Recent progress in determining the population structure of Campylobacter jejuni, and discerning associations between genotypes and specific niches, has emphasized the shortfall in our understanding of the ecology and epidemiology of this bacterium. We examined the natural structure of the C. jejuni community associated with cattle farmland in the UK by structured spatiotemporal sampling of habitats, including livestock and wild animal faeces, environmental water and soil, over a 10-week period within a 100 km2 area. A total of 172 isolates were characterized using multilocus sequence typing into 65 sequence types (STs). Isolates from cattle faeces were significantly over-represented in the ST-61 complex, whereas isolates from wildlife faeces and water were more likely to belong to the ST-45 complex and a number of unusual STs, many of which were first encountered during this study. Sampling within a narrow spatiotemporal window permitted the application of novel statistical methods exploring the relationship between the genetic relatedness and spatial separation of isolates. This approach showed that isolates from the same sampling squares and squares separated by <1.0 km were genetically more similar than isolates separated by greater distances. Our study demonstrates the potential of multilocus sequence typing combined with spatial modelling in exploring natural transmission pathways for C. jejuni. 相似文献

Background
Prevalence of type 2 diabetes (T2D) is increasing worldwide. T2D prevention by lifestyle intervention is effective. Pragmatic scalable interventions are needed, with evidence to efficiently target and monitor such interventions. We report pooled analyses of data from three European trial cohorts: to analyse T2D incidence, sustained weight loss and utility of risk predictors.Methods
We analysed data on 749 adults with impaired glucose tolerance (278 men and 471 women, mean age 56 years, mean BMI 31 kgm−2) recruited between 1993 and 2003, and randomised to intensive lifestyle intervention (I) or lifestyle advice control (C). The intervention aimed to increase physical activity, modify diet, and promote weight loss≥5%. Using Cox-regression survival analysis, we assessed T2D incidence and the impact on T2D incidence of sustained weight loss, and of baseline cut-point values of FINDRISC score, fasting plasma glucose (FPG), and HbA1c.Results
Mean follow-up duration was 3.1 years. T2D was diagnosed in 139 participants (I = 45/379, C = 94/370). Cumulative T2D incidence was 57% lower in the intervention compared with the control group (HR 0.42 (95% CI 0.29 to 0.60) P<0.001). Participants with ≥5% weight loss at one year had 65% lower T2D incidence (HR 0.35 (95% CI 0.22 to 0.56) P<0.001); maintaining ≥5% weight loss for two and three years further reduced T2D incidence. Recommended cut-points to identify those at high risk for T2D would have identified different proportions of European Diabetes Prevention Study (EDIPS) participants with similar hazard-ratios for intervention effect.Conclusions
Pooled analysis of EDIPS trial data reinforces evidence for T2D prevention by lifestyle intervention. Analysis showed the preventive effect of ≥5% weight loss, especially if maintained long term, which has utility for intervention monitoring. Analysis of proposed cut-points demonstrates difficulties in balancing risk and benefit, to efficiently target interventions and suggests evidence is needed to define clinical policy.Trial registrations

