Reverse genetic screen for loss‐of‐function mutations uncovers a frameshifting deletion in the melanophilin gene accountable for a distinctive coat color in Belgian Blue cattle

In the course of a reverse genetic screen in the Belgian Blue cattle breed, we uncovered a 10‐bp deletion (c.87_96del) in the first coding exon of the melanophilin gene (MLPH), which introduces a premature stop codon (p.Glu32Aspfs*1) in the same exon, truncating 94% of the protein. Recessive damaging mutations in the MLPH gene are well known to cause skin, hair, coat or plumage color dilution phenotypes in numerous species, including human, mice, dog, cat, mink, rabbit, chicken and quail. Large‐scale array genotyping undertaken to identify p.Glu32Aspfs*1 homozygous mutant animals revealed a mutation frequency of 5% in the breed and allowed for the identification of 10 homozygous mutants. As expression of a colored coat requires at least one wild‐type allele at the co‐dominant Roan locus encoded by the KIT ligand gene (KITLG), homozygous mutants for p.Ala227Asp corresponding with the missense mutation were excluded. The six remaining colored calves displayed a distinctive dilution phenotype as anticipated. This new coat color was named ‘cool gray’. It is the first damaging mutation in the MLPH gene described in cattle and extends the already long list of species with diluted color due to recessive mutations in MLPH and broadens the color palette of gray in this breed. 相似文献

The transport rate of chalk, clay particles and algal cells (Scenedesmus sp.) through the digestive tract of Streptocephalus
torvicornis and Branchipus schaefferi is described under experimental conditions. Differences in transport rate as well as
in the degree of digestion at a fixed particle density of algae were found. In S. torvicornis, the transport rate is higher
and the digestion of algae lower than in B. schaefferi. These differences might reflect trophic differences related to niche
partitioning in these sympatric species.

The planktonic genus Planktothrix, as other cyanobacteria, shows signals of both homologous and nonhomologous recombination. However, the frequency of recombination and its effect on Planktothrix population structuring is unknown. We isolated 290 Planktothrix strains from seven neighboring lakes in the subalpine Italian region and analyzed these using multilocus sequence typing. Four of six loci analyzed were polymorphic, resulting in 20 distinct multilocus genotypes. Association indices among alleles at different loci were suggestive of an “epidemic population structure,” resulting from an explosive (and temporary) dominance of one genotype against a panmictic background. ClonalFrame analyses supported this view by detecting: (i) three major clades affected by three distinct recombination events, (ii) a recombination rate about equal to the mutation rate, and (iii) the fact that recombination had an impact on introducing molecular diversity more than double the mutation rate. Furthermore, analysis of molecular variance over an annual cycle in three of seven lakes revealed that both local clonal expansion and recombination processes affected among‐lake diversity. Our observations suggest that recombination affects microevolution of Planktothrix and that an epidemic structure can emerge in populations of this genus. 相似文献

Chemical reduction of mitosenes under aerobic conditions in DMSO showed characteristic ESR signals of the mitosene derived semiquinone free radicals. However, these signals diminished strongly upon addition of water to the reaction mixture, indicating a short lifetime of the mitosene semiquinone free radicals under aqueous conditions. In addition, enzymatic one-electron reduction of these mitosenes with either xanthine oxidase or purified NADPH cytochrome P450 reductase under anaerobic conditions showed no signals of the mitosene semiquinone free radicals. Subsequent cyclic voltammetry measurements demonstrated facilitation of the further one-electron reduction of the mitosene semiquinone free radicals in the presence of water in comparison with non-aqueous conditions. The present results strongly suggest that in the presence of water relatively stable hydroquinones are formed upon reduction of mitosenes. Consequently, the steady state concentrations of mitosene semiquinone free radicals will be lowered substantially in aqueous environment. Thus under physiological conditions, two-electron reduction and formation of the mitosene hydroquinone might be important in processes leading to DNA alkylation by these mitosenes. 相似文献

