Cephalosporins are widely used as anti-infectious β-lactam antibiotics in clinic. For the purpose of increasing the yield
of cephalosporin C (CPC) fermentation, especially in an industrial strain, A. chrysogenum genes cefEF and cefG, which encode the ultimate and penultimate steps in CPC biosynthesis, cefT, which encodes a CPC efflux pump, and vgb, which encodes a bacterial hemoglobin gene were transformed in various combinations into an industrial strain of A. chrysogenum. Both PCR and Southern blotting indicated that the introduced genes were integrated into the chromosome of A. chrysogenum. Carbon monoxide difference spectrum absorbance assay was performed and the result showed that Vitreoscilla hemoglobin was successfully expressed in A. chrysogenum and had biological activity. HPLC analysis of fermentation broth of recombinant A. chrysogenum showed that most transformants had a higher CPC production level than the parental strain. Multiple transformants containing
an additional copy of cefG showed a significant increase in CPC production. However, cefT showed little effect on CPC production in this high producer. The highest improvement of CPC titer was observed in the mutant
with an extra copy of cefG + cefEF + vgb whose CPC production was increased by 116.3%. This was the first report that three or more genes were introduced simultaneously
into A. chrysogenum. Our results also demonstrated that the combination of these genes had a synergy effect in a CPC high producer. 相似文献

The diversity and genetic relationship among 29 populations of Chrysanthemum morifolium, one of Chrysanthemum indicum and one of Chrysanthemum nankingense from China were analyzed using morphological traits and molecular markers. Twenty morphological traits were scored as well as 182 ISSR marker-fragments, as amplified by 22 primers [the percentage of polymorphic bands (PPB): 81.87%], and 243 SRAP marker-fragments as generated by 26 primer pairs (PPB: 75.72%). Mantel’s test indicated significant correlation (r = 0.624) of morphological trait and SRAP. By contrast, the morphological trait showed low correlation with ISSR (r = 0.246). Cluster analysis showed groupings of the accessions according to all four methods correlated well with their geographic region of origin, and most populations from the south of China were classified into one cluster and most populations from the north of China were classified into another cluster. Finally, an appropriate strategy for conserving the C. morifolium germplasm was proposed. 相似文献

生态系统服务供需关系研究是自然资源开发和生态环境保护空间差异化管控的重要依据,基于主体功能区划视角的研究对构建生态安全格局、促进区域经济可持续发展具有重要意义。以湖南省各县域为研究单元构建湖南省生态系统服务供需评估指数,运用ESSD系数、局部空间自相关等方法,将湖南省主体功能区划实施前(2006-2011年)和实施后(2012-2017年)两个时间段作对比,分析生态系统服务供需关系的时空变化及空间相关性特征。研究结果表明:(1)主体功能区划政策对湖南省有效保护生态系统,稳定供需关系产生积极导向作用。(2)湖南省生态系统服务供需系数存在显著的空间异质性,始终呈现西高东低的空间分布特征。各类功能区的生态系统服务供需关系存在较大差异,重要开发区主要表现为供不应求,重要生态功能区主要表现为供过于求,农产品主产区主要表现为供需均衡。(3)湖南省生态系统服务供需关系空间负相关性增强,生态系统服务供需空间平衡性差异加剧。 相似文献

Miscanthus spp. show excellent application prospects due to its bioenergy potential and multiple ecological services. Annual N export with biomass harvest from Miscanthus, even without fertilizer supplement, do not reduce soil N levels. The question arises regarding how Miscanthus can maintain stable soil N levels. Metagenomic strategies were used to reveal soil N-cycling-related microbiome and their functional contributions to processes of soil N-cycling based on the comparison among the bare land, cropland, 10-year Miscanthus × giganteus, and 15-year Miscanthus sacchariflorus fields. The results showed that, after long-term cropland-to-Miscanthus conversion (LCMC), 16 of 21 bacterial phyla and all the archaeal phyla exhibited significant changes. Soil microbial denitrification and nitrification functions were significantly weakened, and N fixation (NF) was significantly enhanced. The biosynthesis of amino acids, especially alanine, aspartate, and glutamate metabolism, in soil N-cycling-related microbiome was dramatically promoted. The genus Anaeromyxobacter contributed largely to the NF process after LCMC. Variations in the soil available potassium, available N, organic C, and total N contents drove a functional shift of soil microbiome from cropland to Miscanthus pattern. We conclude that Miscanthus can recruit Anaeromyxobacter communities to enhance NF benefiting its biomass sustainability and soil N balance. 相似文献

The purpose of this study was to investigate the biological effect of miR‐16 on myocarditis and the underlying molecular mechanism. H9c2 cells were treated with 10 µg/mL lipopolysaccharide (LPS) for 12 hours to form a myocarditis injury model. We observed that LPS treatment distinctly decreased the level of miR‐16 in H9c2 cells. Upregulation of miR‐16 increased cell proliferation and reduced cell apoptosis. Then, CD40 was predicted and verified as a target gene of miR‐16 by TargetScan and luciferase reporter assay, respectively. Furthermore, the messenger RNA and protein expression of CD40 are negatively regulated by miR‐16. The relative expression of inflammatory factors was dramatically decreased by the miR‐16 mimic. Cells cotransfected with miR‐16 mimic and si‐CD40 could significantly abolish the injury of cardiomyocytes caused by myocarditis. Our study illustrated that the upregulation of miR‐16 has a protective effect on LPS‐damaged H9c2 cells, which may be achieved by regulating CD40 and the nuclear factor kappa B pathway. 相似文献

分析了来自同一细胞系的体细胞克隆牛甜甜、庆庆、浒娃及来源同一培养转基因体细胞系转基因体细胞克隆牛九妹、乐娃和1个妊娠8个月转基因流产胎牛8C2以及随机抽取的1头鲁西黄牛(LX)、1头褐斯坦牛(HS)在24个微卫星位点标记牛的基因型.结果表明24个多态位点均表现出多态,等位基因数为1~5个,平均为3.17个.根据网上公布的数据,按其最高频率计算,甜甜、庆庆、浒娃、九妹、乐娃、8C2与培养细胞系、转基因细胞系间匹配概率为1.17×10-36,根据本研究观察到的数据计算,匹配概率为1.90×10-23;而与随机抽取的1头鲁西黄牛及褐斯坦牛的基因型分别在23和20个位点上完全不同. 相似文献
