There is a significant body of evidence demonstrating that radiation therapy (XRT) enhances the effect of immune therapy. However, the precise mechanisms by which XRT potentiates the immunotherapy of cancer remain elusive. Here, we report that XRT potentiates the effect of immune therapy via induction of autophagy and resultant trafficking of mannose-6-phopsphate receptor (MPR) to the cell surface. Irradiation of different tumor cells caused substantial up-regulation of MPR on the cell surface in vitro and in vivo. Down-regulation of MPR in tumor cells with shRNA completely abrogated the combined effect of XRT and immunotherapy (CTLA4 antibody) in B16F10-bearing mice without changes in the tumor-specific responses of T cells. Radiation-induced MPR up-regulation was the result of redistribution of the receptor to the cell surface. This effect was caused by autophagy with redirection of MPR to autophagosomes in a clathrin-dependent manner. In autophagosomes, MPR lost its natural ligands, which resulted in subsequent trafficking of empty receptor(s) back to the surface. Together, our data demonstrated a novel mechanism by which XRT can enhance the effect of immunotherapy and the molecular mechanism of this process.  相似文献

Studies of heat shock response show a correlation with local climate, although this is more often across altitudinal than latitudinal gradients. In the present study, differences in constitutive but not inducible components of heat shock response are detected among populations of the Glanville fritillary butterfly Melitaea cinxia L. that exist at the species' latitudinal range limits (Finland and Spain). The study demonstrates that macroclimatic differences between these sites should cause greater exposure of the Spanish population to higher temperatures. Thermal stress treatments are used to estimate differences in the expression of four genes potentially relevant for tolerating these temperatures. For the analysis, three heat‐shock proteins and glyceraldehyde‐3‐phosphate dehydrogenase (G3PDH), a glycolysis enzyme that also modulates cell growth based on metabolic state, are chosen. Two constitutive differences are found between the sites. First, insects from Spain have higher levels of Hsp 21.4 than those from Finland regardless of thermal stress treatment; this protein is not inducible. Second, insects from Finland have higher levels of G3PDH. The two remaining Hsps, Hsp20.4 and Hsp90, show dramatic up‐regulation at higher temperatures, although there are no significant differences between insects from the different populations in either constitutive levels or inducibility. In nature, differences between the study populations likely occur in the expression of all four genes that were studied, although these differences would be directly climate‐induced in Hsp20.4 and Hsp90 and constitutive in Hsp21.4 and G3PDH. Inducibility may mitigate the need for constitutive variation in traits that adapt insects to local climate.  相似文献

Successful pellet production has been reported in literature with cross-linked poly(vinylpyrrolidone), Polyplasdone® XL-10 and INF-10. In the present study, a quality by experimental design approach was used to assess several formulation and process parameter effects on the characteristics of Polyplasdone® XL-10 pellets, including pellet size, shape, yield, usable yield, friability, and number of fines. The hypothesis is that design of experiments and appropriate data analysis allow optimization of the Polyplasdone product. High drug loading was achieved using caffeine, a moderately soluble drug to allow in vitro release studies. A five-factor, two-level, half-fractional factorial design (Resolution V) with center point batches allowed mathematical modeling of the influence of the factors and their two-factor interactions on five of the responses. The five factors were Polyplasdone® level in the powder blend, volume of water in the wet massing step, wet mixing time, spheronizer speed, and spheronization time. Each factor and/or its two-factor interaction with another factor influenced pellet characteristics. The behavior of these materials under various processing conditions and component levels during extrusion-spheronization have been assessed, discussed, and explained based on the results. Numerical optimization with a desirability of 0.974 was possible because curvature and lack of fit were not significant with any of the model equations. The values predicted by the optimization described well the observed responses. The hypothesis was thus supported.  相似文献
