Deferrioxamine B and rhodotorulic acid, iron-chelating agents of microbial origin, exerted a pronounced inhibitory effect on pathogenic Naegleria fowleri at microgram levels. This inhibition was diminished by adding iron to the chelators before incubation with Naegleria isolates. These and related microbial iron chelators occur naturally in the environment. This could be of considerable ecological significance and provides a novel hypothesis to account for the proliferation of pathogenic Naegleria spp. in certain aquatic habitats. 相似文献

Mesenchymal and hematopoietic tissues are important reservoirs of adult stem cells. The potential of tissue resident mesenchymal stem cells (MSCs) to differentiate into cells of mesodermal and ectodermal lineages has been reported previously. We examined the hypothesis that adherent adipose tissue resident mesenchymal stem cells (ASCs) are capable of generating cells with hematopoietic characteristics. When cultured in differentiation media, clonally isolated ASCs develop into cells with hematopoietic attributes. The hematopoietic differentiated cells (HD) express early hematopoietic (c‐kit, PROM1, CD4) as well as monocyte/macrophage markers (CCR5, CD68, MRC1, CD11b, CSF1R). Additionally, HD cells display functional characteristics of monocyte/macrophages such as phagocytosis and enzymatic activity of α‐Naphthyl Acetate Esterase. HD cells are also responsive to stimulation by IL‐4 and LPS as shown by increased CD14 and HLA‐DRB1 expressions and release of IL‐2, IL10, and TNF. Taken together, this study characterizes the potential of ASCs to generate functional macrophages in vitro, and therefore paves way for their possible use in cell therapy applications. J. Cell. Recombinant tissue-type plasminogen activator (rt-PA) from cultures of a genetically manipulated Bowes melanoma cell line (TRBM6) was purified in batches of average volume 451 using an autoclavable, reusable, continuous chromatography system comprising zinc chelate-Sepharose CL4B and lysine-Sepharose CL4B. After eight successive purifications the rt-PA was ultrafiltered to yield a preparation containing 4.9 mg protein/ml and 2.7 X 10(6) IU/ml. Analysis by SDS-polyacrylamide gel electrophoresis followed by staining with Coomassie brilliant blue R250 showed major protein bands at Mr = 63,000 and 65,000; most of the material was in the 1-chain form. The potential usefulness of a simple, rapid continuous chromatography system that can be operated under aseptic conditions is discussed. 相似文献

Trio combines with dock to regulate Pak activity during photoreceptor axon pathfinding in Drosophila
Correct pathfinding by Drosophila photoreceptor axons requires recruitment of p21-activated kinase (Pak) to the membrane by the SH2-SH3 adaptor Dock. Here, we identify the guanine nucleotide exchange factor (GEF) Trio as another essential component in photoreceptor axon guidance. Regulated exchange activity of one of the two Trio GEF domains is critical for accurate pathfinding. This GEF domain activates Rac, which in turn activates Pak. Mutations in trio result in projection defects similar to those observed in both Pak and dock mutants, and trio interacts genetically with Rac, Pak, and dock. These data define a signaling pathway from Trio to Rac to Pak that links guidance receptors to the growth cone cytoskeleton. We propose that distinct signals transduced via Trio and Dock act combinatorially to activate Pak in spatially restricted domains within the growth cone, thereby controlling the direction of axon extension. 相似文献

A rat liver model has been developed for studying preservation by continuous hypothermic perfusion. Perfusion for 5 and 24 hr with a defined solution containing Haemaccel as colloid at a temperature of 5–7 °C was found to result in good preservation of the metabolic activity of the livers as assessed by the ability of the tissue to synthesize urea and take up galactose. In common with other continuous perfusion methods, there was increasing evidence of cell damage as perfusion progressed, and there was an associated depletion of cellular K+ and an increase in tissue water content. There was a considerable increase in vascular resistance, as indicated by a fall in flow rate at a constant pressure in the later stages of perfusion, similar to that which has been reported in other liver preservation experiments using plasma derivatives or Dextran. Further studies are envisaged to attempt to analyze, in a controlled fashion, the nature of the damage incurred by the liver during storage and the requirements for maintenance of a viable graft at hypothermia. 相似文献

