通过温室土壤培养试验,研究了不同添加量玉米和水稻秸秆对淹水土壤pH、CO2及交换态铵的影响.结果表明:在接近中性的土壤中加入秸秆可使土壤pH值降低,4 g·kg-1玉米和水稻秸秆处理的土壤pH值均与对照差异显著(P<0.05),而1 g·kg-1玉米和水稻秸秆处理与对照差异不显著(P>0.05).土壤溶液中的CO2含量随秸秆添加量的增加而增大,1 g·kg-1玉米和水稻秸秆处理的土壤溶液CO2含量最大值分别为35.9%和31.9%(v/v),与对照(25.8%)差异达显著水平(P<0.05),但两者间差异不显著(P>0.05);4 g·kg-1玉米和水稻秸秆处理的土壤溶液CO2含量最大值分别为54.2%和41.8%(v/v),与对照差异极显著(P<0.01),两者间差异也达显著水平(P<0.05).在不施氮肥的情况下,添加秸秆可降低土壤铵态氮浓度,且铵态氮浓度随秸秆添加量的增加而降低,不同添加量处理间差异显著(P<0.05);在施入氮肥的情况下,1 g·kg-1玉米和水稻秸秆处理提高了土壤铵态氮浓度,而4 g·kg-1玉米和水稻秸秆处理降低了土壤铵态氮浓度.无论是否施入氮肥,玉米和水稻秸秆处理的土壤铵态氮浓度差异不显著(P>0.05).  相似文献

The cuticular wax composition of leaves has been analysed in three western European populations (Corsica, central Pyrenees, northern Alps) of Juniperus communis var. saxatilis Pall. (=  J. nana Willd., nom illeg.) and in one population of J. communis L. var. depressa Pursh. from North America (Sierra Nevada). Gas chromatography shows the presence of 13 alkanes in all samples ranging from C23 to C35 with important intraspecific polymorphism in alkane content. The dominant alkanes range from C33 to C35. Alkanes C21 and C22 were found only in Corsica and Sierra Nevada populations. Canonical discriminant analysis separated the J. communis L. var. depressa Pursh. of the population of Sierra Nevada from other populations of J. communis var. saxatilis Pall. on the basis of their higher C31 content and the constant presence of C21 and C22 alkanes. J. communis var. saxatilis Pall. populations from the Pyrenees are close to northern Alps populations characterized by high concentrations of C33, C34 and C35 alkanes. Tests assessing the fear of humans by measuring avoidance or approach reactions of the test animals towards humans can be useful tools in welfare assessment schemes. In this study, we wanted to compare tests assessing the reactions of laying hens towards humans in the home environment to a test performed in a novel environment. As well we investigated inter-test correlations in the home environment. We performed several tests to assess the hens’ approach and avoidance reactions towards an unfamiliar human inside the home environment of laying hens in 14 caged flocks and 10 free-range flocks. We measured the reactions of hens to a stationary person and a moving person approaching individual hens in both systems, as well as reactions to a stationary person trying to touch individual hens in non-cage systems and reactions of caged hens to a person passing by. Additionally, a standardised arena test was performed outside the home environment to compare the birds’ avoidance reactions towards an approaching human in a novel environment. In caged hens no significant correlations of tests performed inside the home environment with the arena test could be found, but there were moderate to high correlations (rs > 0.6) with one parameter of the arena test in non-caged hens. The different tests applied inside the home environment correlated significantly and moderately to highly with each other, supporting the validity of these tests to measure fear of humans. The caged laying hens in our study reacted differently in the novel environment from the non-caged birds, indicating that the birds’ reactions towards humans in an unfamiliar environment depend on the housing system. In summary, a comparison of the level of fear of humans between flocks in different housing systems by using an arena test outside the home environment seems difficult and was not possible in the present study.  相似文献

Understanding how DNA polymerases control fidelity requires elucidation of the mechanisms of matched and mismatched dNTP incorporations. Little is known about the latter because mismatched complexes do not crystallize readily. In this report, we employed small-angle X-ray scattering (SAXS) and structural modeling to probe the conformations of different intermediate states of mammalian DNA polymerase β (Pol β) in its wild-type and an error-prone variant, I260Q. Our structural results indicate that the mismatched ternary complex lies in-between the open and the closed forms, but more closely resembles the open form for WT and the closed form for I260Q. On the basis of molecular modeling, this over-stabilization of mismatched ternary complex of I260Q is likely caused by formation of a hydrogen bonding network between the side chains of Gln260, Tyr296, Glu295 and Arg258, freeing up Asp192 to coordinate MgdNTP. These results argue against recent reports suggesting that mismatched dNTP incorporations follow a conformational path distinctly different from that of matched dNTP incorporation, or that its conformational closing is a major contributor to fidelity.  相似文献

