Impact of resistance exercise during bed rest onskeletal muscle sarcopenia and myosin isoform distribution. J. Appl. Physiol. 84(1): 157-163, 1998.Because resistance exercise (REx) and bed-rest unloading (BRU)are associated with opposing adaptations, our purpose was to test theefficacy of REx against the effects of 14 days of BRU on theknee-extensor muscle group. Sixteen healthy men were randomly assignedto no exercise (NoEx; n = 8) or REx(n = 8). REx performed five sets ofleg press exercise with 80-85% of one repetition maximum (1 RM)every other day during BRU. Muscle samples were removed from the vastuslateralis muscle by percutaneous needle biopsy. Myofiber distributionwas determined immunohistochemically with three monoclonal antibodiesagainst myosin heavy chain (MHC) isoforms (I, IIa, IIx). MHCdistribution was further assessed by quantitative gel electrophoresis.Dynamic 1-RM leg press and unilateral maximum voluntary isometriccontraction (MVC) were determined. Maximal neural activation (root meansquared electromyogram) and rate of torque development (RTD) weremeasured during MVC. Reductions(P < 0.05) in type I (15%) and typeII (17%) myofiber cross-sectional areas were found in NoEx but not inREx. Electrophoresis revealed no changes in MHC isoform distribution. The percentage of type IIx myofibers decreased(P < 0.05) in REx from 9 to 2% anddid not change in NoEx. 1 RM was reduced(P < 0.05) by 9% in NoEx but wasunchanged in REx. MVC fell by 15 and 13% in NoEx and REx,respectively. The agonist-to-antagonist root mean squaredelectromyogram ratio decreased (P < 0.05) 19% in REx. RTD slowed (P < 0.05) by 54% in NoEx only. Results indicate that REx preventedBRU-induced myofiber atrophy and also maintained training-specificstrength. Unlike spaceflight, BRU did not induce shifts in myosinphenotype. The reported benefits of REx may prove useful in prescribingexercise for astronauts in microgravity.

This paper examines differences in the processes by which the cranial base flexes in humans and extends in chimpanzees. In addition, we test the extent to which one can use comparisons of cranial base angles in humans and non-human primates to predict vocal tract dimensions. Four internal cranial base angles and one external cranial base angle were measured in a longitudinal sample of Homo sapiens and a cross-sectional sample of Pan troglodytes. These data show that the processes of cranial base angulation differ substantially in these species. While the human cranial base flexes postnatally in a rapid growth trajectory that is complete by two years, the cranial base in P. troglodytes extends postnatally in a more prolonged skeletal growth trajectory. These comparisons also demonstrate that the rate of cranial base angulation is comparable for different measures, but that angles which incorporate different anterior cranial base measurements correlate poorly. We also examined ontogenetic relationships between internal and external cranial base angles and vocal tract growth in humans to test the hypothesis that cranial base angulation influences pharyngeal dimensions and can, therefore, be used to estimate vocal tract proportions in fossil hominids. Our results indicate that internal and external cranial base angles are independent of the horizontal and vertical dimensions of the vocal tract. Instead, a combination of mandibular and palatal landmarks can be used to predict dimensions of the vocal tract in H. sapiens. The developmental contrasts in cranial base angulation between humans and non-human primates may have important implications for testing hypotheses about the relationship between cranial base flexion and other craniofacial dimensions in hominid evolution.  相似文献

Summary We present evidence of a difference between a storing and non-storing species in lateralization and transfer of spatial memory processing. Using the technique of monocular occlusion, we compared the performance of a food-storing species, the marsh tit (Parus palustris), with a closely related species that does not store food, the blue tit (P. caeruleus), on a task which relies on one-trial learning for the spatial location of hidden food items. In this one-trial associative learning task, the birds had to return to sites in phase II of a trial where they had been allowed to eat some, but not all, of a piece of peanut in phase I. In the first experiment, in which the birds did not wear eye caps, marsh tits required fewer looks in phase II to find the hidden peanut than blue tits. By categorising the sites as seeded, unseeded or not visited in phase I, further analysis suggested that the two species also differ in the way they behave towards the 3 types of site. Marsh tits appear to distinguish between seeded and other sites, irrespective of whether the other sites were known to be empty (unseeded) or not (not visited); whereas blue tits preferentially returned to those sites that have been visited in phase I, irrespective of whether they contained a seed or not. In the second experiment, all birds wore an eye cap on the left or right eye for phase I and II of each trial. For both species, it was demonstrated that although the visual systems fed by both the right and left eye are involved in short-term storage, the right eye system is associated also with long-term storage. Thus, lateralization was found in both blue tits and marsh tits. However, unilateral transfer of these memories was found only in marsh tits, suggesting that there may be a difference between storers and non-storers in the mechanism of memory processing.  相似文献

