This study aims to investigate the mixing characteristics of a transparent sludge simulant in a mechanically agitated model digester using flow visualisation technique. Video images of the flow patterns were obtained by recording the progress of an acid–base reaction and analysed to determine the active and inactive volumes as a function of time. The doughnut-shaped inactive region formed above and below the impeller in low concentration simulant decreases in size with time and disappears finally. The ‘cavern’ shaped active mixing region formed around the impeller in simulant solutions with higher concentrations increases with increasing agitation time and reaches a steady state equilibrium size, which is a function of specific power input. These results indicate that the active volume is jointly determined by simulant rheology and specific power input. A mathematical correlation is proposed to estimate the active volume as a function of simulant concentration in terms of yield Reynolds number.  相似文献

Bone isremoved or replaced in defined locations by targeting osteoclasts andosteoblasts in response to its local history of mechanical loading.There is increasing evidence that osteocytes modulate this targeting bytheir apoptosis, which is associated with locally increasedbone resorption. To investigate the role of osteocytes in the controlof loading-related modeling or remodeling, we studied the effects onosteocyte viability of short periods of mechanical loading applied tothe ulnae of rats. Loading, which produced peak compressive strains of0.003 or 0.004, was associated with a 78% reduction in theresorption surface at the midshaft. The same loading regimen resultedin a 40% relative reduction in osteocyte apoptosis at the samesite 3 days after loading compared with the contralateral side(P = 0.01). The proportion of osteocytes that wereapoptotic was inversely related to the estimated local strain(P < 0.02). In contrast, a single short period ofloading resulting in strains of 0.008 engendered both tissuemicrodamage and subsequent bone remodeling and was associated with aneightfold increase in the proportion of apoptotic osteocytes(P = 0.02) at 7 days. This increase in osteocyteapoptosis was transient and preceded both intracorticalremodeling and death of half of the osteocytes (P < 0.01). The data suggest that osteocytes might use their U-shapedsurvival response to strain as a mechanism to influence boneremodeling. We hypothesize that this relationship reflects a causalmechanism by which osteocyte apoptosis regulates bone'sstructural architecture.

The Fas ligand (FasL)/Fas receptor (CD95) pathway is an important mediator of apoptosis in the immune system and can also mediate cancer cell death. Soluble FasL (sFasL), shed from the membrane-bound form of the molecule by a putative metalloproteinase (MP), may function to locally regulate the activity of membrane-bound FasL. Using a replication-defective recombinant adenovirus-expressing FasL (RAdFasL), we identified a variable ability of different carcinoma cells to respond to FasL-induced cytotoxicity and to shed sFasL. Blockade of FasL cleavage with an MP inhibitor significantly enhanced RAdFasL-induced apoptosis suggesting that sFasL may antagonize the effect of membrane-bound FasL. In support of this concept, a recombinant adenovirus expressing a noncleavable form of FasL (RAdD4) was found to be a potent inducer of apoptosis even at very low virus doses. Our results highlight the therapeutic potential of noncleavable FasL as an antitumor agent and emphasize the important role of MP via the production of sFasL in regulating the response of the Fas pathway. Moreover, these findings have general implications for the therapeutic exploitation of TNF family ligands and for the possible impact of MP-based therapies on the normal physiology of Fas/TNF pathways.  相似文献

