A series of inhibitors of mammalian 15-lipoxygenase based on tryptamine and homotryptamine scaffolds is described. Compounds with aryl substituents at C-2 of the indole core of tryptamine and homotryptamine sulfonamides (e.g., 37a-p) proved to be potent inhibitors of the isolated enzyme. Selected compounds also demonstrated desirable inhibition selectivities over isozymes 5- and P-12-LO.  相似文献

Biological activity of the bacterium Bacillus thuringiensis Berliner (Bt) against insect pests is influenced by the host plants. To understand the underlying mechanism of variation in biological activity of Bt on host plants, we studied the effect of chemicals from the surface of chickpea (Cicer arietinum L., Fabaceae) leaves (ICCC 37 and ICC 506EB), sorghum [Sorghum bicolor (L.) Moench, Poaceae] grain (ICSV 745 and IS 18698), pigeon pea [Cajanus cajan (L.) Millsp., Fabaceae] pods (ICPL 87 and ICPL 332WR), and cotton (Gossypium hirsutum L., Malvaceae) squares (RCH 2 and Bt RCH 2), on which Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) feeds under natural conditions. Surface chemicals extracted in water from host plant leaves were added to the standard artificial diet containing a commercial formulation of Bt or Cry1Ac. Data were recorded on larval and pupal weights, pupation, adult emergence, larval and pupal periods, adult longevity, and fecundity. Weights of H. armigera at 5 days after initiation of the experiment were significantly reduced on artificial diets containing Bt + pod washings of ICPL 87 and ICPL 332WR, grain washings of ICSV 745, or square washings of RCH 2, and Cry1Ac + leaf‐surface washings of ICC 506EB. Pupal weights were lower on diets containing leaf‐surface washings of ICCC 37 + Bt than on standard artificial diet. Larval periods were prolonged on diets containing Bt + leaf‐surface washings of ICCC 37, pod washings of ICPL 87, and square washings of RCH 2, and on standard artificial diet + Cry1Ac. Pupation was significantly higher on standard artificial diet + Cry1Ac than on diets with Bt + grain washings of ICSV 745 and Cry1Ac + square washings of RCH 2 and Bt RCH 2. Adult emergence was lowest on diets with square washings of RCH 2 + Bt and grain washings of ICSV 745 + Cry1Ac. The results suggested that leaf‐surface washings play an important role in biological activity of Bt/Cry1Ac against H. armigera.  相似文献

Rubyspira, a new genus of deep-sea snails (Gastropoda: Abyssochrysoidea) with two living species, derives its nutrition from decomposing whalebones. Molecular phylogenetic and morphological evidence places the new genus in an exclusively deep-sea assemblage that includes several close relatives previously known as fossils associated with Cretaceous cold seeps, plesiosaur bones, and Eocene whalebones. The ability to exploit a variety of marine reducing environments may have contributed to the evolutionary longevity of this gastropod lineage.  相似文献

The earthworm Lumbricus rubellus has been found to inhabit cadmium-rich soils and accumulate cadmium within its tissues. Two metallothionein (MT) isoforms (1 and 2) have been identified and cloned from L. rubellus. In this study, we address the metalation status, metal coordination, and structure of recombinant MT-2 from L. rubellus using electrospray ionization mass spectrometry (ESI-MS), UV absorption, and circular dichroism (CD) spectroscopy. This is the first study to show the detailed mass and CD spectral properties for the important cadmium-containing earthworm MT. We report that the 20-cysteine L. rubellus MT-2 binds seven Cd(2+) ions. UV absorption and CD spectroscopy and ESI-MS pH titrations show a distinct biphasic demetalation reaction, which we propose results from the presence of two metal-thiolate binding domains. We propose stoichiometries of Cd(3)Cys(9) and Cd(4)Cys(11) based on the presence of 20 cysteines split into two isolated regions of the sequence with 11 cysteines in the N-terminal and 9 cysteines in the C-terminal. The CD spectrum reported is distinctly different from any other metallothionein known suggesting quite different binding site structure for the peptide.  相似文献

Induced resistance is one of the important components of host plant resistance to insects. We studied the induced defensive responses in groundnut genotypes with different levels of resistance to the leaf defoliator Helicoverpa armigera and the sap-sucking insect Aphis craccivora to gain an understanding of the induced resistance to insects and its implications for pest management. The activity of the defensive enzymes (peroxidase, polyphenol oxidase, phenylalanine ammonia lyase, superoxide dismutase, ascorbate peroxidase, and catalase) and the amounts of total phenols, hydrogen peroxide, malondialdehyde, and proteins were recorded at 6 days after infestation. Induction of enzyme activities and the amounts of secondary metabolites were greater in the insect-resistant genotypes ICGV 86699, ICGV 86031, ICG 2271, and ICG 1697 infested with H. armigera and A. craccivora than in the susceptible check JL 24. The resistant genotypes suffered lower insect damage and resulted in lower Helicoverpa larval survival and weights than those larvae fed on the susceptible check JL 24. The number of aphids was significantly lower on insect-resistant genotypes than on the susceptible check JL 24. The results suggested that groundnut plants respond to infestation by H. armigera and A. craccivora in a similar way; however, the degree of the response differed across the genotypes and insects, and this defense response is attributed to various defensive enzymes and secondary metabolites.  相似文献

