Virulence of cloned variants of Autographa californica nuclear polyhedrosis virus.

The relative virulence of five different genotypic variants of Autographa californica nuclear polyhedrosis virus was tested by determining the 50% lethal dose of occluded virus for larvae of Trichoplusia ni. The 50% lethal dose values of uncloned virus and the five cloned genotypic variants ranged between 10 and 21 polyhedra per larva, and no statistically significant differences were observed. Cloning has therefore neither enhanced nor decreased the virulence of this potential microbial pesticide.     

Scanning electron microscopy of the disruption of tobacco hornworm, Manduca sexta, midgut by Bacillus thuringiensis endotoxin

The ingestion of Bacillus thuringiensis crystal endotoxin by Manduca sexta causes the destruction of both goblet and columnar cells of the midgut. One hour after ingestion, the microvilli show pathological effects. Nearly complete destruction of the goblet and columnar cells has taken place after 4 hr exposure to the toxin.     

California encephalitis virus activity in mosquitoes and horses in southern Ontario, 1975.

A study was undertaken in 1975 to determine California encephalitis virus activity in southern Ontario. Three thousand and sixty-one mosquitoes, primarily Aedes species, were divided into 104 pools and inoculated into suckling mice. Isolates of snowshoe hare virus were obtained from one pool each of Aedes fitchii and A. triseriatus mosquitoes collected in the Guelph area. Serological testing of horse sera revealed extensive virus activity in southern Ontario and indicated that horses may serve as excellent monitors for California encephalitis virus.     

Growth changes in endocranial capacity in the Cercopithecoidea and Hominoidea

On the basis of examination of 2588 skulls representing 15 genera (56 species) from the Cercopithecoidea, together with 973 skulls representing six genera (10 species) of the Hominoidea, grouped according to stages of dental development, the pattern of postnatal growth change in endocranial volume has emerged as constant throughout Old World Primates. The brain attains 80% of its adult size by the time the deciduous dentition is fully established; 90% by the time the first permanent molars erupt; 95% during the period of eruption of the premolars and second molars; and some 98 % by the time the last permanent teeth are erupting. This constancy in timing of postnatal endocranial growth contrasts with a progressive variation in parameters associated with brain weight-body weight relationship.     

Comparison of infrared coagulation and rubber band ligation for first and second degree haemorrhoids: a randomised prospective clinical trial.

One hundred and thirty seven previously untreated out-patients with first and second degree haemorrhoids were allocated at random to treatment by infrared coagulation (n=66) or rubber band ligation (n=71). Complete follow up was obtained in 122 patients (60 who had undergone infrared coagulation (group 1), and 62 rubber band ligation (group 2)) at periods from three months to one year after completion of treatment. Infrared coagulation produced a satisfactory outcome in 51 patients (85%): 34 were rendered asymptomatic and 17 improved. Rubber band ligation produced a satisfactory outcome in 57 patients (92%): 33 were rendered asymptomatic and 24 improved. Both methods were equally effective in first and second degree haemorrhoids. The incidence of side effects, particularly discomfort, during and after treatment was significantly higher in those treated by rubber band ligation (p less than 0.001). This appeared to be an appreciable deterrent to future patient compliance. The number of patients losing more than 24 hours from work was higher after rubber band ligation than after infrared coagulation. The number of treatments necessary to cure symptoms did not differ significantly between the two methods. Infrared coagulation was significantly faster than rubber band ligation (p less than 0.001). Infrared coagulation is a simple, fast, and effective outpatient method for the treatment of first and second degree haemorrhoids with fewer troublesome side effects and higher patient acceptability than rubber band ligation.     

Scanning electron microscopy of cloned astrocytic lines derived from ethylnitrosourea-induced rat gliomas

Six cloned astrocytoma cell lines derived from four ethylnitrosourea-induced F-344 rat gliomas were viewed by scanning electron microscopy in vitro, and two were examined in vivo after transplantation to the intracerebral site. All clones consisted of stellate cells that were reasonably homogeneous within individual glioma lines. Cell membrane features common to all tumor lines included microvilli, blebs, ruffles, and miniridges, mainly confined to perikarya, and filopodia emanating chiefly from cell processes. One cell line demonstrated a profuse, and another cell line a moderate, degree of microvillous development and cell surface roughening, which in one tumor correlated with rapid in vitro cell doubling time. Both cell lines maintained these topographical appearances when transplanted into brain. These results extend the SEM observations of astrocytomas, particularly in cloned ethylnitrosourea-induced tumors in rats. The confirm that distinct variations in cell membrane topography do occur among tumors of this type, probably irrespective of their origin in humans or rats, and irrespective of their mode of genesis as spontaneous, chemically-induced, or virally-induced tumors.     

The inverse autotransporters of Yersinia ruckeri,YrInv and YrIlm,contribute to biofilm formation and virulence

Yersinia ruckeri causes enteric redmouth disease (ERM) that mainly affects salmonid fishes and leads to significant economic losses in the aquaculture industry. An increasing number of outbreaks and the lack of effective vaccines against some serotypes necessitates novel measures to control ERM. Importantly, Y. ruckeri survives in the environment for long periods, presumably by forming biofilms. How the pathogen forms biofilms and which molecular factors are involved in this process, remains unclear. Yersinia ruckeri produces two surface-exposed adhesins, belonging to the inverse autotransporters (IATs), called Y. ruckeri invasin (YrInv) and Y. ruckeri invasin-like molecule (YrIlm). Here, we investigated whether YrInv and YrIlm play a role in biofilm formation and virulence. Functional assays revealed that YrInv and YrIlm promote biofilm formation on different abiotic substrates. Confocal microscopy revealed that they are involved in microcolony interaction and formation, respectively. The effect of both IATs on biofilm formation correlated with the presence of different biopolymers in the biofilm matrix, including extracellular DNA, RNA and proteins. Moreover, YrInv and YrIlm contributed to virulence in the Galleria mellonella infection model. Taken together, we propose that both IATs are possible targets for the development of novel diagnostic and preventative strategies to control ERM.     

Inter-rater reliability of physiotherapists using the Action Research Arm Test in chronic stroke

Objectives:The purpose of this study is to establish whether physiotherapists’ ratings are consistent, when using the Action Research Arm Test (ARAT) to score a chronic stroke patient.Methods:This was part of a large project establishing the reliability in chronic stroke. This study used a correlational design comparing the association between physiotherapist scores of the same patient, to establish the ARAT’s inter-rater reliability. The COSMIN checklist was followed to enhance the methodology of the study.Results:Twenty physiotherapists (8 female and 12 male) aged between 25 and 53 years were selected. There were no participant dropouts or withdrawals. The sample size was normally distributed. The physiotherapists appeared representative of the UK physiotherapy population, with the exception of gender. The distribution of scores showed a normal distribution with standard deviation of score of 1.9. The Kendall’s W test showed 0.711 of agreement between the raters. The scores achieved statistical significance showing consistency between physiotherapists’ scores with chronic stroke. Limitations of the study were the use of a small single center convenience sample that may reduce the generalizability of the findings.Conclusions:The ARAT is consistent when scored by physiotherapists in a chronic stroke population. The inter-rater reliability range was (0.70 to 0.90) which is categorized as good.