Helicobacter pylori, a major cause of human gastric disease, is a microaerophilic bacterium that contains neither pyruvate nor 2-oxoglutarate dehydrogenase activity. Previous studies (N. J. Hughes, P. A. Chalk, C. L. Clayton, and D. J. Kelly, J. Bacteriol. 177:3953–3959, 1995) have indicated that the major routes for the generation of acetyl coenzyme A (acetyl-CoA) and succinyl-CoA are via pyruvate:flavodoxin oxidoreductase (POR) and 2-oxoglutarate:acceptor oxidoreductase (OOR), respectively. The purified POR is a heterotetrameric protein, with subunits of 48 (PorA), 36 (PorB), 24 (PorC), and 14 (PorD) kDa. In this study OOR has been purified, and it is similarly composed of polypeptides of 43 (OorA), 33 (OorB), 24 (OorC), and 10 (OorD) kDa. Both POR and OOR are oxygen labile and are likely to be major contributors to the microaerophilic phenotype of H. pylori. Unlike POR, OOR was unable to use a previously identified flavodoxin (FldA) as an electron acceptor. Although the purified enzymes were unable to reduce NAD(P), electrons from both pyruvate and 2-oxoglutarate could reduce NADP in cell extracts, consistent with a role for these oxidoreductases in the provision of NADPH as a respiratory electron donor. The H. pylori por, oor, and fldA genes were cloned and sequenced. The deduced por gene products showed significant sequence similarity to archaeal four-subunit 2-oxoacid:acceptor oxidoreductases. However, the amino acid sequences of OorA and -B were more closely related to that of the two-subunit POR of the aerobic halophile Halobacterium halobium. Both porD and oorD encode integral ferredoxin-like subunits. POR and OOR are probably essential enzymes in H. pylori, as insertion inactivation of porB and oorA appeared to be lethal.  相似文献

Thermoinhibition in Tagetes minuta achenes is tightly and rapidly regulated with regard to its imposition and release, with both processes occurring within 2-3h. Germination at high temperatures is almost exclusively regulated by the embryo, while the pericarp appears to play only a minor role. Thermoinhibition in T. minuta could not be alleviated by any single plant growth regulator application, but a combination of treatments that both reduced ABA levels and increased ethylene levels were able to restore germination at supraoptimal temperatures. This suggests a role for both ethylene and ABA in the imposition of thermoinhibition in this species.  相似文献

The increasing focus on plantation forestry as a renewable source of cellulosic biomass has emphasized the need for tools to study the unique biology of woody genera such as Eucalyptus, Populus and Pinus. The domestication of these woody crops is hampered by long generation times, and breeders are now looking to molecular approaches such as marker-assisted breeding and genetic modification to accelerate tree improvement. Much of what is known about genes involved in the growth and development of plants has come from studies of herbaceous models such as Arabidopsis and rice. However, transferring this information to woody plants often proves difficult, especially for genes expressed in woody stems. Here we report the use of induced somatic sector analysis (ISSA) for characterization of promoter expression patterns directly in the stems of Populus and Eucalyptus trees. As a case study, we used previously characterized primary and secondary cell wall-related cellulose synthase (CesA) promoters cloned from Eucalyptus grandis. We show that ISSA can be used to elucidate the phloem and xylem expression patterns of the CesA genes in Eucalyptus and Populus stems and also show that the staining patterns differ in Eucalyptus and Populus stems. These findings show that ISSA is an efficient approach to investigate promoter function in the developmental context of woody plant tissues and raise questions about the suitability of heterologous promoters for genetic manipulation in plant species.  相似文献

The Hauraki Gulf is a large, shallow embayment located north of Auckland City (36°51′S, 174°46′E), New Zealand. Bryde's whales (Balaenoptera edeni) are the most frequently observed balaenopterid in these waters. To assess the use of the Hauraki Gulf for this species, we examined the occurrence and distribution in relation to environmental parameters. Data were collected from a platform of opportunity during 674 daily surveys between March 2003 and February 2006. A total of 760 observations of Bryde's whales were recorded throughout the study period during 371 surveys. The number of Bryde's whales sighted/day was highest in winter, coinciding with the coolest median sea‐surface temperature (14.6°C). Bryde's whales were recorded throughout the Hauraki Gulf in water depths ranging from 12.1–59.8 m (mean = 42.3, SD = 5.1). Cow–calf pairs were most frequently observed during the austral autumn in water depths of 29.9–53.9 m (mean = 40.8, SD = 5.2). Data from this study suggest Bryde's whales in the Hauraki Gulf exhibit a mix of both “inshore” and “offshore” characteristics from the Bryde's whales examined off the coast of South Africa.  相似文献