Jasmonic acid (JA) acts as a signal molecule to induce resistance in plants against herbivores and its levels are elevated in plants after wounding or insect damage. Groundnut is an important crop in many tropical and subtropical regions worldwide, but there is surprisingly little knowledge on its induced defenses against herbivores. The effect of JA as a spray on induced resistance in three groundnut genotypes, namely, ICGV 86699 (resistant), NCAc 343 (resistant), and TMV 2 (susceptible), against Helicoverpa armigera was studied. The activity of oxidative enzymes [peroxidase (POD) and polyphenol oxidase (PPO)] and the amounts of other host plant defense components [total phenols, hydrogen peroxide (H2O2), malondialdehyde (MDA), and protein content] were recorded at 24, 48, 72, and 96 h after pretreatment (1 day) with JA followed by infestation with H. armigera (PJA + HIN) and H. armigera infestation with simultaneous JA application (HIN + JA) to understand the consequences of induced resistance in groundnut. The plant damage, larval survival, and larval weights were also recorded. There was a significant increase in POD and PPO activities and in the amounts of total phenols, H2O2, MDA, and proteins in PJA + HIN- and JA + HIN-treated plants as compared to the plants treated with JA and infested with H. armigera individually and to untreated control plants. Among all the genotypes, the strongest induction of defense was observed in the ICGV 86699 genotype. It is concluded that pretreatment with JA and its application during low levels of insect infestation can increase the levels of host plant resistance against herbivorous insects and reduce the pest-associated losses in groundnut.  相似文献

Background

Control of bovine tuberculosis (bTB) and zoonotic tuberculosis (zTB) has relied upon surveillance and slaughter of infected cattle, milk pasteurisation and public health education. In Cameroon, like many other sub-Saharan African countries, there is limited understanding of current cattle husbandry or milk processing practices or livestock keepers awareness of bTB. This paper describes husbandry and milk processing practices within different Cameroonian cattle keeping communities and bTB awareness in comparison to other infectious diseases.

Study design

A population based cross-sectional sample of herdsmen and a questionnaire were used to gather data from pastoralists and dairy farmers in the North West Region and Vina Division of Cameroon.

Results

Pastoralists were predominately male Fulanis who had kept cattle for over a decade. Dairy farmers were non-Fulani and nearly half were female. Pastoralists went on transhumance with their cattle and came into contact with other herds and potential wildlife reservoirs of bTB. Dairy farmers housed their cattle and had little contact with other herds or wildlife. Pastoralists were aware of bTB and other infectious diseases such as foot-and-mouth disease and fasciolosis. These pastoralists were also able to identify clinical signs of these diseases. A similar proportion of dairy farmers were aware of bTB but fewer were aware of foot-and-mouth and fasciolosis. In general, dairy farmers were unable to identify any clinical signs for any of these diseases. Importantly most pastoralists and dairy farmers were unaware that bTB could be transmitted to people by consuming milk.

Conclusions

Current cattle husbandry practices make the control of bTB in cattle challenging especially in mobile pastoralist herds. Routine test and slaughter control in dairy herds would be tractable but would have profound impact on dairy farmer livelihoods. Prevention of transmission in milk offers the best approach for human risk mitigation in Cameroon but requires strategies that improved risk awareness amongst producers and consumers.  相似文献

The sterile insect technique (SIT) is widely used in integrated programs against fruit fly pests, particularly the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae). Unfortunately, the mass-rearing procedures inherent to the SIT often lead to a reduction in male mating competitiveness. One potential solution involves the pre-release exposure of males to specific attractants. In particular, male exposure to ginger root oil [Zingiber officinale Roscoe (Zingiberaceae); hereafter GRO] has been shown to increase mating success dramatically in field cage trials. Initial studies exposed small groups of males (25 individuals), but more recent work has demonstrated that GRO exposure involving standard storage boxes (containing ≈ 36 000 males) also results in enhanced mating performance. The objective of the present study was to determine whether aromatization of entire trailers, holding ≈ 14 million sterile males from a genetic sexing [temperature sensitive lethal (tsl)] strain, increases male mating success. Independent of the total dose, spatial distribution, or type of dispenser used, sterile males exposed to GRO for a 24-h period displayed greater mating success than non-exposed males in mating cage trials (in which tsl males competed against males from a standard, bisexual strain for females from this same standard strain). Averaged over all experiments, tsl males exposed to GRO obtained 54% of all matings compared to 38% for non-exposed tsl males, an increase of 42%. The implications of these findings for SIT programs against C. capitata are discussed.  相似文献

Actin stress fibers (SFs) play an important role in many cellular functions, including morphological stability, adhesion, and motility. Because of their central role in force transmission, it is important to characterize the mechanical properties of SFs. However, most of the existing studies focus on properties of whole cells or of actin filaments isolated outside cells. In this study, we explored the mechanical properties of individual SFs in living endothelial cells by nanoindentation using an atomic force microscope. Our results demonstrate the pivotal role of SF actomyosin contractile level on mechanical properties. In the same SF, decreasing contractile level with 10 μM blebbistatin decreased stiffness, whereas increasing contractile level with 2 nM calyculin A increased stiffness. Incrementally stretching and indenting SFs made it possible to determine stiffness as a function of strain level and demonstrated that SFs have nearly linear stress-stain properties in the baseline state but nonlinear properties at a lower contractile level. The stiffnesses of peripheral and central portions of the same SF, which were nearly the same in the baseline state, became markedly different after contractile level was increased with calyculin A. Because these results pertain to effects of interventions in the same SF in a living cell, they provide important new understanding about cell mechanics.  相似文献
