The distribution and accumulation of Cu2+, Zn2+ and Cd2+ ionsin the roots of Polygonum cuspidatum, collected from metalliferousand non-metalliferous habitats, were examined. About 90% ofthe metal ions was located in the cell wall fraction of rootsof plants grown in either type of habitat. The metal-ion exchangecapacity of the cell walls is not specific to the plants frommetalliferous habitats, and there were no significant differencesamong plants from the various habitats. The capacity for Cu2+ions was greater than that for Zn2+ or Cd2+ ions. Cu2+ ionshave a high affinity for the cell wall and, thus, it appearsthat the cell wall is a barrier for the transportation of Cu2+into the cytoplasm. An attempt to define a severe suppression of cell-mediated immunity by street rabies virus infection was undertaken by using the mice lethally and peripherally infected with a street rabies virus (1088 strain). The cell-mediated cytotoxic (CMC) activity of the spleen cells from those mice once slightly increased until day 4 after infection but declined rapidly thereafter until their death on days 10 to 12 after infection. In parallel with a decrease of CMC response of the spleen cells from 1088-infected mice, proliferative response to Con A, IL-2 activity in the culture supernatants of Con A-induced proliferation, responsiveness to exogenously added IL-2 and to Con A to express IL-2R, of those cells became suppressed, and the marked decrease of the total number of spleen cells was observed. Selective depletion of CD4+ and CD8+ cells in the spleens, abnormalities of IL-1 and E-type prostaglandins (PGE2) production or production of inhibitory component able to block IL-2 activity by spleen cells were not observed and these factors did not appear to be associated with the suppression of proliferative response to Con A. However, an apparent association of CD8+ cells in the suppression of differentiation of pre-cytotoxic lymphocytes (CTL) into CTL was demonstrated in the co-culture experiments of the spleen cells from 1088-infected mice with spleen cells of mice infected with an attenuated rabies virus (ERA strain) which can induce higher levels of CMC response. There was no evidence of the productive replication of rabies virus in thymus and spleen of 1088-infected mice. The relationship of these observations to current theories on virus-induced immunosuppression was discussed. 相似文献

To investigate the mechanism of Cu tolerance of Polygonum cuspidatum,the Cu-binding protein was isolated from the cytoplasm of theroot and rhizome from plants growing in a Cu-contaminated habitat.A Cu-containing compound with a molecular weight of 9,500 wasobtained from the cytoplasm of the root and the cortex of therhizome by Bio-Gel P-30 column chromatography. The amount ofthis compound increased with increasing Cu content in the cytoplasm.The compound was purified by passage through a DEAE-CellulofineAL column and further by high performance liquid chromatography.The purified compound was a metallothionein-like protein, becauseit had high contents of cysteine (31.3%) and acidic amino acids(16.5%) and a very low proportion of aromatic amino acids. Cells of the roots ofA. yokoscense growing on metalliferous habitats were fractionated into their cell wall and cytoplasmic components. About 70–90% of the
total copper, zinc and cadmium was located in the cell wall. Copper had a markedly greater affinity for the cell wall than
zinc and cadmium, and was prevented from entering the cytoplasm. A large proportion of these heavy metals in the cell wall
were exchanged as ions. The capacity of the cell wall for exchanging metal ions inA. yokoscense was higher than in other plants growing on metalliferous habitats. However, compared with different ferns unable to grow
on metalliferous habitats, this capacity was not unique toA. yokoscense. Consequetly, the root cell wall ofA. yokoscense is considered to be an important site of metal ion storage and may play the role of an excretory organ for heavy metals.
On the other hand, as proportion of the heavy metls was transported to the cytoplasm, where the metal content was much higher

Angiogenesis is an essential process in the progression of malignant tumors. However, little is known of the angioarchitecture in primary oral malignant melanoma. We sought to determine this by the use of periodic acid-Schiff (PAS) stain, endothelial markers (CD34, CD105) and laminin, and by transmission electron microscopy in two cases. The results demonstrated that endothelium-lined vessels dominated the tumor microvasculature and these stained positively for PAS, laminin, and endothelial markers. Mosaic and tumor-lined vessels were infrequently encountered. Most PAS-positive patterned networks and loops ultrastructurally represented intratumor microhemorrhages that probably arose secondary to tumor vessel leakiness. Vascular channels of the vasculogenic mimicry type were rare. They stained for laminin but not for endothelial markers. 相似文献