Previous studies have shown that Eicosapentaenoic Acid (EPA) and Docosahexaenoic Acid (DHA) exhibit triacylglycerol (TAG) lowering effect in vitro and in vivo by down-regulating the Sterol Regulating Element Binding Protein (SREBP-1c) and reducing the expression levels of lipogenic genes. However, there is no evidence on the effect of Docosapentaenoic Acid (DPA) on SREBP-1c expression levels. DPA is a long chain n-3 fatty acid present in our diet through fish, red meat and milk of ruminant animals. Therefore, this study aimed to elucidate the effect of DPA on liver fatty acid synthesis in an in vitro model using rat liver cells. Our results suggested that DPA incubation (50μM) for 48h (like EPA and DHA) caused a significant decrease in the mRNA expression levels of SREBP-1c, 3-Hydroxy-3-Methyl-Glutaryl-Coenzyme A reductase (HMG-CoA reductase), Acetyl Coenzyme A Carboxylase (ACC-1) and Fatty Acid Synthase (FASn) compared with Oleic Acid (OA) and also a decrease in the protein levels of SREBP-1 and ACC-1. A time-course fatty acid analysis showed that DPA and EPA are interconvertable in the cells; however, after 8h of incubation with DPA, the cell phospholipids contained mainly DPA. The gene expression profiling of the lipogenic genes repeated at 8h confirmed that the inhibitory effect of DPA on mRNA expression levels of the lipogenic genes was most likely due to DPA itself and not due to its conversion into EPA.  相似文献

1. Understanding and accurately predicting the spatial patterns of habitat use by organisms is important for ecological research, biodiversity conservation and ecosystem management. However, this understanding is complicated by the effects of spatial scale, because the scale of analysis affects the quantification of species-environment relationships. 2. We therefore assessed the influence of environmental context (i.e. the characteristics of the landscape surrounding a site), varied over a large range of scales (i.e. ambit radii around focal sites), on the analysis and prediction of habitat selection by African elephants in Kruger National Park, South Africa. 3. We focused on the spatial scaling of the elephants' response to their main resources, forage and water, and found that the quantification of habitat selection strongly depended on the scales at which environmental context was considered. Moreover, the inclusion of environmental context at characteristic scales (i.e. those at which habitat selectivity was maximized) increased the predictive capacity of habitat suitability models. 4. The elephants responded to their environment in a scale-dependent and perhaps hierarchical manner, with forage characteristics driving habitat selection at coarse spatial scales, and surface water at fine spatial scales. 5. Furthermore, the elephants exhibited sexual habitat segregation, mainly in relation to vegetation characteristics. Male elephants preferred areas with high tree cover and low herbaceous biomass, whereas this pattern was reversed for female elephants. 6. We show that the spatial distribution of elephants can be better understood and predicted when scale-dependent species-environment relationships are explicitly considered. This demonstrates the importance of considering the influence of spatial scale on the analysis of spatial patterning in ecological phenomena.  相似文献

Here we aim to demonstrate that in arid environments the competitive balance between species can be determined by niche separation with either nitrogen or water as the relevant niche axis. To do this we sampled roots <2 mm in diameter for 5 soil pits equidistant between two coexisting species, a shrub and a grass. Using stable carbon and nitrogen isotope ratios of fine roots we determine both photosynthetic pathway and rooting depth. We also examine the distribution of soil moisture and nitrogen relative to root biomass. Our results for root biomass and stable isotope ratios of fine roots demonstrate both niche separation and competition for resources. Root biomass is highest at the top of the profile where soil nitrogen is highest and soil moisture is lowest. We conclude that while there is competition for resources in the middle of the profile, competition is mitigated by photosynthetic pathway. The facultative CAM shrub grows whenever the soil at the surface is wet enough. The C4 photosynthetic pathway of the grass is more nitrogen and water use efficient making it better adapted to the low nitrogen in the middle of the profile and low summer rainfall.  相似文献
