The I locus of the common bean, Phaseolus vulgaris, controls the development of four different phenotypes in response to inoculation with Bean common mosaic virus, Bean common mosaic necrosis virus, several other related potyviruses, and one comovirus. We have generated a high-resolution linkage map around this locus and have aligned it with a physical map constructed with BAC clones. These clones were obtained from a library of the cultivar "Sprite," which carries the dominant allele at the I locus. We have identified a large cluster of TIR-NBS-LRR sequences associated within this locus, which extends over a distance >425 kb. Bean cultivars from the Andean or Mesoamerican gene pool that contain the dominant allele share the same haplotypes as revealed by gel blot hybridizations with a TIR probe. In contrast, beans with a recessive allele display simpler and variable haplotypes. A survey of wild accessions from Argentina to Mexico showed that this multigene family has expanded significantly during evolution and domestication. RNA gel blot analysis indicated that the TIR family of genes plays a role in the response to inoculations with BCMV or BCMNV.  相似文献

The Roseobacter group and SAR11 clade constitute high proportions of the marine bacterioplankton, but only scarce information exists on the abundance of distinct populations of either lineage. Therefore, we quantified the abundance of the largest cluster of the Roseobacter group, the RCA (Roseobacter clade affiliated) cluster together with the SAR11 clade by quantitative PCR in the southern and eastern North Sea. The RCA cluster constituted up to 15 and 21% of total bacterial 16S ribosomal RNA (rRNA) genes in September 2005 and May 2006, respectively. At a few stations, the RCA cluster exceeded the SAR11 clade, whereas at most stations, SAR11 constituted higher fractions with maxima of 37%. In most samples, only one RCA ribotype was detected. RCA abundance was positively correlated with phaeopigments, chlorophyll, dissolved and particulate organic carbon (POC), turnover rates of dissolved free amino acids (DFAAs), temperature, and negatively correlated with salinity. The SAR11 clade was only correlated with POC (negatively, May) and with DFAA turnover rates (positively, September). An abundant RCA strain, ‘Candidatus Planktomarina temperata'', was isolated from the southern North Sea. This strain has an identical 16S rRNA gene sequence to the dominant RCA ribotype. Detection of the pufM gene, coding for a subunit of the reaction center of bacteriochlorophyll a, indicates the potential of the isolate for aerobic anoxygenic photosynthesis. Our study shows that a distinct population of the RCA cluster constitutes an abundant bacterioplankton group in a neritic sea of the temperate zone and indicates that this population has an important role during decaying phytoplankton blooms.  相似文献

BackgroundStudies have found a relationship between decreased immunity and increased incidence of cancer.MethodsA systematic review of observational studies evaluating the incidence of cancer in both organ recipients and people with HIV/AIDS compared with the general population. Eligible studies were searched up to March 2011 in the following databases: Pubmed, Embase, Scielo, Cancerlit and Google scholar. In this study, the standardized incidence ratios (SIR) of cancer in people with HIV/AIDS and of organ transplant recipients were compared with those found among the general population.ResultsTwenty-five studies of transplant and HIV-associated cancer risk, involving 866 776 people with HIV/AIDS or organ recipients and 21 260 new cases of cancer, were included. The risk for the development of new cancer cases was higher among people with HIV/AIDS (SIR = 4, IC95% 3.78–4.24) and who received organs (SIR = 3.28, IC95% 3.06–3.52) when compared with the general population.ConclusionSimilar SIR in both immunocompromised populations suggests that the weakened immune system is responsible for the increased risk of new cases of cancer among these groups. Research investments are needed to develop effective cancer prevention strategies in these populations.  相似文献

Tocopherols are essential micronutrients in human and animal nutrition due to their function as lipophilic antioxidants. They are exclusively synthesized by photosynthetic organisms including higher plants. Despite the attributed beneficial health effects and many industrial applications, research on the tocopherol biosynthetic pathway and its regulation in plants is still limited. In the work presented here we performed a detailed biochemical characterization of a gamma-tocopherol methyltransferase (gamma-TMT) from Arabidopsis thaliana and of a gamma-TMT purified from Capsicum annuum fruits, a tissue with high accumulation of tocopherols. The biochemical characteristics of both enzyme preparations were remarkably similar including substrate specificities. Both enzymes converted delta- and gamma- into beta- and alpha-tocopherol, respectively, but beta-tocopherol was not accepted as a substrate, pointing to a specific methylation at the C(5)-position of the tocopherol aromatic head group. A kinetic analysis performed with the Arabidopsis enzyme was consistent with an iso-ordered bi-bi type reaction mechanism. Our results emphasize the role of gamma-TMT in regulating the spectrum of accumulated tocopherols in plants.  相似文献

The matricellular SPARC-family member hevin (Sparc-like 1/SPARCL-1/SC1/Mast9) contributes to neural development and alters tumor progression in a range of mammalian models. Based on sequence similarity, we hypothesized that proteolytic digestion of hevin would result in SPARC-like fragments (SLF) that affect the activity and/or location of these proteins. Incubation of hevin with matrix metalloproteinase-3 (MMP-3), a protease known to cleave SPARC, produced a limited number of peptides. Sequencing revealed the major proteolytic products to be SPARC-like in primary structure. In gliomas implanted into murine brain, a SLF was associated with SPARC in the neovasculature but not with hevin, the latter prominent in the astrocytes encompassed by infiltrating tumor. In this model of invasive glioma that involves MMP-3 activity, host-derived SLF was not observed in the extracellular matrix adjacent to tumor cells. In contrast, it occurred with its homolog SPARC in the angiogenic response to the tumor. We conclude that MMP-3-derived SLF is a marker of neovessels in glioma, where it could influence the activity of SPARC.  相似文献

