose production was impaired in MG4KO mice and was restored to control levels in AG4Tg-MG4KO. 2-deoxyglucose uptake during the clamp was increased approximately twofold in white adipose tissue but remained reduced in skeletal muscle of AG4Tg-MG4KO mice. AG4Tg and AG4Tg-MG4KO mice have a slight increase in fat mass, a twofold elevation in serum free fatty acids, an approximately 50% increase in serum leptin, and a 50% decrease in serum adiponectin. In MG4KO mice, serum resistin is increased 34% and GLUT4 overexpression in fat reverses this. Overexpression of GLUT4 in fat also reverses the enhanced clearance of an oral lipid load in MG4KO mice. Thus overexpression of GLUT4 in fat reverses whole body insulin resistance in MG4KO mice without restoring glucose transport in muscle. This effect occurs even though AG4Tg-MG4KO mice have increased fat mass and low adiponectin and is associated with normalization of elevated resistin levels.  相似文献

A reversed-phase high-performance liquid chromatography (RP-HPLC) methodology for the qualitative and quantitative analysis of human thyrotropin (hTSH) in CHO cell conditioned medium and in purified preparations has been set up and validated for accuracy, precision and sensitivity. A recovery test indicated a bias of less than 2% and intra-day and inter-day quantitative determinations presented relative standard deviations (RSD) always <7%, while sensitivity was 0.2 microg (RSD=5.6%). The novel methodology was applied to the study of the best cultivation conditions and was able to detect a significant difference in retention time (t(R)) between pituitary and recombinant hTSH, probably reflecting the influence of the heterogeneity of the carbohydrate moiety on the hydrophobic properties of the molecule.  相似文献

Background

Traditional raft (jangada), piúba wood raft (jangada de pau de piúba), six-log raft (jangada de seis paus), and wooden raft (jangada de pau) are some of the names given to the traditional Brazilian watercrafts created from the buoyancy of bound logs. The traditional raft is a watercraft used and built by artisan fishermen who have, throughout generations, kept and improved knowledge related to this practice and the use of the plant species they need as raw materials. Active groups of these fishermen and their watercrafts are distributed along 200?km of the coast of the state of Bahia. The fishermen interviewed in this study are at the southern limit of distribution for the use of this type of vessel.

Methods

This study aimed to characterize the use of the arboreal species applied in the construction of the traditional raft in the municipalities of Uruçuca, Ilhéus, and Canavieiras in the southern State of Bahia, Brazil. For this purpose, structured and semi-structured interviews were individually conducted with 36 fishermen, and walking tours were conducted with specialists in the construction of the watercraft.

Results

We observed that the raftsmen use 21 species to construct the traditional raft. The features of the wood, such as density, flexibility, and availability, are the main criteria applied to choose the arboreal species. Some species are preferred, such as pau de jangada (Apeiba tibourbou) and biriba (Eschweilera ovata), which are the most frequently employed in watercraft manufacturing.

Conclusions

The southern Bahia population is familiar with the different tree species that are linked to their fishing activities. The main link between the fishermen and the useful species is present in the practice of raft construction. Currently, the restricted access to raw materials limits this practice, which consequently results in the cultural erosion of this community.

Global warming is expected to cause significant changes in the distribution of species worldwide, altering ecosystem function and services. These impacts can be exacerbated by synergism with other global change drivers, such as biological invasions. Furcraea foetida (Asparagaceae) is a CAM species native to Central America and northern South America that is currently invading coastal ecosystems in the Atlantic Forest biome in Brazil. The species propagates clonally by bulbils, forming dense clusters that exclude native species. We addressed whether warming may favor or impair development of F. foetida bulbils based on functional traits and assessed whether plant cover of native species influenced F. foetida establishment in a rocky shore ecosystem. We used open-top chambers during summer and autumn, which increased air temperature during daylight hours by 0.4 °C relative to ambient conditions. This short-term warming effect increased the leaf length of bulbils but had no effect on number of leaves, dry mass, specific leaf area, leaf dry matter content (LDMC), or photosynthetic efficiency of photosystem II (Fv/Fm). However, an increase in vegetation cover decreased LDMC and helped maintain high Fv/Fm, having an indirect positive effect on F. foetida establishment that suggests a facilitation interaction in this extreme environment. Even though the manipulated temperature did not reach IPCC (Intergovernmental Panel on Climate Change) forecasts for 2100, our data show that even small, short-term changes in temperature affected plant performance. Thus, we suggest that F. foetida may be favored in a scenario of climate change, increasing its negative effects on biodiversity of coastal ecosystems.  相似文献

