Spontaneously forming small unilamellar vesicles (SULVs) are easy to prepare and show great promise for use in delivering therapeutic payloads. We report of SULVs made up of the ternary phospholipid mixture, dimyristoyl-phosphatidylcholine (DMPC), dihexanoyl-phosphatidylcholine (DHPC) and dimyristoyl-phosphatidylglycerol (DMPG), which have been characterized by small angle neutron scattering (SANS). These low-polydispersity (0.14-0.19) SULVs range in size (i.e., radius) from 110 to 215 Å and are capable of entrapping, and subsequently releasing, hydrophilic molecules (e.g., fluorescent dyes and quenchers) in a controlled fashion over two different temperature ranges. The low-temperature release mechanism involves the SULVs transforming into discoidal micelles, with an onset temperature (To) of ~ 32 °C, while the high-temperature release mechanism is more gradual, presumably the result of defects formed through the continuous dissolution of DHPC into solution. Both of these mechanisms differ from other, previously reported thermosensitive liposomes.  相似文献

Very little is known about the molecular basis of autosomal recessive MR (ARMR) because in developed countries, small family sizes preclude mapping and identification of the relevant gene defects. We therefore chose to investigate genetic causes of ARMR in large consanguineous Iranian families. This study reports on a family with six mentally retarded members. Array-based homozygosity mapping and high-resolution microarray-based comparative genomic hybridization (array CGH) revealed a deletion of approximately 150–200 kb, encompassing the promoter and the first six exons of the MCPH1 gene, one out of four genes that have been previously implicated in ARMR with microcephaly. Reexamination of affected individuals revealed a high proportion of prematurely condensed chromosomes, which is a hallmark of this condition, but in spite of the severity of the mutation, all patients showed only borderline to mild microcephaly. Therefore the phenotypic spectrum of MCPH1 mutations may be wider than previously assumed, with ARMR being the only consistent clinical finding.  相似文献

Using small-angle neutron scattering and dynamic light scattering, we have constructed partial structural phase diagrams of lipid mixtures composed of the phosphatidylcholines dimyristoyl and dihexanoyl doped with calcium ions (Ca2+) and/or the negatively charged lipid, dimyristoyl phosphatidylglycerol (DMPG). For dilute solutions (lipid concentration < or =1 wt %), spontaneously forming unilamellar vesicles (ULVs) were found, and their polydispersity was determined to be approximately 20%. The stability of the Ca2+- or DMPG-doped ULVs was monitored over a period of 4 days and their structural parameters (e.g., average outer radius, ) were found to be insensitive to the lipid concentration (Clp). However, doping the dimyristoyl/dihexanoyl system with both Ca2+ and DMPG resulted in ULVs whose was found to be Clp dependent. The of DMPG-doped ULVs remained unchanged over an extended period of time (at least 4 days), a good indication of their stability.  相似文献

The present study was carried out to evaluate and compare the volumetric densities of subepithelial infiltrated connective tissue (ICT) geographically in bleeding and non-bleeding gingivae with special reference to comparisons of the relationships with their corresponding stereomicroscopic findings. A total of 51 biopsies (25 bleeding and 26 non-bleeding gingival specimens) was obtained from 33 patients. After stereomicroscopic examinations, tissue sections were obtained with H&E stain. On each section, the density of ICT was calculated by volumetric analysis. The results showed that the volume of ICT in bleeding-gingiva was significantly greater than that in non-bleeding gingiva. However, there was no specific relationship between the ICT of gingiva and the visibility of vasculature by stereomicroscopy. These results suggest that increased infiltrated inflammatory volumes of gingiva may play a role in the presenting of bleeding on clinical probing, but appear not to be related to the visibility of vasculature by stereomicroscopy.  相似文献

Multimodal communication plays an important role in pollination biology. Bees have evolved multimodal communication to recruit nestmates to rewarding food sources. Highly social bees can use visual and chemical information to recruit nestmates to rich food sources. However, no studies have determined if this information is redundant or has an additive effect such that multimodal information is more attractive than either modality presented by itself to free-flying bees. We tested the effect of two modalities, forager-deposited odor marks and the visual presence of foragers, on the orientation of stingless bee (Scaptotrigona mexicana) recruits. Our results show that odor marks alone were significantly more attractive than multimodal information, and that multimodal information was significantly more attractive than visual forager presence alone. Given the high olfactory sensitivity and limited visual acuity of insects, odor marks likely attracted recruits over a greater distance than the visual presence of nestmates. Thus, multimodal information in S. mexicana is redundant, not additive, in terms of orientation to food sources.  相似文献

Because of their biomechanical significance, cross-sectional geometric properties of long bone diaphyses (areas, second moments of area) have been increasingly used in a number of form/function studies, e.g., to reconstruct body mass or locomotor mode in fossil primates or to elucidate allometric scaling relationships among extant taxa. In the present study, we test whether these biomechanical section properties can be adequately estimated using biplanar radiographs, as compared to calculations of the same properties from computer digitization of cross-sectional images. We are particularly interested in smaller animals, since the limb bone cortices of these animals may not be resolvable using other alternative noninvasive techniques (computed tomography). The test sample includes limb bones of small (25–5,000 g) relatively generalized quadrupedal mammals—mice, six species of squirrels, and Macaca fascicularis. Results indicate that biplanar radiographs are reasonable substitutes for digitized cross-sectional images for deriving areas and second moments of area of midshaft femora and humeri of mammals in this size range. Potential application to a variety of questions relating to mechanical loading patterns in such animals is diverse. © 1993 Wiley-Liss, Inc.  相似文献