To understand how pulse and oilseed crops might use nitrogen (N) more efficiently under varying levels of water and N availability in soil, we conducted a 2-year field study to monitor N accumulation in aboveground (AG-N) and root material at five growth stages, for canola (Brassica napus L.), mustard (Brassica juncea L.), chickpea (Cicer arietinum L.), dry pea (Pisum sativum L.) and lentil (Lens culinaris Medicum) alongside spring wheat (Triticum aestivum L.). Crops were grown in lysimeters (15 cm diameter × 100 cm deep) installed in the field in southern Saskatchewan, Canada. AG-N in all crops was greater under high-water than under low-water conditions. In oilseeds and wheat, AG-N increased until flowering then tended to level off, while in pulses it increased gradually to maturity. At maturity, dry pea and wheat had the greatest AG-N and mustard the least. Enhanced water availability increased seed N but did not affect straw N; consequently, N harvest index was greater under high-water than under low-water conditions. Root N increased until late-flowering or late-pod (dough stage in wheat) then decreased to maturity. Mustard had the lowest root N, chickpea the second lowest, and canola, wheat, dry pea, and lentil the highest. Improved water availability increased root N for oilseeds and wheat but did not affect root N in pulses. At maturity, average root N of oilseeds, pulses, and wheat was 14, 17, and 20 kg ha-1, respectively. At the seedling stage pulse crops had about 27% of total plant N in their roots, a much greater proportion than for the non-legumes. However, by maturity all crops had about 14% of plant N in their roots. Soil NO3-N increased gradually between seedling and maturity in non-legumes but in pulses there was a sharp spike at early flowering. Estimated apparent net N mineralized was similar for wheat and pulse crops which were greater than for canola and mustard. Soil N amounts and temporal change patterns varied substantially among crops evaluated, and these differences need to be considered in the development of diverse cropping systems where cereals, legumes, and oilseeds are included in rotation systems.  相似文献

Haemopathological changes attributed to heavy metal poisoning were observed in blood smears of Liza macrolepis (Smith) taken after exposures of 96 h to graded doses (mg/l) of copper (0.11–1.80), lead (1.15–18.36), and mercury (0.04–0.59), in a flow-through marin bioassay system. In general, changes in leucocytic profile appear to be correlated with pathological changes caused by increasing copper and mercury concentrations. By contrast, blood samples of mullets exposed to lead, showed significant polychromasia and + anisocytosis regardless of concentrations. The RBC count, haemoglobin content, and haemotocrit percentages were less valuable in diagnosis of copper and mercury effects.These manifestations of poisoning by trace elements bear a resemblance to the pathological changes that have been shown clinically and experimentally in mammals. Consequently, blood measurements on marine organisms may be diagnostic of undeirably high levels of copper and mercury, and so may constitute useful indicators of marine pollution.  相似文献

Glucagon-like peptide-2 (GLP-2) is a nutrient-dependent, proglucagon-derived gut hormone that shows promise for the treatment of short bowel syndrome (SBS). Our objective was to investigate how combination GLP-2 + enteral nutrients (EN) affects intestinal adaption in a rat model that mimics severe human SBS and requires parenteral nutrition (PN). Male Sprague-Dawley rats were assigned to one of five groups and maintained with PN for 18 days: total parenteral nutrition (TPN) alone, TPN + GLP-2 (100 μg·kg(-1)·day(-1)), PN + EN + GLP-2(7 days), PN + EN + GLP-2(18 days), and a nonsurgical oral reference group. Animals underwent massive distal bowel resection followed by jejunocolic anastomosis and placement of jugular catheters. Starting on postoperative day 4, rats in the EN groups were allowed ad libitum access to EN. Groups provided PN + EN + GLP-2 had their rate of PN reduced by 0.25 ml/day starting on postoperative day 6. Groups provided PN + EN + GLP-2 demonstrated significantly greater body weight gain with similar energy intake and a safe 80% reduction in PN compared with TPN ± GLP-2. Groups provided PN + EN + GLP-2 for 7 or 18 days showed similar body weight gain, residual jejunal length, and digestive capacity. Groups provided PN + EN + GLP-2 showed increased jejunal GLP-2 receptor (GLP-2R), insulin-like growth factor-I (IGF-I), and IGF-binding protein-5 (IGFBP-5) expression. Treatment with TPN + GLP-2 demonstrated increased jejunal expression of epidermal growth factor. Cessation of GLP-2 after 7 days with continued EN sustained the majority of intestinal adaption and significantly increased expression of colonic proglucagon compared with PN + EN + GLP-2 for 18 days, and increased plasma GLP-2 concentrations compared with TPN alone. In summary, EN potentiate the intestinotrophic actions of GLP-2 by improving body weight gain allowing for a safe 80% reduction in PN with increased jejunal expression of GLP-2R, IGF-I, and IGFBP-5 following distal bowel resection in the rat.  相似文献
