Strains of Bacillus cereus can produce a heat-stable toxin (cereulide). In this study, 101 Bacillus strains representing 7 Bacillus species were tested for production of heat-stable toxins. Strains of B. megaterium, B. firmus and B. simplex were found to produce novel heat-stable toxins, which showed varying levels of toxicity. B. cereus strains (18 out of 54) were positive for toxin production. Thirteen were of serovar H1, and it was of interest that some were of clinical origin. Two were of serovars 17B and 20, which are not usually implicated in the emetic syndrome. Partial purification of the novel B. megaterium, B. simplex and B. firmus toxins showed they had similar physical characteristics to the B. cereus emetic toxin, cereulide.  相似文献

Abstract

Herein we report the quantification of purine lesions arising from gamma-radiation sourced hydroxyl radicals (HO?) on tertiary dsDNA helical forms of supercoiled (SC), open circular (OC), and linear (L) conformation, along with single-stranded folded and non-folded sequences of guanine-rich DNA in selected G-quadruplex structures. We identify that DNA helical topology and folding plays major, and unexpected, roles in the formation of 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxo-dG) and 8-oxo-7,8-dihydro-2'-deoxyadenosine (8-oxo-dA), along with tandem-type purine lesions 5′,8-cyclo-2′-deoxyguanosine (5′,8-cdG) and 5′,8-cyclo-2′-deoxyadenosine (5′,8-cdA). SC, OC, and L dsDNA conformers together with folded and non-folded G-quadruplexes d[TGGGGT]4 (TG4T), d[AGGG(TTAGGG)3] (Tel22), and the mutated tel24 d[TTGGG(TTAGGG)3A] (mutTel24) were exposed to HO? radicals and purine lesions were then quantified via stable isotope dilution LC-MS/MS analysis. Purine oxidation in dsDNA follows L?>?OC???SC indicating greater damage towards the extended B-DNA topology. Conversely, G-quadruplex sequences were significantly more resistant toward purine oxidation in their unfolded states as compared with G-tetrad folded topologies; this effect is confirmed upon comparative analysis of Tel22 (~50% solution folded) and mutTel24 (~90% solution folded). In an effort to identify the accessibly of hydroxyl radicals to quadruplex purine nucleobases, G-quadruplex solvent cavities were then modeled at 1.33?Å with evidence suggesting that folded G-tetrads may act as potential oxidant traps to protect against chromosomal DNA damage.  相似文献

Conduction along the optic nerve is often slowed in multiple sclerosis (MS). This is typically assessed by measuring the latency of the P100 component of the Visual Evoked Potential (VEP) using electroencephalography. The Visual Evoked Spread Spectrum Analysis (VESPA) method, which involves modulating the contrast of a continuous visual stimulus over time, can produce a visually evoked response analogous to the P100 but with a higher signal-to-noise ratio and potentially higher sensitivity to individual differences in comparison to the VEP. The main objective of the study was to conduct a preliminary investigation into the utility of the VESPA method for probing and monitoring visual dysfunction in multiple sclerosis. The latencies and amplitudes of the P100-like VESPA component were compared between healthy controls and multiple sclerosis patients, and multiple sclerosis subgroups. The P100-like VESPA component activations were examined at baseline and over a 3-year period. The study included 43 multiple sclerosis patients (23 relapsing-remitting MS, 20 secondary-progressive MS) and 42 healthy controls who completed the VESPA at baseline. The follow-up sessions were conducted 12 months after baseline with 24 MS patients (15 relapsing-remitting MS, 9 secondary-progressive MS) and 23 controls, and again at 24 months post-baseline with 19 MS patients (13 relapsing-remitting MS, 6 secondary-progressive MS) and 14 controls. The results showed P100-like VESPA latencies to be delayed in multiple sclerosis compared to healthy controls over the 24-month period. Secondary-progressive MS patients had most pronounced delay in P100-like VESPA latency relative to relapsing-remitting MS and controls. There were no longitudinal P100-like VESPA response differences. These findings suggest that the VESPA method is a reproducible electrophysiological method that may have potential utility in the assessment of visual dysfunction in multiple sclerosis.  相似文献

Polyandry is widespread and influences patterns of sexual selection, with implications for sexual conflict over mating. Assessing sperm precedence patterns is a first step towards understanding sperm competition within a female and elucidating the roles of male- and female-controlled factors. In this study behavioural field data and genetic data were combined to investigate polyandry in the chokka squid Loligo reynaudii. Microsatellite DNA-based paternity analysis revealed multiple paternity to be the norm, with 79% of broods sired by at least two males. Genetic data also determined that the male who was guarding the female at the moment of sampling was a sire in 81% of the families tested, highlighting mate guarding as a successful male tactic with postcopulatory benefits linked to sperm deposition site giving privileged access to extruded egg strings. As females lay multiple eggs in capsules (egg strings) wherein their position is not altered during maturation it is possible to describe the spatial / temporal sequence of fertilisation / sperm precedence There were four different patterns of fertilisation found among the tested egg strings: 1) unique sire; 2) dominant sire, with one or more rare sires; 3) randomly mixed paternity (two or more sires); and 4) a distinct switch in paternity occurring along the egg string. The latter pattern cannot be explained by a random use of stored sperm, and suggests postcopulatory female sperm choice. Collectively the data indicate multiple levels of male- and female-controlled influences on sperm precedence, and highlights squid as interesting models to study the interplay between sexual and natural selection.  相似文献