TGR5 is a G protein-coupled receptor that mediates bile acid (BA) effects on energy balance, inflammation, digestion, and sensation. The mechanisms and spatiotemporal control of TGR5 signaling are poorly understood. We investigated TGR5 signaling and trafficking in transfected HEK293 cells and colonocytes (NCM460) that endogenously express TGR5. BAs (deoxycholic acid (DCA), taurolithocholic acid) and the selective agonists oleanolic acid and 3-(2-chlorophenyl)-N-(4-chlorophenyl)-N, 5-dimethylisoxazole-4-carboxamide stimulated cAMP formation but did not induce TGR5 endocytosis or recruitment of β-arrestins, as assessed by confocal microscopy. DCA, taurolithocholic acid, and oleanolic acid did not stimulate TGR5 association with β-arrestin 1/2 or G protein-coupled receptor kinase (GRK) 2/5/6, as determined by bioluminescence resonance energy transfer. 3-(2-chlorophenyl)-N-(4-chlorophenyl)-N, 5-dimethylisoxazole-4-carboxamide stimulated a low level of TGR5 interaction with β-arrestin 2 and GRK2. DCA induced cAMP formation at the plasma membrane and cytosol, as determined using exchange factor directly regulated by cAMP (Epac2)-based reporters, but cAMP signals did not desensitize. AG1478, an inhibitor of epidermal growth factor receptor tyrosine kinase, the metalloprotease inhibitor batimastat, and methyl-β-cyclodextrin and filipin, which block lipid raft formation, prevented DCA stimulation of ERK1/2. Bioluminescence resonance energy transfer analysis revealed TGR5 and EGFR interactions that were blocked by disruption of lipid rafts. DCA stimulated TGR5 redistribution to plasma membrane microdomains, as localized by immunogold electron microscopy. Thus, TGR5 does not interact with β-arrestins, desensitize, or traffic to endosomes. TGR5 signals from plasma membrane rafts that facilitate EGFR interaction and transactivation. An understanding of the spatiotemporal control of TGR5 signaling provides insights into the actions of BAs and therapeutic TGR5 agonists/antagonists. 相似文献

A study was conducted to determine which bud (terminal or lateral) breaks first, and thereby exerts primigenic dominance,
on ‘Granny Smith’ and ‘Golden Delicious’, 1-year-old apple (Malus × domestica Borkh.) shoots grown in two locations in the Western Cape, South Africa, with differing degrees of chilling. Primigenic dominance
of laterals was more common in a warm area than a cool area, and more common in ‘Granny Smith’ than ‘Golden Delicious’. Laterals
rarely broke before the terminal in ‘Golden Delicious’, and so differences in lateral development due to position of first
bud to break were only analyzed in ‘Granny Smith’ shoots from this point on in the study. In ‘Granny Smith’, lateral budbreak
and growth was influenced by the position of the first bud to break on the shoot, but did not differ between locations. On
‘Granny Smith’ shoots with primigenic dominance of the terminal, lateral budbreak and growth was suppressed, in accordance
with the typical ‘delayed foliation’ commonly observed in warm winter climates. However, when at least one lateral broke before

The Great Escarpment is the major mountain system in South Africa, yet very few biological surveys have been conducted outside of the well-known Drakensberg section. This is surprising given the important role that mountains play in local and global biodiversity patterns. In this study, small mammal diversity and community composition were estimated at three high altitude (>1,700?m) sites within the Sneeuberg Mountain Complex (SMC) of the Great Escarpment, South Africa from June 2009 to May 2010. The influences of selected environmental variables on diversity were also tested. Of 423 live-captures, a total of 292 unique individuals of 12 small mammal species (one shrew, one elephant shrew and 10 rodents) were identified during 5,280 trap nights. No single environmental variable could account for the variation observed in diversity measurements but vegetation height appeared to be the most important factor to influence the number of individuals captured. It is hypothesised that the high species richness and diversity of small mammals observed in the SMC compared to other parts of the Great Escarpment is due to the SMC being located in a transition zone of the Grassland and Nama-Karoo biomes. Our results suggest that the SMC could be important in conserving small mammal species from western and eastern assemblages across South Africa. 相似文献