The nucleoside triphosphatase (NTPase)/helicase associated with nonstructural protein 3 of West Nile (WN) virus was purified from cell culture medium harvested from virus-infected Vero cells. The purification procedure included sequential chromatography on Superdex-200 and Reactive Red 120 columns, followed by a concentration step on an Ultrogel hydroxyapatite column. The nature of the purified protein was confirmed by immunoblot analysis using a WN virus-positive antiserum, determination of its NH(2) terminus by microsequencing, and a binding assay with 5'-[(14)C]fluorosulfonylbenzoyladenosine. Under optimized reaction conditions the enzyme catalyzed the hydrolysis of ATP and the unwinding of the DNA duplex with k(cat) values of 133 and 5.5 x 10(-3) s(-1), respectively. Characterization of the NTPase activity of the WN virus enzyme revealed that optimum conditions with respect to the Mg(2+) requirement and the monovalent salt or polynucleotide response differed from those of other flavivirus NTPases. Initial kinetic studies demonstrated that the inhibition (or activation) of ATPase activity by ribavirin-5'-triphosphate is not directly related to changes in the helicase activity of the enzyme. Further analysis using guanine and O(6)-benzoylguanine derivatives revealed that the ATPase activity of WN virus NTPase/helicase may be modulated, i.e., increased or reduced, with no effect on the helicase activity of the enzyme. On the other hand the helicase activity could be modulated without changing the ATPase activity. Our observations show that the number of ATP hydrolysis events per unwinding cycle is not a constant value.  相似文献

Heterologous prime-boost immunization protocols using different gene expression systems have proven to be successful tools in protecting against various diseases in experimental animal models. The main reason for using this approach is to exploit the ability of expression cassettes to prime or boost the immune system in different ways during vaccination procedures. The purpose of the project was to study the ability of recombinant vaccinia virus (VV) and bacterial plasmid, both carrying the NS1 gene from tick-borne encephalitis (TBE) virus under the control of different promoters, to protect mice against lethal challenge using a heterologous prime-boost vaccination protocol.  相似文献

Phosphoinositides play a central role in the control of several cellular events including actin cytoskeleton organization. Here we show that, upon infection of epithelial cells with the Gram-negative pathogen Shigella flexneri, the virulence factor IpgD is translocated directly into eukaryotic cells and acts as a potent inositol 4-phosphatase that specifically dephosphorylates phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P(2)] into phosphatidylinositol 5-monophosphate [PtdIns(5)P] that then accumulates. Transfection experiments indicate that the transformation of PtdIns(4,5)P(2) into PtdIns(5)P by IpgD is responsible for dramatic morphological changes of the host cell, leading to a decrease in membrane tether force associated with membrane blebbing and actin filament remodelling. These data provide the molecular basis for a new mechanism employed by a pathogenic bacterium to promote membrane ruffling at the entry site.  相似文献

People can eat a food without having a strong preference for it, and people can prefer a food without eating it. Given this seeming disconnect between attitude and behavior, which type of measure or segment can best be used to profile or identify loyal consumer segments of a food, such as soy? This research compares a usage‐based method (heavy‐light‐nonusers) with a new attitude‐based method (seeker‐neutral‐avoider), and finds that the attitude‐based method differentiates purchase‐related intentions better than the usage‐based method. Implications for profiling consumer taste patterns and consumer segments are provided.  相似文献

Ustilago maydis, the causal agent of corn smut disease, displays dimorphic growth in which it alternates between a unicellular, nonpathogenic yeast-like form and a dikaryotic, pathogenic filamentous form. Previously, a constitutively filamentous haploid mutant was obtained. Complementation of this mutant led to the isolation of the gene encoding adenylate cyclase, uac1. Secondary mutagenesis of a uac1 disruption strain allowed the isolation of a large number of suppressor mutants, termed ubc, for Ustilago bypass of cyclase, lacking the filamentous phenotype. Analysis of one of these suppressor mutants previously led to the identification of the ubc1 gene, encoding the regulatory subunit of cAMP-dependent protein kinase. In this report we describe the isolation of cosmids containing three new ubc genes, termed ubc2, ubc3, and ubc4. We also describe the morphology of the ubc2, ubc3, and ubc4 mutants in a uac1- background as well as in a background with a functional uac1 gene. In addition, we describe several mutant strains not complemented with any of the genes currently in hand and that are thus presumed to possess mutations in additional ubc genes.
