Summary The changes in the serum glycoproteins and the development of specific antibodies were examined during the course of experimental coccidioidomycosis in rats. The total glycoprotein, seromucoid hexose, seromucoid protein, and haptoglobin levels were signigicantly higher in sera from infected animals compared with sera from non-infected controls. These changes were evident at three days but not at two weeks following inoculation withC. immitis. The non-seromucoid hexose and total protein concentrations were not significantly different between infected animals and controls. Photosystem I (PS I) is a large pigment–protein complex embedded in the thylakoid membranes that performs light-driven electron transfer across the thylakoid membrane. Carbon nanotubes exhibit excellent electrical conductivities and excellent strength and stiffness. In this study, we generated PSI–carbon nanotube conjugates dispersed in a solution aimed at application in artificial photosynthesis. PS I complexes in which a carbon nanotube binding peptide was introduced into the middle of the PsaE subunit were conjugated on a single-walled carbon nanotube, orienting the electron acceptor side to the nanotube. Spectral and photoluminescence analysis showed that the PS I is bound to a single-walled carbon nanotube, which was confirmed by transmission electron microscopy. Photocurrent observation proved that the photoexcited electron originated from PSI and transferred to the carbon nanotube with light irradiation, which also confirmed its orientated conjugation. The PS I–carbon nanotube conjugate will be a useful nano-optoelectronic device for the development of artificial systems. 相似文献

Environmental changes influence foraging behavior for most animals. Dolphinfish, Coryphaena hippurus, are epipelagic predators and have a cosmopolitan tropical to warm-temperate (>20°C) distribution. We simultaneously obtained
the ambient temperature and the foraging behavior (i.e., swimming speed, depth and tailbeat acceleration) of dolphinfish,
using an acceleration data-logger in May, September, October, November 2007, June 2008, May and July 2010 for 8 individuals.
Although the dolphinfish spent a mean ± standard deviation of 43.4 ± 27.7% of their time at the surface (0–5 m), dive excursions
from the surface (DES) were observed in all individuals and maximum DES depths ranged from 50.1 to 95.4 m. DES events resulted
dives below the thermocline for these dolphinfish, and there was a significantly positive relationship between the isothermal
layer depth (ILD) and DES depth. Our results demonstrate that dolphinfish avoided the rapid thermal change beyond the thermocline,
and their prey is most likely found in the upper layers of the thermocline. Gliding behavior during the DES phase was also
observed and dolphinfish gradually descended to deeper waters with gliding. The gliding time was longer when the ILD was deeper,
and fish tended to dive deeper. We suggest that dolphinfish adopt gliding behavior to search a broader range of depths for

Three enzyme forms (CR1, CR2 and CR3) of carbonyl reductase were purified from chicken liver with using 4-benzoylpyridine as a substrate. CR1 was a dimeric enzyme composed of two identical 25-kD subunits. CR2 and CR3 were monomeric enzymes whose molecular weights were both 32 kD. CR1 exhibited 17 beta-hydroxysteroid dehydrogenase activity as well as carbonyl reductase activity in the presence of both NADP(H) and NAD(H). CR2 and CR3 had similar properties with regard to substrate specificity and inhibitor sensitivity. They could exhibit the activity only with NADPH and had no hydroxysteroid dehydrogenase activity. CR2 and CR3 cross-reacted with anti-chicken kidney carbonyl reductase antibody, though CR1 did not. The results suggest that CR1 is a hydroxysteroid dehydrogenase, and CR2 and CR3 are similar to each other and to the kidney enzymes. 相似文献

The effect of light and temperature on the growth and photosynthesis of the Japanese agarophyte, Gracilariopsis chorda (Gracilariaceae, Rhodophyta), was determined to better understand its physiology so that we could identify candidates for mass cultivation. Above the photosynthetic active radiation of 66 μmol photons m?2 s?1, photosynthetic rates saturated for all strains that were collected from six different locations (Hokkaido, Chiba, Tokushima, Saga, Kagoshima, and Okinawa); furthermore, either photosynthesis or growth was observed at all temperature treatments examined in our study (4–32 °C for photosynthesis, 16–32 °C for growth experiments). We identified a temperature range for optimal photosynthesis and growth, which occurred within 20.1–29.1 °C and roughly correlated with the water temperatures of the collection locations and strongly suggests that this species tolerates a wide variety of water temperature. In particular, the Kagoshima strain had the widest range of optimal temperatures (20.8–29.1 °C), whereas the Saga strain had the narrowest range (23.1–27.3 °C). It is important to note that all the optimal temperature ranges overlapped among the strains; therefore, no definitive distinction can be determined. The broad tolerance to temperatures commonly observed from northern to southern Japan suggests that the cultivation of this species should succeed during spring to summer in the majority of the coastal regions in Japan. 相似文献

The responses of photochemical efficiency to desiccation and salinity gradients in an intertidal edible brown macroalga, Sargassum fusiforme (Harvey) Setchell (Sargassaceae, Fucales), were determined using a pulse amplitude modulation (PAM)-chlorophyll fluorometer. The effective quantum yields (ΔF/Fm'; = ΦPSII) of photosystem II (PSII) dropped to zero after 360-min aerial exposure under low irradiance (20 μmol photons m−2 s−1) and 120-min exposure under high irradiance (700 μmol photons m−2 s−1) for this species at 20°C and 50% relative humidity. Under these conditions, ΔF/Fm' failed to recover to initial levels even after 1-day rehydration in seawater. In general, ΔF/Fm' decreased as desiccation reduced the absolute water content (AWC, %). Nevertheless, when AWC was above ca. 20%, ΔF/Fm' was mostly restored to initial levels after 1-day rehydration in seawater, suggesting strong tolerance to dehydration. Furthermore, S. fusiforme appeared to tolerate a broad range of salinity (i.e. 15–50 psu) during six days of culture; however, ΔF/Fm' declined when salinity was <10 and 60 psu. Strong tolerance to dehydration and salinity stress likely provides S. fusiforme an advantage that allows it to flourish in the intertidal habitat. 相似文献

In order to synthesize interferon‐γ glycoform having an oligosaccharide at the 97 position by a semi‐synthetic method, interferon‐γ‐polypeptide‐(1–94)‐α‐hydrazide was prepared by the specific Cys‐cyanylation of polypeptide‐(1–94)‐Cys‐His6 expressed from E. coli and subsequent hydrazinolysis in 22% yield (two steps). This polypeptide‐α‐hydrazide was then converted into corresponding polypeptide‐α‐thioester under NaNO2/acid conditions followed by thiolysis in 83% yield. When the spores of Bacillus megaterium QM B1551 (ATCC 12872) were incubated with 5 mM CdCl2 at 30 C, they underwent the early germination events, such as loss of heat resistance and release of calcium dipicolinate, in the same way as when they were germinated by glucose + KNO3. However, germination by CdCl2 caused no increase in the reducing groups in the cortex and no excretion of glucosamine-containing materials due to the hydrolysis of the cortex peptidoglycan. Addition of CdCl2 at any time during germination by glucose + KNO3 inhibited the release of glucosamine-containing materials from the spores, whereas removal of cadmium from the CdCl2-germinated spores by treatment with cysteine restored the hydrolysis of peptidoglycan. These results suggested that CdCl2 caused the early events of spore germination but prevented the spores from undergoing the events following germination by inhibiting the enzymatic lysis of the cortex peptidoglycan. The conclusion from the study is that cortex degradation is not always required for the initiation of germination. 相似文献