We examine the utility of stable carbon and nitrogen isotope variations to characterize the length of the nursing/lactation period and age at weaning for two northern Pacific otariid species, the northern fur seal (Callorhinus ursinus) and California sea lion (Zalophus californianus). We used two sampling strategies to measure ontogenetic trends in isotope value, and compared our results to observational data on the reproductive strategies used by these otariids. For Zalophus, we found evidence for 15N enrichment and 13C‐depletion in bone collagen representing the first and second year of growth, which is consistent with the ~12–14‐mo weaning age in this population after a suitable turnover rate for bone collagen is considered. Analysis of individual tooth annuli from a different suite of Zalophus specimens suggests that half of the individuals were weaned at ~12 mo of age, and half were dependent on milk for a portion of their second year. For Callorhinus, bone collagen for age classes that contain pre‐weaned individuals were 15N‐enriched, but values were significantly lower in specimens between 6 and 20 mo of age. These 15N‐enriched values, presumably acquired during nursing between 0 and 4 mo of age in Callorhinus, were not present in specimens older than 12 mo of age. Thus complete bone collagen turnover in young‐of‐the‐year occurs in 8–10 mo. 15N enrichment is evident in the first annulus of female Callorhinus individuals, but is not detectable in males. Analyses of Callorhinus tooth annuli show no ontogenetic trends in δ13C values. Our study indicates that nitrogen, and in some cases carbon, isotopes can be used to assess reproductive strategies in marine mammals. When coupled with accurate age estimates based on bone growth regressions, this isotopic technique can be applied to historical or fossil otariids to gain insight into the flexibility of maternal strategies within and across species. 相似文献

Background
The definition of "clinical asthma remission" is based on absence of symptoms and use of medication. However, in the majority of these subjects airway inflammation is still present when measured. In the present study we investigated whether "complete asthma remission", additionally defined by the absence of bronchial hyperresponsiveness (BHR) and the presence of a normal lung function, is associated with the absence of airway inflammation.Methods
Patients with a former diagnosis of asthma and a positive histamine provocation test were re-examined to identify subjects with complete asthma remission (no asthma symptoms or medication, PC20 histamine > 32 mg/ml, FEV1 > 90% predicted). Patients with PC20 histamine ≤ 32 mg/ml were defined as current asthmatics and were divided in two groups, i.e. asthmatics with and without BHR to adenosine 5'monophoshate (AMP). Sputum induction was performed 1 week before and 1 hour after AMP provocation. Sputum induction and AMP provocation were previously shown to be sensitive markers of airway inflammation.Results
Seven patients met criteria for complete asthma remission. Twenty-three were current asthmatics, including twelve without hyperresponsiveness to AMP. Subjects with complete asthma remission showed no AMP-induced sputum eosinophilia (median (range) 0.2 (0 - 4.6)% at baseline and 0.2 (0 - 2.6)% after AMP). After AMP, current asthmatics had a significant increase in sputum eosinophils (0.5 (0 - 26.0)% at baseline and 2.6 (0 - 32.0) % after AMP), as had the subgroup of current asthmatics without hyperresponsiveness to AMP (0.2 (0 - 1.8)% at baseline and 1.3 (0 - 6.3)% after AMP).Conclusions
Subjects with complete asthma remission, in contrast to subjects with current asthma, do not respond with eosinophilic inflammation in sputum after AMP provocations. These data lend support to the usefulness of the definition of complete asthma remission. 相似文献49.

Visitors place a complex array of demands on conservation reserves, including provisions for recreation. Rising recreation demand includes a new suite of activities ranging from adventure racing, music events, and motorised activities to extreme sports. Policy implications raise questions anew: what is the fundamental purpose of conservation reserves—nature conservation or recreation or both, and where should the emphasis lie? There is a risk that the current and future emphasis appears to be on increased commercialisation, marketing of conservation reserves as music and/or sporting event venues, places where personal physical challenges can be undertaken alongside a mentality that celebrates human achievement rather than the appreciation of nature! Such a trend may de-emphasise visitor perceptions of conservation reserves as tools for nature conservation. The reported trend in recreational activity requires debate, policy direction and target areas need protected area management effectiveness evaluation to assess conservation implications. 相似文献
