We previouslyshowed that increased macrophage andPGE2 production with age is due toenhanced cyclooxygenase (COX) activity and COX-2 expression. This studydetermined the effect of vitamin E supplementation on macrophagePGE2 synthesis in young and old mice and its underlying mechanism. Mice were fed 30 or 500 parts permillion vitamin E for 30 days. Lipopolysaccharide (LPS)-stimulated macrophages from old mice produced significantly morePGE2 than those from young mice.Vitamin E supplementation reversed the increasedPGE2 production in old mice buthad no effect on macrophage PGE2production in young mice. In both LPS-stimulated and unstimulated macrophages, COX activity was significantly higher in old than in youngmice at all intervals. Vitamin E supplementation completely reversedthe increased COX activity in old mice to levels comparable to those ofyoung mice but had no effect on macrophage COX activity of young miceor on COX-1 and COX-2 protein or COX-2 mRNA expression in young or oldmice. Thus vitamin E reverses the age-associated increase in macrophagePGE2 production and COX activity.Vitamin E exerts its effect posttranslationally, by inhibiting COXactivity. 相似文献

In plant cells, autophagy is required for efficient recycling of cytoplasmic macromolecules in vacuoles. It was previously shown that autophagy-deficient mutants also exhibited hypersensitivity to various abiotic stresses, such as salt, osmotic changes, heat, drought, and oxidative damage. However, it has not been clearly determined whether autophagy is induced or inhibited by these environmental stressors. Using the GFP-ATG8 (green fluorescent protein fused to AUTOPHAGY-RELATED PROTEIN 8) processing assay and confocal microscopy, we assessed autophagic flux of Arabidopsis seedlings exposed to salt stress. Treatment with 150 mM NaCl resulted in an increase in the processing of GFP-ATG8. Notably, the effects of concanamycin A, an inhibitor of vacuolar proton pumps, on GFP-ATG8 processing indicated that the apparent increase in GFP-ATG8 processing by salt-induced stress was due to inefficient vacuolar degradation of the GFP moiety processed from GFP-ATG8. Salt and osmotic stresses did not increase the abundance of autophagic vesicles in the root cells. Although NaCl, KCl, and mannitol did not greatly inhibit the vacuolar trafficking of GFP-ATG8, LiCl partially inhibited autophagy. These data indicated that NaCl stress neither increases nor substantially inhibits autophagic flux. Our work illustrates the importance of autophagic flux analysis to assess the effect of abiotic stresses on plant autophagy. 相似文献

The 3D-QSAR study of quinolinediones which showed potent inhibitory effect on the acetylcholine induced vasorelaxation of rat aorta with the endothelium was conducted by CoMSIA. We have prepared and evaluated the antibacterial activities of a series of substituted methylenepiperidinyl and methylenepyrrolidinyl oxazolidinones against several gram-positive strains including the resistant strains of Staphyloccus and Enterococcus, such as MRSA, CRSA, MSSA and VRE. A series of isoxazolyl tetrahydropyridinyl oxazolidinones with various substituents at the 3-position of the isoxazole ring have been synthesized and their in vitro antibacterial activities (MIC) were evaluated against several Gram-positive strains including the resistant strains of Staphyloccus and Enterococcus, such as MRSA and VRE. 城市绿地生态系统是城市景观的重要组成部分,具有重要的生态与文化价值。鸟类是城市绿地生态系统的指示类群,研究人类活动对鸟类资源的影响能够为城市生态景观的建设与维护提供重要理论依据。本研究于2021年10月至2022年5月,采用样线法对武汉市不同干扰强度的城市绿地内林鸟群落进行调查。共记录到鸟类11目34科100种,其中国家二级重点保护鸟类9种,在物种组成上以雀形目为主(76种,占调查到总鸟种数的76%),在居留型上以留鸟为主(42种,42%),在区系上主要属于东洋界(45种,45%)。繁殖季鸟类物种数高于非繁殖季,主要是由于夏候鸟和旅鸟的增加导致。在不同干扰强度中,重度干扰斑块的平均鸟类个体数最多,轻度干扰斑块的鸟类物种丰富度、多样性指数和均匀度指数最高,而中度干扰斑块的鸟类个体数、物种丰富度、多样性指数和均匀度指数均为最低,且繁殖季和非繁殖季鸟类群落结构在不同人为干扰强度中的格局未发生变化。综上所述,武汉市城市绿地的鸟类多样性较为丰富,随着干扰强度增加,鸟类多样性出现非线性差异,揭示了人为干扰与自然干扰对生态影响的差异;在面积狭小、破碎化严重的斑块中,人为干扰可能有利于城市绿地鸟类多样性的维持。 相似文献

Pharmacophore modelling, docking and virtual screening have become important tool in drug discovery process. Serotonin 2C (5-HT2C) receptor ligands have got major attention for their therapeutic uses as antidepressant and anorectic agents. Two step pharmacophore and docking based virtual screening was done using 5-HT2C agonists. Two common feature pharmacophore directed virtual hits had submicromolar activity. Refined pharmacophore with excluded volumes was constructed and combined with homology model based docking. Best hit from this virtual screening showed IC50 of 20.1 nM. Similarity search of this hit compound resulted more active ligand with 7.8 nM activity. 相似文献

In vitro metabolic stability of dopamine D(3)/D(4) receptor antagonists and identification of their metabolites by high-performance liquid chromatography (HPLC) coupled with ion-trap mass spectrometry (ITMS) were assessed in rat liver microsomes. The compounds were divided into three cassette groups for rapid quantitative analysis of multiple drugs and simultaneous detection of their metabolites. The samples from incubation with rat liver microsomes were pooled into designed cassette groups and analyzed by HPLC/electrospray ITMS in full-scan mode. The metabolic stability of the drugs was determined by comparing their signals after incubation for 0 and for 30min. The metabolic stability of the examined dopamine receptor antagonists was in the range of 9.9-84.4%. In addition, the present cassette analysis allowed the simultaneous detection of metabolites formed during the same incubation without having to reanalyze the samples. The metabolites were first characterized by nominal mass measurement of the corresponding protonated molecules. Subsequent multistage tandem mass spectrometry on the ion-trap instrument allowed characterization of the structure of the detected metabolites. N,O-dealkylation and ring hydroxylation reactions were identified as major metabolic reactions in piperazinylalkylisoxazole derivatives. These results suggested that the present approach is useful for the rapid evaluation of metabolic stability and structural characterization of metabolites within a short period in new drug discovery. 相似文献