Summary The results reported in this paper demonstrate lateralization and transfer of spatial memory processing in an adult, food-storig bird. A monoclonal antibody (C219) that recognizes the P-glycoprotein (Mr = 170,000) in plasma membranes of multidrug-resistant Chinese hamster ovary (CHO) cell lines was used to assay renal brush border membrane (BBM) and basolateral membrane (BLM) fractions for the presence of a cross-reactive polypeptide. The C219 antibody bound to a 155,000 dalton protein in immunoblots of rat BBM but not BLM proteins resolved by sodium dodecyl sulfate gel electrophoresis. The corresponding human kidney BBM and dog kidney BBM proteins had molecular weights of 170,000 and 160,000 respectively. The glycoprotein nature of the renal protein was shown by its sensitivity to N-glycanase treatment which reduced the apparent molecular weight of the dog protein to 120,000. In addition, dog P-glycoprotein could be bound to and eluted from immobilized wheat germ agglutinin. The molecular weight, antibody crossreactivity, glycosidase sensitivity and lectin binding show that this protein is a normal kidney analogue of the P-glycoprotein induced in multidrug resistant cell lines.  相似文献

The human erythrocyte anion exchange protein, Band 3, was reacted with N-ethylmaleimide (NEM) in cells to a stoichiometry of 5.3 mol NEM per mol Band 3, indicating that all NEM-reactive cysteines in Band 3 were labeled. Quantitatively NEM-blocked Band 3 was still able to bind to and be eluted by reducing agents from a mercurial affinity resin, [p-(chloromercuribenzamido)ethylene]amino-Sepharose. Reaction of NEM-blocked Band 3 with p-chloromercuribenzoate (pCMB) did not prevent binding to the resin due to exchange of pCMB for the immobilized mercurial. pCMB has been reported to inhibit water and urea permeation across the red cell membrane, and this has been attributed to reaction with a NEM-reactive sulfhydryl in Band 3. The interaction of Band 3 with the immobilized ligand directly demonstrates the reaction of NEM-blocked Band 3 with a mercurial and indicates that the NEM-unreactive, pCMB-reactive sulfhydryl residue is accessible to within approximately equal to 12 A (the distance from the solid support to the Hg) of the surface of the solubilized Band 3 protein.  相似文献

Acromegalic patients suffer from a number of cardiovascular, metabolic, and rheumatologic problems, and they may also have an increased incidence of malignancy. We reviewed the literature concerning the reversibility of acromegalic complications. Hypertension, myocardial hypertrophy, left ventricular dysfunction and some rheumatologic abnormalities often continue despite successful treatment of the acromegaly. In contrast, glucose intolerance, soft tissue changes, and carpal tunnel syndrome usually resolve when the acromegaly is cured. Studies of the incidence and mortality of cancer in acromegaly are conflicting, but several suggest an increased incidence of colorectal cancer.  相似文献

Coinjection strategy for visual identification of transgenic mice   总被引:10,自引:0,他引:10  
Transgenic mice were generated by coinjection of a dominant marker gene that induces fur and eye pigmentation (a tyrosinase minigene) plus an unrelated DNA construction that has a γ-glutamyl transferase (γGT) promoter linked to aras oncogene. Mice transgenic for γGT-ras could be identified in the first and all subsequent generations by simple visual inspection for pigmentation. Furthermore, the γ-glutamyl transferase promoter was active in kidney but not skin of the transgenic mice, indicating that the cointegrated DNA was active and independently expressed. These results confirm that the tyrosinase minigene can be used for coinjections to allow rapid visual identification of transgenic mice.  相似文献

Because women's preferences for male masculinity reflect tradeoffs between the benefits of greater genetic health and the costs of lower paternal investment, variables that affect the importance of these costs and benefits also affect masculinity preferences. Concern about disease and pathogens may be one such variable. Here we show that disgust sensitivity in the pathogen domain is positively correlated with facial masculinity preferences, but disgust sensitivity in the moral and sexual domains is not. Our findings present novel evidence that systematic variation in women's preferences for masculine men reflects factors that influence how women resolve the tradeoff between the benefits and costs associated with choosing a masculine partner.  相似文献
