A number of recent studies show that activation of CR3 on dendritic cells (DCs) suppresses TLR-induced TNF-alpha and IL-12 production and inhibits effective Ag presentation. Although the proposed physiologic role for these phenomena is immune suppression due to recognition of iC3b opsonized apoptotic cells by CR3, all of the aforementioned investigations used artificial means of activating CR3. We investigated whether iC3b opsonized apoptotic cells could induce the same changes reported with artificial ligands such as mAbs or iC3b-opsonized RBC. We explored the kinetics of iC3b opsonization in two models of murine cell apoptosis, gamma-irradiated thymocytes and cytokine deprivation of the IL-3 dependent cell line BaF3. Using a relatively homogenous population of early apoptotic cells (IL-3 deprived BaF3 cells), we show that iC3b opsonized apoptotic cells engage CR3, but this interaction is dispensable in mediating the anti-inflammatory effects of apoptotic cells. TLR-induced TNF-alpha and IL-12 production by bone marrow-derived DCs occurs heterogeneously, with apoptotic cells inhibiting only certain populations depending on the TLR agonist. In contrast, although apoptotic cells induced homogeneous IL-10 production by DCs, IL-10 was not necessary for the inhibition of TNF-alpha and IL-12. Furthermore, because the ability of iC3b opsonization to enhance phagocytosis of apoptotic cells has been controversial, we report that iC3b opsonization does not significantly affect apoptotic cell ingestion by DCs. We conclude that the apoptotic cell receptor system on DCs is sufficiently redundant such that the absence of CR3 engagement does not significantly affect the normal anti-inflammatory processing of apoptotic cells.  相似文献

This desk study deals with the mechanisms and parameters affecting particles separation from wastewater in mainly upflow anaerobic reactors. Despite the fact that the functioning of upflow anaerobic sludge blanket (UASB) systems depends on both physical parameters and biological processes, the physical parameters have been barely reported in the literature. The reason is that the underlying mechanisms are very complex and depend on various interrelated parameters. In addition, the lack of a serious attempt to gather the entire physical theme into one picture has resulted in just a superficial understanding of this field of science. Better understanding of the interaction and role of these parameters is essential for the development of anaerobic treatment technologies. In this study, the various parameters that might affect the solid liquid separation process by filtration through the sludge bed of a UASB have been elaborated. These parameters have been classified into (1) reactor operational conditions (temperature, organic loading rate, hydraulic retention time and upflow velocity), (2) influent characteristics (influent concentration, influent particle size and influent particle charge) and (3) sludge bed characteristics (particle size distribution, extracellular polymeric substances, and charge). The overall output of this study includes (1) a literature review, (2) structuring of this field of science, and (3) highlighting fields where research is needed.  相似文献

In the mixture of lipids and proteins which comprise pulmonary surfactant, the dominant protein by mass is surfactant protein A (SP-A), a hydrophilic glycoprotein. SP-A forms octadecamers that interact with phospholipid bilayer surfaces in the presence of calcium. Deuterium NMR was used to characterize the perturbation by SP-A, in the presence of 5 mM Ca2+, of dipalmitoyl phosphatidylcholine (DPPC) properties in DPPC/egg-PG (7:3) bilayers. Effects of SP-A were uniformly distributed over the observed DPPC population. SP-A reduced DPPC chain orientational order significantly in the gel phase but only slightly in the liquid-crystalline phase. Quadrupole echo decay times for DPPC chain deuterons were sensitive to SP-A in the liquid-crystalline mixture but not in the gel phase. SP-A reduced quadrupole splittings of DPPC choline β-deuterons but had little effect on choline α-deuteron splittings. The observed effects of SP-A on DPPC/egg-PG bilayer properties differ from those of the hydrophobic surfactant proteins SP-B and SP-C. This is consistent with the expectation that SP-A interacts primarily at bilayer surfaces.  相似文献

A qualitative sludge characterisation technique called "sludge filterability technique" has been developed. This technique enables the determination of the sludge potential for the physical removal of solids, weighing the effect of different process parameters on solids removal and identifying the mechanisms of solids removal in an upflow anaerobic sludge bed system. In this paper guidelines for conducting the test are given and a "standardised" set-up is presented. The experimental set-up and protocol are simple and the results can be obtained in a period as short as a few hours. A sludge sample is added to an upflow reactor incubated at 4 degrees C, to limit gas production, washed with an anaerobically pre-treated and suspended solids free wastewater to remove solids washed out from the sludge, and then fed with a model substrate, prepared from fish meal with a standard procedure. Several experimental runs were conducted to validate and optimise the technique. The results showed that the technique is reliable, workable and reproducible.  相似文献