Phytochromes are dimeric photoreceptors that regulate a range of responses in plants and microorganisms through interconversion of red light-absorbing (Pr) and far-red light-absorbing (Pfr) states. Photoconversion between these states is initiated by light-driven isomerization of a bilin cofactor, which triggers protein structural change. The extent of this change, and how light-driven structural changes in the N-terminal photosensory region are transmitted to the C-terminal regulatory domain to initiate the signalling cascade, is unknown. We have used pulsed electron-electron double resonance (PELDOR) spectroscopy to identify multiple structural transitions in a phytochrome from Synechocystis sp. PCC6803 (Cph1) by measuring distances between nitroxide labels introduced into the protein. We show that monomers in the Cph1 dimer are aligned in a parallel ‘head-to-head’ arrangement and that photoconversion between the Pr and Pfr forms involves conformational change in both the N- and C-terminal domains of the protein. Cryo-trapping and kinetic measurements were used to probe the extent and temporal properties of protein motions for individual steps during photoconversion of Cph1. Formation of the primary photoproduct Lumi-R is not affected by changes in solvent viscosity and dielectric constant. Lumi-R formation occurs at cryogenic temperatures, consistent with their being no major structural reorganization of Cph1 during primary photoproduct formation. All remaining steps in the formation of the Pfr state are affected by solvent viscosity and dielectric constant and occur only at elevated temperatures, implying involvement of a series of long-range solvent-coupled conformational changes in Cph1. We show that signalling is achieved through ultrafast photoisomerization where localized structural change in the GAF domain is transmitted and amplified to cause larger-scale and slower conformational change in the PHY and histidine kinase domains. This hierarchy of timescales and extent of structural change orientates the histidine kinase domain to elicit the desired light-activated biological response. 相似文献

Metallothioneins, MT's, are low-molecular-weight, cysteine-rich, polypeptides that complex ‘soft’ metal ions in thiol clusters.
They are structurally diverse. Some MT's are gene products, while others are secondary metabolites. Two of the three classes
of MT have been identified in algae. Eukaryotic algae possess the secondary metabolites referred to as class III MT. There
is no unequivocal evidence that MT genes occur in eukaryotic algae. However, the products of MT genes have been identified
in cyanobacteria. These genes and their metal regulatory elements remain to be isolated and characterized.
MT's have attracted interest from researchers involved in a wide range of disciplines including bioinorganic chemistry, biochemistry,
molecular biology, physiology, toxicology, environmental science and medicine. Although, the precise physiological roles of
these polypeptides remain undefined, a large number of functions have been speculated. These molecules chelate toxic trace
metals, such as Cd, thereby reducing the concentration of cytotoxic, free-metal ions. Furthermore, some MT's are believed
to be involved in zinc and copper homoeostasis. Future studies should reveal whether or not some of the diversity of MT structure

Alzheimer's disease is a major neurodegenerative disease of the brain, the incidence of which increases dramatically in old age. Currently, no drugs are available to halt or slow the progression of this disease, which poses an ever-expanding burden on health services, families and society. The prion protein has become infamous owing to its role as the causative agent of the transmissible spongiform encephalopathies such as Creutzfeldt-Jakob disease in humans. However, our view of the prion protein as an unwanted, harmful entity has been challenged recently. Abstract. 1. The emigration behaviour of the army ant Dorylus ( Anomma ) molestus was studied in the montane forest of Mt Kenya. This species forages by massive swarm raids (mean width 10.3 m ± 4.6 m SD), which are assumed to have a strong negative impact on the densities of prey populations.
2. For non-reproductive colonies the stay duration in a nest is highly variable (median 17, range 3–111 days). This suggests that the frequency of emigrations is not dictated by a brood cycle as an underlying endogenous pattern generator.
3. Colony density is high (mean nearest neighbour's distance 82.1 m ± 29.4 m SD) and mean foraging range is 75.0 m, so encounters with neighbouring colonies occur frequently.
4. The straight line emigration distance is on average 92.7 m (± 29.7 SD). The emigration direction is random with respect to absolute bearing and also relative to the direction of the previous emigration. However, the emigration direction is influenced by the location of the nearest neighbour. Colonies typically emigrate directly away from their nearest neighbour.
5. Local food depletion is likely to be the ultimate cause for emigrations in this species, because emigration distance is larger than foraging range and colonies move away from their nearest neighbour. A small percentage of emigrations may be triggered by pangolin attacks on nests.
