The viral genome of Kaposi's sarcoma-associated herpesvirus (KSHV) persists as an extrachromosomal plasmid in latently infected cells. The KSHV latency-associated nuclear antigen (LANA) stimulates plasmid maintenance and DNA replication by binding to an approximately 150-bp region within the viral terminal repeats (TR). We have used chromatin immunoprecipitation assays to demonstrate that LANA binds specifically to the replication origin sequence within the KSHV TR in latently infected cells. The latent replication origin within the TR was also bound by LANA-associated proteins CBP, double-bromodomain-containing protein 2 (BRD2), and the origin recognition complex 2 protein (ORC2) and was enriched in hyperacetylated histones H3 and H4 relative to other regions of the latent genome. Cell cycle analysis indicated that the minichromosome maintenance complex protein, MCM3, bound TR in late-G(1)/S-arrested cells, which coincided with the loss of histone H3 K4 methylation. Micrococcal nuclease studies revealed that TRs are embedded in a highly ordered nucleosome array that becomes disorganized in late G(1)/S phase. ORC binding to TR was LANA dependent when reconstituted in transfected plasmids. DNA affinity purification confirmed that LANA, CBP, BRD2, and ORC2 bound TR specifically and identified the histone acetyltransferase HBO1 (histone acetyltransferase binding to ORC1) as a potential TR binding protein. Disruption of ORC2, MCM5, and HBO1 expression by small interfering RNA reduced LANA-dependent DNA replication of TR-containing plasmids. These findings are the first demonstration that cellular replication and origin licensing factors are required for KSHV latent cycle replication. These results also suggest that the KSHV latent origin of replication is a unique chromatin environment containing histone H3 hyperacetylation within heterochromatic tandem repeats.  相似文献

Abstract. Studies of seedling demography and dynamics were carried out in a dry forest at Pinkwae, Ghana (rainfall 1100 mm). Seedlings of tree and liana species in permanent transects were tagged, identified, measured and recensused at bimonthly intervals. A total of 1931 seedlings in 53 species were followed over 14 censuses during a 2-yr period. Germination and mortality were seasonal in their distribution; mortality was highest in dry periods. Seedling density varied seasonally, with the highest densities in comparatively wet periods. Diversity of seedlings was low, corresponding to that of the adult assemblage. Survivorship of tagged seedlings was recorded during the study period. The cohort tagged at the first census, comprising 1033 individuals of mixed ages and species, lost 61.9% during a 24-month period. Many seedlings were found to be rather large at the initial post-germination stage, with strong root development. Vegetative reproduction by root suckers or epicormic shoots was quite common at the site, although the majority of regeneration was by seeds. The current seedling assemblage appears to be representative, in terms of composition and diversity, of the adult species assemblage at Pinkwae.  相似文献

The cranial capacity of Olduvai Hominid 7 is estimated to be 690 cc, with a standard uncertainty range of 538 to 868 cc. The estimate is derived from a systematic consideration of the relationships between Bregma-Asterion chords and cranial capacities obtained from a large sample of Homo sapiens and Pan troglodytes and from available fossil hominids. Subpopulations of B-lymphocytes traffic to different sites and organs to provide diverse and tissue-specific functions. Here, we provide evidence that epigenetic differences confer a neuroinvasive phenotype. An EBV+ B cell lymphoma cell line (M14) with low frequency trafficking to the CNS was neuroadapted to generate a highly neuroinvasive B-cell population (MUN14). MUN14 B cells efficiently infiltrated the CNS within one week and produced neurological pathologies. We compared the gene expression profiles of viral and cellular genes using RNA-Seq and identified one viral (EBNA1) and several cellular gene candidates, including secreted phosphoprotein 1/osteopontin (SPP1/OPN), neuron navigator 3 (NAV3), CXCR4, and germinal center-associated signaling and motility protein (GCSAM) that were selectively upregulated in MUN14. ATAC-Seq and ChIP-qPCR revealed that these gene expression changes correlated with epigenetic changes at gene regulatory elements. The neuroinvasive phenotype could be attenuated with a neutralizing antibody to OPN, confirming the functional role of this protein in trafficking EBV+ B cells to the CNS. These studies indicate that B-cell trafficking to the CNS can be acquired by epigenetic adaptations and provide a new model to study B-cell neuroinvasion associated CNS lymphoma and autoimmune disease of the CNS, including multiple sclerosis (MS).  相似文献

Early modern and archaic humans are associated with similar lithic industries in the Middle Paleolithic of the southern Levant, but new data suggest that they used the environment in different ways. Evidence from analyses of seasonally deposited increments of the teeth of the animals they hunted suggests that modern humans primarily practiced a strategy ofcirculating seasonal mobility, while archaic humans in the same region 30,000 years later were more residentially mobile. Analyses of their lithic hunting technology further suggest that archaic humans hunted more frequently than did modern humans. We argue that this greater hunting intensity may have been a strategy for coping with the consequences of resource biodepletion resulting from long-term, multiseasonal occupation of sites. These behavioral contrasts may be related to some of the morphological differences between early modern and archaic humans.  相似文献
