Inherited retinal degeneration due to loss of photoreceptor cells is a leading cause of human blindness. These cells possess a photosensitive outer segment linked to the cell body through the connecting cilium (CC). While structural defects of the CC have been associated with retinal degeneration, its nanoscale molecular composition, assembly, and function are barely known. Here, using expansion microscopy and electron microscopy, we reveal the molecular architecture of the CC and demonstrate that microtubules are linked together by a CC inner scaffold containing POC5, CENTRIN, and FAM161A. Dissecting CC inner scaffold assembly during photoreceptor development in mouse revealed that it acts as a structural zipper, progressively bridging microtubule doublets and straightening the CC. Furthermore, we show that Fam161a disruption in mouse leads to specific CC inner scaffold loss and triggers microtubule doublet spreading, prior to outer segment collapse and photoreceptor degeneration, suggesting a molecular mechanism for a subtype of retinitis pigmentosa.Inherited retinal degeneration due to loss of photoreceptor cells is a leading cause of human blindness. The Wenchuan earthquake (Richter scale 8) on 12 May 2008 in southwestern China caused widespread ecosystem damage in the Longmenshan area. It is important to evaluate natural vegetation recovery processes and provide basic information on ecological aspects of the recovering environment after the earthquake. To circumvent the weather limits of remote sensing in the Wenchuan earthquake-hit areas, and to meet the need for regional observation analyses, three Landsat TM images pre- and post-earthquake in Mao County were used for analysis. Post-earthquake normalized difference vegetation index (NDVI) values were compared to pre-earthquake values with an NDVI-based index differencing method to determine the extent to which the vegetation was damaged in relation to the pre-earthquake pattern, and the rate of recovery was evaluated. The spatial characteristics of vegetation loss and natural recovery patterns were analyzed in relation to elevation, slope and aspect. The results indicated that severely damaged sites occurred mainly in river valleys, within a range of 1,500?C2,500?m elevation and on slopes of 25?C55°. The distance from rivers, rather than the distance from active faults, controls the damage patterns. After 1?year of natural regeneration, 36?% of the destroyed areas showed a decrease in NDVI value, 28.8?% showed very little change, 19.1?% showed an increase, and 16.1?% also increased with a recovery rate greater than 100?%. Moreover, there is a good correlation between recovery rate and both slope and elevation, but recovery patterns in the damaged area are complicated. MicroRNAs (miRNAs) are endogenously expressed RNAs consisting of 20–24 nucleotides. These molecules are thought to repress
protein translation by binding to target mRNAs. However, biological functions have not been assigned to most of the 175 porcine
miRNAs registered in miRBase (release 15.0). In an effort to uncover miR-103 important in pigs, we examined the integrative
tissue expression profile and gene ontology (GO) term enrichment of predicted target genes to determine the global biological
functions of miR-103. Our results demonstrated that miR-103 is involved in various biological processes including brain development,
lipid metabolism, adipocyte differentiation, hematopoiesis, and immunity. Moreover, we also experimentally verified effects
of miR-103 in porcine preadipocytes. miR-103 levels increased in differentiating adipocytes, and inhibition of miR-103 effectively
inhibited preadipocyte differentiation. In addition, mRNA levels of the putative miR-103 target RAI14 were higher in miR-103 inhibitor-treated adipocytes. These results demonstrate that miR-103 is involved in porcine preadipocyte
differentiation and may act through the putative target gene RAI14. In a word, our data provide new insights into the global biological role of miR-103. 相似文献

Peptide sequencing plays a fundamental role in proteomics. Tandem mass spectrometry, being sensitive and efficient, is one of the most commonly used techniques in peptide sequencing. Many computational models and algorithms have been developed for peptide sequencing using tandem mass spectrometry. In this paper, we investigate general issues in de novo sequencing, and present results that can be used to improve current de novo sequencing algorithms. We propose a general preprocessing scheme that performs binning, pseudo-peak introduction, and noise removal, and present theoretical and experimental analyses on each of the components. Then, we study the antisymmetry problem and current assumptions related to it, and propose a more realistic way to handle the antisymmetry problem based on analysis of some datasets. We integrate our findings on preprocessing and the antisymmetry problem with some current models for peptide sequencing. Experimental results show that our findings help to improve accuracies for de novo sequencing. 相似文献

The identification of human cancer-related microRNAs (miRNAs) is important for cancer biology research. Although several identification methods have achieved remarkable success, they have overlooked the functional information associated with miRNAs. We present a computational framework that can be used to prioritize human cancer miRNAs by measuring the association between cancer and miRNAs based on the functional consistency score (FCS) of the miRNA target genes and the cancer-related genes. This approach proved successful in identifying the validated cancer miRNAs for 11 common human cancers with area under ROC curve (AUC) ranging from 71.15% to 96.36%. The FCS method had a significant advantage over miRNA differential expression analysis when identifying cancer-related miRNAs with a fine regulatory mechanism, such as miR-27a in colorectal cancer. Furthermore, a case study examining thyroid cancer showed that the FCS method can uncover novel cancer-related miRNAs such as miR-27a/b, which were showed significantly upregulated in thyroid cancer samples by qRT-PCR analysis. Our method can be used on a web-based server, CMP (cancer miRNA prioritization) and is freely accessible at http://bioinfo.hrbmu.edu.cn/CMP. This time- and cost-effective computational framework can be a valuable complement to experimental studies and can assist with future studies of miRNA involvement in the pathogenesis of cancers. 相似文献

工业园区层面的生态效率评价是生态效率研究领域有待深入探索的课题。TOPSIS方法则可能是适用于生态效率评价的一种方法,它以空间统计学为基础,借助于多目标决策问题的理想解和负理想解来进行排序,能够客观地比较和评价不同样本点综合指标的优劣。本文根据生态效率和循环经济理论,结合《综合类生态工业园区标准(试行)》,建立工业园区循环经济评价指标体系,运用TOPSIS方法对苏州高新区、苏州工业园区生态工业园和无锡新区生态工业示范园区的生态效率进行综合评价,结果表明:三者的生态效率综合排名由高到低依次为:无锡新区生态工业示范园区> 苏州工业园区生态工业园> 苏州高新区,且3园区在经济发展、资源能源利用率、物质循环利用、环境污染控制等子系统有所优劣。并针对各园区的各自存在的不足提出了相应的改进和发展建议。 相似文献

透明质酸(HA)是一种在医药及化妆品领域具有广泛应用的天然粘多糖。兽疫链球菌(Streptococcuszooepidemicus)是工业上生产透明质酸的菌种之一。透明颤菌血红蛋白(VHb)具有增强细胞摄氧的作用。对生产透明质酸的兽疫链球菌进行了基因改造:将兽疫链球菌HA的合成基因hasABC以及合成透明颤菌血红蛋白的vgb基因(Vitreoscillahemoglobingene,vgb)分别或同时插入阳性菌表达质粒pEU308中,通过电转化导入兽疫链球菌中。通过一氧化碳(CO)差光谱检测到了VHb的表达。在摇瓶实验中,同时带有hasABC和vgb基因的重组菌比野生菌的透明质酸产量提高了30%。而在发酵罐中,带有这2个基因的重组菌的透明质酸产量达到了6.9g/L,高于重组菌5.5g/L的产量。实验结果表明,vgb基因的存在促进了细胞的生长,hasABC操纵子的过表达增强了透明质酸的合成。首次将VHb导入兽疫链球菌中,获得了表达,并证明其对菌体生长及透明质酸合成有促进作用。通过研究,VHb将可以在阳性菌中获得更广泛的应用。 相似文献