In this paper, we focus on the impact on soil organic carbon (SOC) of two dedicated energy crops: perennial grass Miscanthus x Giganteus (Miscanthus) and short rotation coppice (SRC)‐willow. The amount of SOC sequestered in the soil is a function of site‐specific factors including soil texture, management practices, initial SOC levels and climate; for these reasons, both losses and gains in SOC were observed in previous Miscanthus and SRC‐willow studies. The ECOSSE model was developed to simulate soil C dynamics and greenhouse gas emissions in mineral and organic soils. The performance of ECOSSE has already been tested at site level to simulate the impacts of land‐use change to short rotation forestry (SRF) on SOC. However, it has not been extensively evaluated under other bioenergy plantations, such as Miscanthus and SRC‐willow. Twenty‐nine locations in the United Kingdom, comprising 19 paired transitions to SRC‐willow and 20 paired transitions to Miscanthus, were selected to evaluate the performance of ECOSSE in predicting SOC and SOC change from conventional systems (arable and grassland) to these selected bioenergy crops. The results of the present work revealed a strong correlation between modelled and measured SOC and SOC change after transition to Miscanthus and SRC‐willow plantations, at two soil depths (0–30 and 0–100 cm), as well as the absence of significant bias in the model. Moreover, model error was within (i.e. not significantly larger than) the measurement error. The high degrees of association and coincidence with measured SOC under Miscanthus and SRC‐willow plantations in the United Kingdom, provide confidence in using this process‐based model for quantitatively predicting the impacts of future land use on SOC, at site level as well as at national level.  相似文献

This article examines the interplay of ethnic conflict and economic interests in Northern Ireland, specifically for the case of the shipbuilding industry, which received massive state aid despite its terminal decline. Parkin's theory of dual social closure by members of the subordinate class but dominant status group to monopolize resources and opportunities is employed to examine the interests and actions of Protestant workers. The contribution of local institutions including Extended Internal Labor Markets to the salience of Protestant group solidarity is examined from a political economy perspective. Political accommodation between Protestant labour and the Northern Ireland government, and later British government political fears of antagonizing Loyalist workers, facilitated huge aid to a failing industry.  相似文献

Cell-free translation of the Semliki Forest virus-specific 26S RNA yielded primarily capsid protein. After treatment of the protein synthesising reaction with 25 mM EDTA, the capsid protein cosedimented with the large ribosomal subunit in sucrose gradients, and banded with the subunit at a density of 1.54 gm/cm3 in CsCl. Exposure to 0.5 M KCl released the protein from the subunit. Similar binding of the virus capsid protein to the large ribosomal subunit has been observed in infected HeLa cells, although its function is not clear. The nonstructural proteins, which are the major products translated from the virion 42S RNA, did not associate with sedimenting structures.  相似文献

In-situ artificial stream channels and ex-situ laboratory mesocosms were used to measure the responses of macroinvertebrate assemblages, from upland and lowland tropical streams, to high loads of fine clay sediment. Significant responses were observed mainly in the in-situ channels in the upland stream, where densities and the number of taxa were lower in the treatment channels than in controls. There was no evidence of any taxon being particularly sensitive to sedimentation, with a general decrease in densities across several taxa and differences only detectable for abundant taxa. Animals moved downstream in response to the treatment, but only a short distance within the channels. However, further colonization after the treatment was impeded in the treatment channels. In the mesocosm experiments, the upland macroinvertebrate assemblage demonstrated some negative effects; however, the lowland fauna was very tolerant to sedimentation, even when exposure was extended to 15 days. Together, the mesocosm and in-situ experiments indicate that there is a behavioural response to sedimentation because of a change in the habitat, and that the lowland macroinvertebrate assemblage is more tolerant of sedimentation, at least in the short term. Nano-functionalized products such as UV protective paints additives, antimicrobial food packaging, and fuel additives offering reduced CO2 emissions have the potential to secure a significant Irish market share in the near future. This scoping study gives a first estimation of nanomaterial surface water concentrations and population ingestional exposure through drinking water resulting from these products. As nanomaterial behavior in wastewater treatment plants (WWTPs) and water treatment plants (WTPs) is currently unclear, bridging data relating to potentially relevant materials (pharmaceuticals and metal removal efficiencies in WWTPs; pathogen removal efficiencies in WTPs) are employed in this study. Mean nanomaterial removal efficiencies of 59.8% and 70.2% were predicted for Irish WWTPs, between 96.95% and 0% for Irish WTPs. Predicted nano-scale TiO2 concentrations in surface waters (resulting from exterior paints) were 2 orders of magnitude greater than that of Ag (resulting from food packaging) and CeO2 (resulting from fuel additives), respectively. Predicted surface and drinking water concentrations were unlikely to pose any ecotoxicological or human health risk, although nano-scale TiO2 and Ag may warrant monitoring as part of standard surface water monitoring schemes. Future research should be directed toward characterizing the behavior of different categories of nanomaterials within WWTP processes.  相似文献
