Cutting edge: Th2 response induction by dendritic cells: a role for CD40.   总被引:8,自引:0,他引:8  
We investigated the influence of dendritic cell (DC) CD40 expression on Th2 and Th1 development by in vivo transfer of Ag-pulsed bone marrow-derived DC generated from wild-type (WT) or CD40(-/-) mice. Contrary to expectation, CD40(-/-) DC primed with Ag that inherently induce a Th2 response (soluble egg Ag from Schistosoma mansoni) failed to induce a Th2 response or any compensatory Th1 response, whereas CD40(-/-)DC primed with Ag that inherently induce a Th1 response (Propionibacterium acnes) generated a competent Th1 response. Thus, DC expression of CD40 is a prerequisite for initiation of Th2, but not Th1, responses by these Ag. Consistent with this, CD154(-/-) mice, unlike WT mice, failed to mount a Th2 response when directly injected with schistosome eggs but mounted a normal Th1 response after challenge with P. acnes. CD40-CD154 interaction can therefore play a major role in Th2 response induction.  相似文献

The disease barrier hypothesis is one long-standing explanation of the temporal discrepancies between the initial colonization of North and South America. The model postulates an epidemiological barrier that prohibited or slowed the initial migration into South America during the Late Pleistocene. Using data from ethnographically documented hunter–gatherers, the theoretical foundations of the hypothesis are explored. In addition, likely demographic effects to colonizing populations are postulated and compared to disease-response mechanisms in foraging societies. Based on identified disease conditions deemed necessary to maintain a prohibitive barrier, it is suggested that disease transmission rates in the initial colonizing populations of the New World were likely extremely limited and insufficient to support the disease barrier concept.  相似文献

Affecting almost twenty percent of thepopulation in industrialized countries,allergic diseases such as asthma and seasonalrhinitis result from the development ofimmediate hypersensitivity to otherwiseinnocuous components of the environment. Pollen, the male gametophyte of flowering plantspecies, is one of the most predominant sourcesof environmental allergens, and a significantcause of allergic diseases. This reviewdiscusses the nature of pollen proteins asallergens, their effect on the human immunesystem and their mode of environmentaltransmission, including effects of theinteraction between pollen aeroallergens andair pollution. The influence of cross-reactivepollen allergens to the incidence of foodsensitivities is also discussed. Floweringplant species that possess allergenic pollen,identified with allergens cloned from thesespecies, are also discussed.  相似文献

Microscopic analyses of tundra soils from northern central Siberia, Taimyr Peninsula (74.5°N, 98.5°E) were performed in order to investigate spatial variation of fungal and bacterial biomass. Biomass figures of fungi and bacteria (µg C g-1 dry wt.) were measured from 11 permafrost soil pits. Fungal biovolume of up to 3.5 mm3 g-1 dry wt. (median 0.19 mm3 g-1 dry wt.) and a maximum hyphal length of 393 m g-1 dry wt. (median 21 m g-1 dry wt.) were determined. Fungal biomass was found up to 455 µg C g-1 dry wt. (median 24 µg C g-1 dry wt.). The amounts generally decreased with depth but increased within organic horizons. Little fungal biomass was found in the unvegetated soils or deep horizons above the permafrost table. Bacterial counts ranged from 0.16 to 7.38*109 g-1 dry wt. and bacterial biomass ranged from 0.68 to 20.38 µg C g-1 dry wt. (median 6.19 µg C g-1 dry wt.) because of small cell volume (median 0.04 µm3). Microbial biomass was generally dominated by fungi as shown by the ratio of fungal to bacterial biomass, which was between 0 and 174.1 (median 4.5). Plant cover and soil organic matter content were found to be the important keys in understanding microbial ecology in arctic tundra soils.  相似文献

Metachromatic leukodystrophy (MLD) is a recessive autosomal disease which is biochemically characterized by an accumulation of sulfatides (sulfogalactosylceramides) mainly in oligodendrocytes and macrophages/microglia. The deficient enzyme is a lysosomal hydrolase, cerebroside sulfate sulfatase (arylsulfatase A). MLD is both a dysmyelinating and a demyelinating disease. The main clinical forms are infantile or juvenile, but some forms appear at adulthood. This disease involves also neuronal cells as sulfatides are also present in neurons in which the defect in degradation occurs also. We have studied 12 cases of adult MLD and clearly distinguished two clinical forms. One of them was characterized by mainly central nervous system motor signs (pyramidal, cerebellar, and seldom dystonia) and a peripheral neuropathy. The other form always started by behavioural abnormalities with modifications of mood, peculiar social reactions; a progressive mental deterioration occurred also. The diagnosis of schizophrenia was often mentioned. Most of these patients remained for many years without any neurological symptoms, and the diagnosis was only made when neurological signs appeared, or when Magnetic Resonance Imaging (MRI) was performed. MRI showed a diffuse demyelination, bilateral and often symmetrical, which could be temporarily limited to the periventricular areas. The diagnosis of adult MLD was biochemical, evidencing the low activity of arylsulfatase A (ASA) and sulfatide accumulation. To determine the respective participation of neurons and glial cells in the physiopathology of both the motor forms and the psycho-cognitive forms, our first approach was to search for mutations differing according to the clinical status. Motor forms involved the major adult ASA mutation P426L in a homozygote form in contrast to psycho-cognitive forms which involved as a compound heterozygote a specific I179S mutation.  相似文献

Colloidal silver has been known to have unique antimicrobial activity that may be useful in the construction of antibacterial materials (self-cleaning materials) to aid in the fight against bacteria-related infections. In this study, silver-coated TiO2 (Ag/TiO2) particles prepared through the photo-reduction of Ag+ were investigated as an antibacterial agent against Escherichia coli and Staphylococcus aureus. The deposition of Ag onto the surface was confirmed with SEM and EDS analysis of the post-reaction particles. It was also determined that the initial concentration of Ag+ in solution played a significant role in the effective size of the post-irradiation particles. The antibacterial effectiveness of the Ag/TiO2 was evaluated through the determination of the minimum inhibitory concentration (MIC) of AgTiO2 for each species of bacteria. The MIC values for the Ag/TiO2, on both E. coli and S. aureus, were much lower than the MIC values for Ag metal, and quite comparable to the MIC values for AgNO3. A disc diffusion/antibiotic sensitivity test was also performed using the Ag/TiO2 particles and the results compared with the results obtained for Ag metal, AgNO3 and common antibacterial agents; tetracycline, chloramphenicol, erythromycin, and neomycin. The zone of inhibition diameters for the Ag/TiO2 particles were found to be comparable with those of the other antimicrobial agents.  相似文献
