The Pac1 ribonuclease of Schizosaccharomyces pombe is a member of the RNase III family of double-strand-specific ribonucleases. To examine RNA structural features required for efficient cleavage by the Pac1 RNase, we tested a variety of double-stranded and hairpin RNAs as substrates for the enzyme. The Pac1 RNase required substrates that have a minimal helix length of about 20 base pairs. The enzyme cut both strands of the helix at sites separated by two base pairs. However, Pac1 was also able to make a single-stranded cleavage within an internal bulge of an authentic Escherichia coli substrate at the same site chosen by RNase III. Pac1 efficiently degraded the structurally complex adenovirus VA RNA(I), but was inactive against the short HIV-1 TAR RNA hairpin. These results indicate that the Pac1 RNase prefers straight, perfect helices, but it can tolerate internal bulges that do not distort the helix severely. Like its homologue from Saccharomyces cerevisiae, the Pac1 RNase cleaved at two in vivo RNA processing sites in a hairpin structure in the 3' external transcribed spacer of the S. pombe pre-rRNA, suggesting a role for the enzyme in rRNA maturation. 相似文献

以小米(Setaria italica)为材料,克隆了含有叶绿体psbA基因的2.2kb EcoRⅠ片段,测定了该基因5'末端非编码区的核苷酸序列。序列分析显示psbA基因5'末端非编码区存在着与原核类似的启动子结构,其“-10”区的序列为TATACT,与原核生物“-10”共有基序(Consensus motif)仅相差一个核苷酸;其“-35”区的序列为TTGACA,与原核生物“-35”共有基序完全相同。另外,在“-10”区和“-35”区之间还存在着一个类似真核启动子结构的“TATATA”保守序列。这些结果表明小米psbA基因的启动子既具有原核的特征又具有真核的特征。小米psbA基因的mRNA前导序列长87bp,与高粱完全一致,而比水稻多出了“CTATTTT”7个核苷酸,比小麦、大麦和黑麦多出了“TTTT”4个核苷酸。因此推测在禾本科的C_3和C_4植物之间,psbA基因mRNA前导序列区的差异可能具有普遍性。计算机分析结果显示,以上6种植物的psbA基因mRNA前导序列区内均能形成小的茎环结构,而且这段“CTATTTT”额外序列恰好位于茎环结构中,造成了6种植物间茎环大小的差异。这一小的二级结构可能对psbA基因的表达调控有一定的影响。 相似文献

Individual differences in mind and behavior are believed to reflect the functional variability of the human brain. Due to the lack of a large-scale longitudinal dataset, the full landscape of variability within and between individual functional connectomes is largely unknown. We collected 300 resting-state functional magnetic resonance imaging (rfMRI) datasets from 30 healthy participants who were scanned every three days for one month. With these data, both intra- and inter-individual variability of six common rfMRI metrics, as well as their test-retest reliability, were estimated across multiple spatial scales. Global metrics were more dynamic than local regional metrics. Cognitive components involving working memory, inhibition, attention, language and related neural networks exhibited high intra-individual variability. In contrast, inter-individual variability demonstrated a more complex picture across the multiple scales of metrics. Limbic, default, frontoparietal and visual networks and their related cognitive components were more differentiable than somatomotor and attention networks across the participants. Analyzing both intra- and inter-individual variability revealed a set of high-resolution maps on test-retest reliability of the multi-scale connectomic metrics. These findings represent the first collection of individual differences in multi-scale and multi-metric characterization of the human functional connectomes in-vivo, serving as normal references for the field to guide the use of common functional metrics in rfMRI-based applications. 相似文献

Background and Aims
It has been proposed that variation in pollinator preferences or a fluctuating environment can act to maintain flower colour polymorphism. These two hypotheses were tested in an aquatic monocot Butomus umbellatus (Butomaceae) with a pink or white gynoecium in the field population.Methods
Pollinator visitation was compared in experimental arrays of equivalent flowering cymes from both colour morphs. Seed set was compared between inter- and intramorph pollination under different water levels to test the effect of fluctuating environment on seed fertility.Key Results
Overall, the major pollinator groups did not discriminate between colour morphs. Compared with the white morph, seed production in the pink morph under intermorph, intramorph and open pollination treatments was significantly higher when the water level was low but not when it was high. Precipitation in July was correlated with yearly seed production in the pink morph but not in the white morph.Conclusions
The results indicated that the two colour morphs differed in their tolerance to water level. Our study on this aquatic plant provides additional evidence to support the hypothesis that flower colour polymorphism can be preserved by environmental heterogeneity. 相似文献998.

