The biological oxygen demand (BOD) of filtered water from Lake Wingra, Wisconsin is significantly higher in the littoral zone than in the pelagial zone. Laboratory experiments indicate that BOD is not influenced by water temperature at the time of sampling or by enrichment with nitrate or ammonia. Rather, enrichment with macrophyte leachate sharply increases BOD, and enrichment with phosphate produces a small but significant increase in BOD. We conclude that high BOD in littoral waters of the lake is an indication of production of labile organic matter in the water by dense beds of the macrophyte Myriophyllum spicatum.  相似文献

A detergent-solubilized form of H-2b (dH-2b) has been purified 1500-fold from RBL-5 tumor cells. The purification was accomplished by deoxycholate solubilization of purified plasma membranes, gel filtration, Lens culinaris lectin affinity chromatography, and affinity chromatography on a sheep anti-dH-2b immunoadsorbent. Both alloantigen and beta 2-microglobulin were monitored by radioimmunoassay during purification. The final product was judged to be greater than 90% pure by the following criteria: 1) sodium dodecyl sulfate-polyacrylamide electrophoresis which showed the expected 2-component structure of histocompatibility antigens, i.e. a heavy chain and beta 2-microglobulin; 2) amino acid composition which was comparable to the known compositions of other H-2 and HLA molecules; 3) NH2-terminal sequencing which gave a unique sequence for the heavy chain, and the reported sequence for beta 2-microglobulin; and 4) immunoprecipitation of the bulk of the preparation by appropriate alloantisera.  相似文献

A 21-year-old male patient underwent aortic and mitral valve replacement for progressive cardiac failure due to acute bacterial endocarditis. Ischemic myocardial contracture developed during attempts to restore cardiac activity following hypothermic, ischemic, cardioplegic arrest. An abdominal left ventricular assist device (ALVAD) was implanted and supported the circulation for nearly six days prior to cardiac transplantation. The preoperative EKG showed sinus tachycardia with left anterior hemiblock. Postoperatively, there was complete electromechanical dissociation. The postoperative EKG showed a superior and leftward shift of the axis. There was a marked loss of QRS voltage and variable degrees of atrioventricular block. At times, only P waves were present. On the fourth postoperative day, there was an axis shift to the extreme right. Prior to transplantation, sinus rhythm returned, and the axis shifted leftward once again. The common denominator of all the abnormal postoperative electrocardiograms was the conspicuous low voltage that probably signified early and extensive myocardial damage. To our knowledge, this is the first instance wherein a sequential electrocardiographic analysis of stone heart syndrome has been undertaken.  相似文献

Experimental intrauterine growth retardation was studied in sheep. Endometrial caruncles (anlagen of maternal cotyledon) were removed before pregnancy and at a second operation, catheters were implanted into the ewe and fetus at 105-135 days of pregnancy. Three groups of fetuses were defined: low birthweight-for-dates (small-caruncle), normal birthweight-for-dates (normal-sized-caruncle) from ewes which had endometrial caruncles removed and the controls. The mean placental weights in these groups were 139 plus or minus 5 g, 283 plus or minus 46 g, 334 plus or minus 22 g respectively. The brains, kidneys and adrenals of the small-caruncle-fetuses were significantly greater in proportion to body weight than in the controls and the appearance of ossification centres was delayed. Arterial oxygen tension was lower and packed cell volume higher in the small-caruncle-fetuses (PaO2 15 plus or minus 0.6 mmHg; packed cell volume 37.3 plus or minus 1.6%) and normal sized caruncles (PaO2 20.7 plus or minus 1.2 mmHg; packed cell volume 35.2 plus or minus 0.7%) than in the controls (PaO2 23.2 plus or minus 0.7 mmHg; packed cell volume 29.8 plus or minus 0.7%). Plasma concentrations of glucose (0.65 plus or minus 0.12 micromol/ml), lactate (0.9 plus or minus 0.1 micromol/ml) and pyruvate (0.08 plus or minus 0.025 micromol/ml) were lower in small-caruncle fetuses than in the control fetuses (glucose 1.05 plus or minus 0.06 micromol/ml, lactate 1.83 plus or minus 0.7 micromol/ml, pyruvate 0.21 plus or minus 0.06 micromol/ml). The corresponding values for the normal-sized-caruncle fetuses were glucose 0.71 plus or minus 0.12, lactate 1.18 plus or minus 0.7 and pyruvate 0.12 plus or minus 0.03 micromol/ml. The plasma concentration of alanine in the small-caruncle-fetuses (0.25 plus or minus 0.09 micromol/ml) was higher than in the normal-sized-caruncle (0.073 plus or minus 0.009 micromol/ml) or control fetuses (0.12 plus or minus 0.013 micromol/ml). The results indicate that fetal growth retardation due to restriction of placental growth after removal of endometrial caruncles is associated with chronic hypoxaemia, polycythaemia and hypoglycaemia. The restriction of nutrient supply probably accounts for the altered pattern of fetal growth but the relative importance of the changes observed remains uncertain.  相似文献

