Rapid visco analysis (RVA) was performed to study the pasting properties of mixtures of wheat flour and tuber starches, i.e., potato starch (PS), sweet potato starch (SPS), yam starch (YS), and cassava starch (CS), at 10–50% starch in the mixtures. Lower phosphorus and higher amylose contents were observed in CS, followed by YS, SPS, and PS. The peak, breakdown, final, and setback viscosities of the control wheat flour were lower than those of the control PS, SPS, YS, and CS. The peak viscosity of wheat–PS mixtures was higher than those of the wheat–SPS, wheat–YS, and wheat–CS because of the higher phosphorus and lower amylose content of PS, which resulted in higher swelling of PS than that of SPS, YS, and CS. The breakdown viscosities increased as the starch content of the PS, SPS, and CS in the mixtures increased to up to the 50%, and the values tended to decrease in the wheat–YS mixture. The setback viscosities of wheat–SPS, wheat–YS, and wheat–CS increased significantly as the starch content increased from 10% to 50%, and that of wheat–PS dropped dramatically at 50%. The findings in this work provide evidence that tuber starches could be used as a partial substitute for wheat flour in some wheat-based products.  相似文献

Bioactive endophytic streptomycetes from the Malay Peninsula   总被引:1,自引:0,他引:1  
Three novel endophytic streptomycetes have been isolated and characterized from plants with ethnobotanical uses on the Malay Peninsula including: Thottea grandiflora (family -Aristolochiaceae), Polyalthia spp. (family -Annonaceae), and Mapania sp. (family -Cyperaceae). Each isolate, as studied by scanning electron microscopy, has small hyphae, and produces typical barrel-shaped spores arising by hyphal fragmentation. Interestingly, although none has any detectable antibacterial killing properties, each has demonstrable killing activity against one or more pathogenic fungi including organisms such as Phytophthora erythroseptica, Pythium ultimum, Sclerotinia sclerotiorum, Mycosphaerella fijiensis and Rhizoctonia solani. Molecular biological studies on the rRNA gene sequence of each isolate revealed that it is distinct from all other genetic accessions of streptomyectes in GenBank, and each bears some genetic similarity to other streptomycetes. The bioactivity of each microbe was extractable in various organic solvents.  相似文献

From the world's tiny flying bugs to gigantic dobsonflies, inflight locomotion of a flying creature requires complex biomechanical strategies to cope with air turbulence. These unpredictable changes in ambient airflow strength and direction may destabilize body posture and orientation. To record this behaviour in further detail, we scientifically examined how houseflies (Musca domestica) respond to air turbulence. We then, three-dimensionally reconstructed body and wings motion of continuously perturbated houseflies using high-speed videography under laboratory condition. The findings confirmed that houseflies, in general, do not initiate flight when average ambient air speed exceeds ~0.63 ms?1 at approximately ~2% of relative turbulent intensity. This finding contrasts with flies which immediately take-off after being released. During mild turbulent conditions, flies performed take-off but with severe and active modulation of body postures. In addition, the body roll angle fluctuates more severely (18.5-fold increase) compared to yaw (7-fold of increment) and pitch (6.4-fold of increment) during turbulence, highlighting that body roll stability is highly sensitive. This research extends our current knowledge on flies' behaviours during turbulence and how insects achieve their superior flight performance.  相似文献

A successful in vitro Agrobacterium-mediated transformation protocol was developed for Mimulus aurantiacus, a model species for ecological and evolutionary genetics and a promising ornamental plant. Three binary vectors were tested, each containing the hptII selectable marker gene and one of the reporter genes: gusA, EGFP or ZsGreen, all of them under CaMV 35S promoter. Genetic transformation was achieved through 4 days of co-cultivation of leaf, petiole and hypocotyl explants with Agrobacterium tumefaciens strain LBA 4404. Explants produced transformed callus tissue on solid modified Murashige and Skoog medium supplemented with 1 mg L?1 6-benzylaminopurine, 0.5 mg L?1 1-naphthaleneacetic acid, 30 g L?1 sucrose and 20 or 50 mg L?1 hygromycin B. All three reporter genes were expressed in callus tissue but the intensity of expression gradually decreased during further plant development. The new reporter gene ZsGreen proved suitable for plant transformation experiments since very intense and bright fluorescence was detected. Out of 1,760 co-cultured explants, 110 plants were regenerated and all of them were found to be PCR positive for the selection and/or reporter genes. Chemiluminescent Southern blot analysis revealed that 91 % of the regenerated plants (100 T0 plants) contained T-DNA integrated in their genome. Transformation efficiency varied from 1.4 to 23.3 % for hypocotyl and petiole explants, respectively. Integration of some backbone sequences in plant genomes was confirmed in 75.3 % of T0 plants. Using this protocol, stable transformants expressing selectable marker gene hptII and one of the reporter genes (gusA, ZsGreen or EGFP) were obtained in 4–5 months.  相似文献

Breast cancer is the most common cause of deaths and the most frequently diagnosed cancer among women worldwide. This study aimed to determine the prevalence of breast cancer screening, specifically on clinical breast examination, and the predictors of its uptake among women in Malaysia. A cross-sectional study was carried out in five selected districts whereby women aged between 20 to 64 years old, from a total of 1000 households were interviewed. A total of 1192 women responded to the survey of which 53.3% reported had ever done clinical breast examination. Significant associations with clinical breast examination were noted for income and distance from the hospital. These factors should be considered in developing interventions aimed at promoting clinical breast examination. In particular, healthcare providers should be proactive in raising awareness about clinical breast examination among women in Malaysia.  相似文献

The 72 known members of the flavivirus genus include lethal human pathogens such as Yellow Fever, West Nile, and Dengue viruses. There is at present no known chemotherapy for any flavivirus and no effective vaccines for most. A common genomic organization and molecular mechanisms of replication in hosts are shared by flaviviruses with a viral serine protease playing a pivotal role in processing the viral polyprotein into component polypeptides, an obligatory step in viral replication. Using the structure of the dengue serine protease complexed with a protein inhibitor as a template, we have identified five compounds, which inhibit the enzyme. We also describe parallel inhibitory activity of these compounds against the West Nile virus Protease. A few of the compounds appear to provide a template for design of more potent and specific inhibitors of the dengue and West Nile virus proteases. Sequence similarities among flaviviral proteases suggests that such compounds might also possibly inhibit other flaviviral proteases.  相似文献