Peptidoglycan recognition proteins (PGRPs), which have been identified in most animals, are pattern recognition molecules that involve antimicrobial defense. Resulting from extraordinary expansion of innate immune genes, the amphioxus encodes many PGRPs of diverse functions. For instance, three isoforms of PGRP encoded by Branchiostoma belcheri tsingtauense, termed BbtPGRP1~3, are fused with a chitin binding domain (CBD) at the N-terminus. Here we report the 2.7 Å crystal structure of BbtPGRP3, revealing an overall structure of an N-terminal hevein-like CBD followed by a catalytic PGRP domain. Activity assays combined with site-directed mutagenesis indicated that the individual PGRP domain exhibits amidase activity towards both DAP-type and Lys-type peptidoglycans (PGNs), the former of which is favored. The N-terminal CBD not only has the chitin-binding activity, but also enables BbtPGRP3 to gain a five-fold increase of amidase activity towards the Lys-type PGNs, leading to a significantly broadened substrate spectrum. Together, we propose that modular evolution via domain shuffling combined with gene horizontal transfer makes BbtPGRP1~3 novel PGRPs of augmented catalytic activity and broad recognition spectrum. 相似文献

Aims Grassland is the most widely distributed vegetation type on the Xizang Plateau. Accurate remote sensing estimation of the grassland aboveground biomass (AGB) in this region is influenced by the types of vegetation indexes (VIs) used, the grain size (resolution) of the remote sensing data and the targeted ecosystem features. This study attempts to answer the following questions: (i) Which VI can most accurately reflect the grassland AGB distribution on the Xizang Plateau? (ii) How does the grain size of remote sensing imagery affect AGB reflection? (iii) What is the spatial distribution pattern of the grassland AGB on the plateau and its relationship with the climate?Methods We investigated 90 sample sites and measured site-specific AGBs using the harvest method for three grassland types (alpine meadow, alpine steppe and desert steppe). For each sample site, four VIs, namely, Normalized Difference VI (NDVI), Enhanced VI, Normalized Difference Water Index (NDWI) and Modified Soil-Adjusted VI (MSAVI) were extracted from the Moderate Resolution Imaging Spectroradiometer (MODIS) products with grain sizes of 250 m and 1 km. Linear regression models were employed to identify the best estimator of the AGB for the entire grassland and the three individual grassland types. Paired Wilcoxon tests were applied to assess the grain size effect on the AGB estimation. General linear models were used to quantify the relationships between the spatial distribution of the grassland AGB and climatic factors.Important findings The results showed that the best estimator for the entire grassland AGB on the Xizang Plateau was MSAVI at a 250 m grain size (MSAVI 250 m). For each individual grassland type, the best estimator was MSAVI at a grain size of 250 m for alpine meadow, NDWI at a grain size of 1 km for alpine steppe and NDVI at a grain size of 1 km for desert steppe. The explanation ability of each VI for the grassland AGB did not significantly differ for the two grain sizes. Based on the best fit model (AGB =-10.80 + 139.13 MSAVI 250 m), the spatial pattern of the grassland AGB on the plateau was characterized. The AGB varied from 1 to 136g m ?2. Approximately 59% of total spatial variation in the AGB for the entire grassland was explained by the combination of the mean annual precipitation (MAP) and mean annual temperature. The explanatory power of MAP was weaker for each individual grassland type than that for the entire grassland. This study illustrated the high efficiency of the VIs derived from MODIS data in the grassland AGB estimation on the Xizang Plateau due to the vegetation homogeneity within a 1×1 km pixel in this region. Alkaliphiles are considered more suitable chassis than traditional neutrophiles due to their excellent resistance to microbial contamination. Alkaliphilic Bacillus sp. N16-5, an industrially interesting strain with great potential for the production of lactic acid and alkaline polysaccharide hydrolases, can only be engineered genetically by the laborious and time-consuming homologous recombination. In this study, we reported the successful development of a CRISPR/Cas9-based genome editing system with high efficiency for single-gene deletion, large gene fragment deletion and exogenous DNA chromosomal insertion. Moreover, based on a catalytically dead variant of Cas9 (dCas9), we also developed a CRISPRi system to efficiently regulate gene expression. Finally, this efficient genome editing system was successfully applied to engineer the xylose metabolic pathway for the efficient bioproduction of D -lactic acid. Compared with the wild-type Bacillus sp. N16-5, the final engineered strain with XylR deletion and AraE overexpression achieved 34.3% and 27.7% increases in xylose consumption and D -lactic acid production respectively. To our knowledge, this is the first report on the development and application of CRISPR/Cas9-based genome editing system in alkaliphilic Bacillus, and this study will significantly facilitate functional genomic studies and genome manipulation in alkaliphilic Bacillus, laying a foundation for the development of more robust microbial chassis. 相似文献
