Sixteen Lactobacillus plantarum strains, isolated from fermented table olives, were studied for the presence and expression of genes involved in the production of bacteriocins, pheromones and other peptides. The presence of 13 genes that belong to pln locus was monitored, while for the study of gene expression, producer strains were cultured in growth medium with variant salinity (0, 4, 6, and 8 % NaCl) and pH (3.5, 4.0, 4.5, and 6.4). The effect of producer strain on the growth of indicator microorganisms was evaluated using a well diffusion assay. In parallel, Real-Time PCR was employed to monitor the genetic expression of plnE/F and plnJ/K genes for strains that revealed the highest antimicrobial activity. The well diffusion assay showed that the growth of Lactobacillus pentosus was inhibited by six L. plantarum strains when cultured on control medium (0 % NaCl, pH 6.4). Moreover, when the same growth medium was supplemented with 4 and 6 % NaCl, the growth of L. pentosus was inhibited by three and two L. plantarum strains, respectively. Growth of L. pentosus was favoured when L. plantarum strains were cultured on a growth medium with lowered pH (3.5, 4.0, and 4.5). No inhibition of pathogens was observed, but in a few cases, inhibition of Aureobasidium pullulans was detected. The Real-Time PCR assay revealed that the expression of genes was dependent on producer strains and growth phase, whereas inhibition of indicator strains was enhanced in earlier stages of the growth curve in the presence of NaCl, although similar counts were obtained.  相似文献

On treating the blue-green alga Anacystis nidulans with dimethylsuberimidate up to 70% of the free NH2 of the photosynthetic membrane is amidinated, and presumably inter- and intramolecular cross-links are established in the membrane proteins. Amidination destroys the ability of A. nidulans to photoreduce HCO3? but leaves the photochemical activities of Photosystems II and I nearly intact. With added electron acceptors, photosynthetic O2 evolution can be demonstrated both with permeable cells (permeaplasts) prepared by digestion of the cell wall of dimethylsuberimidate-reacted A. nidulans with lysozyme, as well as with heavy membrane particles (36 000 × g) prepared from dimethylsuberimidate-reacted cells.Permeaplasts prepared from dimethylsuberimidate-reacted cells resist damage in hypoosmotic medium, whereas those prepared from unreacted cells are induced to release C-phycocyanin. On the other hand, the former are inactivated more easily by heat stress than the latter. On this basis, it is concluded that cross-linking with dimethylsuberimidate confers functional instability to photosynthetic membranes.  相似文献

The woodland history of the northern mountainous parts of Greece is considered in the context of pollen profiles from bogs in Rhodopes, Lailias-Vrontou, Paiko, Voras and Pieria. In the time period covered by these diagrams (only those from Voras and Lailias extend back to the 7th millennium B.P.) the overall trends in the woodland history at the particular sites are comparable but there are also substantial differences that are attributed to the effects of human influence which varied in time, intensify and extent. The earlier phase, which corresponds to the Atlantic period of Firbas, is characterized by mixed deciduous woodlands with Quercus or Tilia the main components. This is followed by the phase (Subboreal) in which coniferous woodlands, which consisted of Abies and Pinus, dominated. In this time, Fagus gradually expanded. In the final phase (Subatlantic), Abies becomes more or less extinct and Fagus has a dominant role. These developments broadly correspond with those recorded in pollen profiles from Central Europe.  相似文献

Cancer is a clinical situation caused by uncontrolled cell division and is responsible for a large number of deaths worldwide. Colchicine is a classical antimitotic, tubulin-binding agent (TBA) which is being explored for its antitumor activities, although its tubulin-binding ability leads to some toxicity toward normal cells proliferation. Colchicine derivatives are considered as potent antitumor compounds with less toxicity compared to colchicine. Derivatives with substituted functional groups at A-ring (methoxy), B-ring (acetamide) or C-ring (methoxy) have been synthesized via chemical and microbial routes and show modified bioactivities and altered tropolonic functionality. Earlier reports, in combination with our group’s research findings, suggest that microbial biotransformation is an efficient choice for the production of bioactive colchicine derivatives. This route has gained significant interest in the mass production of regio-specific, cost-effective, safe and eco-friendly derivatives. The present review paper critically analyzes and discusses the development and application of colchicine derivatives as a potent antitumor molecule and their production through a microbial transformation process. The information provided in this review might assist in the stimulation of new ideas regarding the development of alternative therapeutic agent(s) for cancer treatment.  相似文献

The present study investigated the mite fauna and the relative abundance of mites present in foliage and litter of two adjacent and similar alfalfa fields, differing only in the number of cuttings, in Kopais Valley (Central Greece) through 2008–2010. We also examined the relationship between assemblage patterns of Mesostigmata, Oribatida and Prostigmata by comparing their population fluctuation, population density, species richness and diversity. Spatial distribution of common dominant and influent mite species was also estimated. Our results showed that both fields supported a very rich and similar mite fauna with eight new species records for alfalfa of Greece, although these species have been previously reported from other habitats in Greece. The pattern of population fluctuation in foliage was similar in both fields, unlike the fluctuation in litter. Population density significantly differed between fields, being higher in the less harvested field, except Prostigmata. Species richness in litter was higher in the less harvested field, whereas it was higher in the foliage of the more harvested field, apart from that of prostigmatic mites in litter, which was higher in the more harvested field, and that of oribatid mites in foliage, which was higher in the less harvested field. The diversity of mites was higher in the more harvested field, with the exception of prostigmatic mites. The spatial distribution of mites in foliage and litter was aggregated in both fields. Our results indicate that despite the considerable similarity of the study fields, the different harvesting frequency might have disturbed differently the mite communities hosted in foliage and litter.  相似文献

Summary A new solid phase synthesis of the growth-modulating tripeptide Gly-His-Lys is described. 2-Chlorotrityl chloride resin and 9-fluorenylmethoxycarbonyl-(Fmoc), 4-methyltrityl-(Mtt) protecting groups were used. The synthetic tripeptide was tested for its activity against bacteria, yeast and fungi. The in vitro effect of the tripeptide on DNA, RNA and protein synthesis was studied as well.  相似文献

A prototype system for passive intracranial monitoring using microwave radiometry is proposed. It comprises an ellipsoidal conductive wall cavity to achieve beamforming and focusing, in conjunction with sensitive multiband receivers for detection. The system has already shown the capability to provide temperature and/or conductivity variations in phantoms and biological tissue. In this article, a variant of the initially constructed modality is theoretically and experimentally investigated. Specifically, dielectric matching materials are used in an effort to improve the system's focusing attributes. The theoretical study investigates the effect of dielectric matching materials on the system's detection depth, whereas measurements with phantoms focus on the investigation of the system's detection level and spatial resolution. The combined results suggest that the dielectric matching layers lead to the improvement of the system's detection depth and temperature detection level. Also, the system's spatial resolution is explored at various experimental setups. Theoretical and experimental results conclude that with the appropriate combination of operation frequencies and dielectric layers, it is possible to monitor areas of interest inside human head models with a variety of detection depths and spatial resolutions.