Small, benthic, cryptic fishes represent a species-rich guild on marine substrates. Most of them are micropredators that feed on crustaceans that are often smaller than 1 mm. Typical examples are seahorses and pipefishes (Syngnathidae), most gobies (Gobiidae), dragonets (Callionymidae) and triplefins (Tripterygiidae). Previous work on the yellow black-headed triplefin Tripterygion delaisi demonstrated that it actively redirects downwelling sunlight sideways using its iris and can use this to locally illuminate objects of interest. We call this form of active sensing using light “diurnal active photolocation”. Visual modelling predicted that light redirection can be sufficient to induce a perceivable change in luminance in the eyes of one of its prey species, a cryptic gammarid crustacean (Cheirocratus sp.), over distances of 1–2 cm. Empirical validation, however, was not possible because measurements of predation distances have not been quantified for free-ranging, small, benthic fishes before. Here, we present interaction distances measured from videos of T. delaisi approaching and striking at prey in the field. Out of 160 recordings, we were able to quantify 78 prey approaching distances and 100 striking distances. Approaching distances ranged from 2.1 to 4.1 cm (interquartile range, IQR) and involved one to five approaching steps before the actual strike occurred. The distance over which the final strike took place varied from 0.7 to 1.6 cm (IQR). Both approaching and striking distances increased with fish body size. We conclude that most approaching sequences started too far away to be explained by prey detection through light redirection. Striking distances, however, fell well with the distances predicted by the model. We conclude that if diurnal active photolocation plays a role in prey detection, it is during the final decision whether to strike or not. 相似文献

Invasions by introduced ant species can be ecologically destructive and affect a wide range of taxa, particularly native ants. Invasive ant species often numerically dominate ant communities and outperform native ant species in effective resource acquisition. Here, we describe interactions between the invasive ant Anoplolepis gracilipes (Smith) and resident ant species in disturbed habitats in NE Borneo. We measured interference competition abilities of A. gracilipes by performing arena bioassays between two A. gracilipes colonies and seven local ant species, and measured its effective resource competition at baits within supercolonies and at supercolony boundaries. Furthermore, we compared ant species diversity and composition at baits among (A) core areas of A. gracilipes supercolonies, (B) supercolony boundaries and (C) outside supercolonies. Anoplolepis gracilipes was behaviorally dominant over most ant species except Oecophylla smaragdina. Within supercolonies, A. gracilipes discovered all food baits first, and monopolized the vast majority throughout the course of the experiment. At supercolony boundaries, A. gracilipes discovered baits later than resident ant species, but subsequently monopolized half of the baits. Furthermore, the activity and diversity of the ant community within A. gracilipes supercolonies was lower than at its boundaries and outside supercolonies, and the ant communities differed significantly between infested and noninfested areas. Our study supports the hypothesis that successful establishment of A. gracilipes in anthropogenically disturbed habitats may negatively affect resident ant communities through high levels of direct interspecific aggression and almost complete monopolization of resources within high‐density supercolonies. 相似文献

This paper is a preliminary report of the ability of IKONOS multispectral satellite imagery with a very high spatial resolution of 1 metre to distinguish two mangrove species in Sri Lanka belonging to the same genus (Rhizophora apiculata and R. mucronata). Not only is this an advancement for the monitoring of forests, it is even more important considering their patchy nature in Sri Lankan mangroves (in contrast to classically zoned forests). Apart from congeneric distinction, intro‐gressive species (Acrostichum aureum) can also be detected from IKONOS imagery, which is important in the early warning for cryptic ecological changes that may affect mangrove species composition (both floral and faunal) and functioning. The results tabulate the usage of various image composites, transformations and classifications, and indicate the danger of too much detail in remote sensing, and the need to apply an optimum resolution. We also highlight that the highest resolutions (as in pansharpened multispectral composites) remain invaluable for visual ecological investigations, which are not at all outdated by new digital satellite images of (sub)metre spatial resolution and their possibility for computer‐aided analysis. 相似文献
