The design features of gustatory systems are considered in relation to the need for insect herbivores (or indeed any animal) to regulate their nutrient intake. The gustatory system (defined to include both peripheral and central components) should ideally be designed to reflect: (a) the average nutritional requirements of the insect during its various developmental stages; (b) the predictably unpredictable variation in nutrient supply-demand which occurs about that average; and (c) the distribution and composition of foods available in the environment. A mathematical model is presented which makes detailed predictions regarding behavioural and electrophysiological responses to foods or simple solutions varying in nutrient content.  相似文献

DNA, isolated from age-synchronous senescent populations of Caenorhabditis elegans has been quantitatively and qualitatively analyzed for the presence of 5-methylcytosine. High performance liquid chromatography on two wild-type and several mutant strains of C. elegans failed to detect any 5-methylcytosine. The restriction endonuclease isoschizomers, HpaII and MspI, were used to digest genomic DNA after CsCl purification and failed to detect any 5' cytosine methylation at any age. We conclude that C. elegans does not contain detectable (0.01 mole percent) levels of 5-methylcytosine.  相似文献

Non-muscle-invasive bladder cancer (NMIBC) is one of the most common malignant tumors in the urological system with a high risk of recurrence, and effective non-invasive biomarkers for NMIBC relapse are still needed. The human urinary proteome can reflect the status of the microenvironment of the urinary system and is an ideal source for clinical diagnosis of urinary system diseases. Our previous work used proteomics to identify 1643 high-confidence urinary proteins in the urine from a healthy population. Here, we used bioinformatics to construct a cancer-associated protein-protein interaction (PPI) network comprising 16 high-abundance urinary proteins based on the urinary proteome database. As a result, platelet-derived growth factor receptor beta (PDGFRB) was selected for further validation as a candidate biomarker for NMIBC diagnosis and prognosis. Although the levels of urinary PDGFRB showed no significant difference between patients pre- and post-surgery (n = 185, P>0.05), over 3 years of follow-up, urinary PDGFRB was shown to be significantly higher in relapsed patients (n = 68) than in relapse-free patients (n = 117, P<0.001). The levels of urinary PDGFRB were significantly correlated with the risk of 3-year recurrence of NMIBC, and these levels improved the accuracy of a NMIBC recurrence risk prediction model that included age, tumor size, and tumor number (area under the curve, 0.862; 95% CI, 0.809 to 0.914) compared to PDGFR alone. Therefore, we surmise that urinary PDGFRB could serve as a non-invasive biomarker for predicting NMIBC recurrence.  相似文献

We are describing a system for the introduction, selection, and expression of eucaryotic genes in higher eucaryotic cells. The carrier consisted of the herpes simplex virus 1 (HSV-1) tk gene covalently linked to an HSV-1 alpha promoter directed away from the tk gene. In this study we fused to the alpha promoter the 5' transcribed noncoding sequences and the coding sequences of the chicken oviduct ovalbumin gene. Cells converted to the TK+ phenotype with this chimeric fragment produced an ovalbumin precursor which was processed and secreted into the extracellular fluid. The ovalbumin gene utilized the HSV-1 alpha promoter and was regulated as a viral gene inasmuch as inversion of the genomic DNA relative to the alpha promoter resulted in no ovalbumin synthesis, and production of ovalbumin was enhanced after superinfection with HSV-1. Synthesis of ovalbumin was not detected when cDNA was linked to the HSV-1 alpha promoter. The carrier system described in this study is suitable for introduction, selection, and expression of eucaryotic genes whose natural promoter is either weak or requires the presence of regulatory elements which may be absent from undifferentiated cells in culture.  相似文献

Haematological and serum biochemistry values were determined for 13 adult, free-living American mink, Neovison vison, in southern England live trapped as part of a longer term research project. Serum samples were tested for the presence of antibodies against Toxoplasma gondii, Aleutian disease virus (ADV) and canine distemper virus (CDV). Animals were examined to assess ectoparasite burden; faecal samples were examined for the presence of gut parasites and bacteria (identified via culture). Post-mortem examinations were carried out on four individuals shot during on-going control operations. Haematological and serum biochemistry values for most individuals were similar to published values for captive mink. Neutrophil/lymphocyte ratios were high in two animals (possibly due to trap-associated stress). Three individuals had high levels of creatinine, urea and the liver enzymes, alanine transaminase, aspartate transaminase, alkaline phosphatise and gamma-glutamyl transpeptidase. Six of 12 mink tested positive for antibodies to T. gondii and 8 of 12 tested positive for antibodies to ADV; none tested positive for antibodies to CDV. No significant bacteria or parasites were detected in faecal samples. Post-mortem examinations in three cases showed no significant lesions but the fourth animal had Skrjabingylus nasicola nematodes in the nasal passages, lung lesions suggestive of adiaspiromycosis, cholangiohepatitis possibly indicative of Pseudamphistomum truncatum infection and tubulointerstitial nephritis associated with renal calculi.  相似文献

In Drosophila, neural precursors are formed in a spaced pattern separated by intervening epidermal cells. Segregation of neural and epidermal lineages relies on cellular interactions. Failure of this cell communication, as in the mutants Notch (N), Delta, and shaggy, results in most or all of the cells becoming neural. Cells mutant for N and shaggy, but not Delta, autonomously adopt the neural fate when adjacent to wild-type cells in mosaics. Furthermore, wild-type cells adopt the epidermal fate if adjacent cells express a lower level of N activity than themselves, but produce neural precursors if adjacent cells express a higher level of N activity. This shows that there is competition between the cells and that the N protein is required for the mechanism whereby the cells choose between alternative fates. It also suggests that N acts as a receptor for an inhibitory signal emanating from the neural precursors.  相似文献

The Russian barley cultivar Nevsky lacks 3 hordein and accumulates most of its hordein in the lumen of the endoplasmic reticulum and only a minor portion in the vacuole. In wild type barley and all other temperate cereals, storage proteins are deposited in the vacuole. F1 crosses revealed that the Nevsky phenotype is recessive; but the extent of hordein accumulation in the endoplasmic reticulum in F2 endosperm lacking 3 hordein was very much less than in the Nevsky parent. In order to study the Nevsky endosperm phenotype we have measured the levels of seven proteins and two mRNAs involved in protein folding in the ER lumen or ER to Golgi transport during endosperm development. The protein levels were unaltered in Nevsky as compared to the wild-type variety Bomi. When the levels of these seven proteins were correlated with the rate of hordein accumulation, four of these (HSP70, PDI, Sar1p and Sec18p) were consistently up-regulated with hordein synthesis. Accumulation of hordein in the endoplasmic reticulum appears to be determined by the absence of 3 hordein, or the product of a gene closely linked to it, plus one or more other recessive genes.  相似文献