Summary A knowledge of the precise location of neurosecretory cell bodies is a prerequisite for studying the synthesis and subsequent processing of neurosecretory polypeptides stored in axon terminals comprising the sinus gland of the crustacean eyestalk. Structural data establish that the X organ in the medulla terminalis ganglion (mtXo) of the crayfish eyestalk represents 90–95% of the cell bodies actively synthesizing neurosecretory vesicles stored in the neurohemal sinus gland (Fig. 4). These cell bodies transport rather than accumulate neurosecretory vesicles as judged by light and electron microscopy suggesting that neurohormone precursors, but not subsequently stored products, might be found there. Two-dimensional electrophoresis of sinus gland and mtXo homogenates support this hypothesis. In crayfish, lobster and blue crab, stained two-dimensional gels display a number of sinus gland-specific polypeptides whose high concentrations and low molecular weights are consistent with stored neurosecretory material (Table 1). These neuropeptides are not detected in mtXo homogenates or in non-neurosecretory neural tissue with Coomassie Blue staining. By decreasing the porosity of the second dimension, the two-dimensional gel technique has proven useful in determining the molecular weights of a variety of neurosecretory polypeptides stored in the sinus gland. The crayfish and lobster store several polypeptides of ca. 7,000 Dalton. The blue crab stores two 7,000, two 13,000 and three 20,000 Dalton sinus gland polypeptides detected in stained gels.Following a 4 h incubation in3H-labelled amino acids, predominantly labelled 19,000–21,000 Dalton polypeptides are detected in crayfish mtXo homogenates by 2-D gel autoradiography (Fig. 12). Concomitantly, three labelled polypeptides (4,000–10,000 Dalton) appear in the sinus gland (Fig. 13), suggesting that they are cleaved from 19,000–21,000 Dalton molecules. When histone is oxidized by peroxidase, its basicity (hence its complexing with DNA) is reduced: this reduction causes further alterations in the effect of histone upon the heat denaturation, acid precipitation, and breakdown by DNase of DNA, alterations which indicate that the regulation by histone of DNA expression may become abnormal. If oxidized species of histone should accumulate in the tissues in old age, the alteration mentioned might be a contributory factor of senescence.  相似文献

Summary The plasmatocytes are the major phagocytic blood-cell type in the haemolymph of the wax-moth, Galleria mellonella. In the present study, these cells were allowed to attach to tissue culture dishes for 1 h, rinsed and then incubated with latex beads for up to 72 h. These cells were then fixed for routine transmission electron microscopy and acid phosphatase cytochemistry. Intracellular latex particles were found in tight, ill-defined phagosomes, which were often clearly associated with the Golgi complexes of the plasmatocytes. Fusion of both primary lysosomes and multivesicular bodies with the phagosomes occasionally occurred and this resulted in the accumulation of an acid phosphatase positive reaction product around the test particles. Subsequent experiments showed that this acid phosphatase activity was mainly associated with the primary lysosomes. The results of the lysosome/latex interactions are compared with those obtained from similar studies on the digestive mechanisms in other phagocytes.We are grateful to Professor E.W. Summary A woman in the fourth year of agnogenic myeloid metaplasia was found to have partial deletion of the long arm of chromosome 20 [46,XX, del(20)(q11)] in mitoses of presumably immature myeloid cells from unstimulated cultures of peripheral blood and bone marrow. Cytogenetic studies of peripheral blood lymphocytes showed a normal female karyotype.  相似文献

Synopsis Latent period, rise time, contraction time, and half relaxation time from isotonic contractions of isolated white muscle samples from skipjack tuna, Katsuwonus pelamis, were determined at 20°, 27°, and 34° C. These parameters were found to be inversely proportional to temperature (Q10 = 1.47, 1.67, 1.62, and 1.72, respectively). The data show that contraction time and the effect of temperature on contraction time of skipjack tuna white muscle are not unique when compared to other equal-sized teleosts. Based on contraction time, maximum swimming speeds at each muscle temperature were calculated and found not significantly to exceed the maximum speeds of other equal-sized teleosts, when comparisons are made at the same white muscle temperatures  相似文献