Traditional Knowledge and Management of Caryocar coriaceum Wittm. ( Pequi ) in the Brazilian Savanna, Northeastern Brazil Caryocar coriaceum Wittm. (pequi) is a tree native to northeastern Brazil that produces fruits of importance to human populations of the region. This species serves as a resource for the livelihoods of many people and represents a source of income in many rural communities in northeastern Brazil. This study aimed to evaluate the knowledge, use, collection practices, and forms of local management related to C. coriaceum in a rural community in the state of Ceará, Chapada do Araripe, northeast Brazil. Pequi collectors were identified and constituted the sample of respondents (N = 56). The knowledge of C. coriaceum is uniformly distributed in the community. The categories food and derivatives (oil) were the most relevant. Considering all use categories, there was a significant difference between the knowledge of men and women (p <0.05, 95% confidence interval) based on the Shannon-Wiener and Hurlbert’s PIE diversity indices. The main form of management applied to pequi trees was collection of its fruits, followed by the practice locally referred to as zelar (similar to the management practice “protection”) and sowing seeds. All of the informants stated that they commercialize pequi fruits. The mean income from fruit sales is less than the mean income obtained via selling pequi oil, as the latter product is of greater commercial value. Among the nine most relevant collection sites, Barreiro Novo (savanna) was highlighted, which is part of both a protected area and the anthropogenic landscape of the region, and has great economic and cultural importance for pequi collectors.  相似文献

Background

Knowing the national disease burden of severe influenza in low-income countries can inform policy decisions around influenza treatment and prevention. We present a novel methodology using locally generated data for estimating this burden.

Methods and Findings

This method begins with calculating the hospitalized severe acute respiratory illness (SARI) incidence for children <5 years old and persons ≥5 years old from population-based surveillance in one province. This base rate of SARI is then adjusted for each province based on the prevalence of risk factors and healthcare-seeking behavior. The percentage of SARI with influenza virus detected is determined from provincial-level sentinel surveillance and applied to the adjusted provincial rates of hospitalized SARI. Healthcare-seeking data from healthcare utilization surveys is used to estimate non-hospitalized influenza-associated SARI. Rates of hospitalized and non-hospitalized influenza-associated SARI are applied to census data to calculate the national number of cases. The method was field-tested in Kenya, and validated in Guatemala, using data from August 2009–July 2011. In Kenya (2009 population 38.6 million persons), the annual number of hospitalized influenza-associated SARI cases ranged from 17,129–27,659 for children <5 years old (2.9–4.7 per 1,000 persons) and 6,882–7,836 for persons ≥5 years old (0.21–0.24 per 1,000 persons), depending on year and base rate used. In Guatemala (2011 population 14.7 million persons), the annual number of hospitalized cases of influenza-associated pneumonia ranged from 1,065–2,259 (0.5–1.0 per 1,000 persons) among children <5 years old and 779–2,252 cases (0.1–0.2 per 1,000 persons) for persons ≥5 years old, depending on year and base rate used. In both countries, the number of non-hospitalized influenza-associated cases was several-fold higher than the hospitalized cases.

Conclusions

Influenza virus was associated with a substantial amount of severe disease in Kenya and Guatemala. This method can be performed in most low and lower-middle income countries.  相似文献

Different species of Leishmania can cause a variety of medically important diseases, whose control and treatment are still health problems. Telomere binding proteins (TBPs) have potential as targets for anti-parasitic chemotherapy because of their importance for genome stability and cell viability. Here, we describe LaTBP1 a protein that has a Myb-like DNA-binding domain, a feature shared by most double-stranded telomeric proteins. Binding assays using full-length and truncated LaTBP1 combined with spectroscopy analysis were used to map the boundaries of the Myb-like domain near to the protein only tryptophan residue. The Myb-like domain of LaTBP1 contains a conserved hydrophobic cavity implicated in DNA-binding activity. A hypothetical model helped to visualize that it shares structural homology with domains of other Myb-containing proteins. Competition assays and chromatin immunoprecipitation confirmed the specificity of LaTBP1 for telomeric and GT-rich DNAs, suggesting that LaTBP1 is a new TBP.  相似文献
