Age-related changes in ovarian development characteristics and plasma sex steroids in female Murray cod were examined throughout their second, third and fourth years of life to better understand the physiological and endocrine processes associated with puberty in this species in captivity. Spawning performance of 2+ and 3+ year old females was also assessed to identify ontogenetic differences in egg fertility. Puberty was acquired in 38% of 1+ year old females and 100% of age 2+ females. By age 3+, all females had developed full (adult) reproductive function. Ovarian development in pubertal fish was characterised by a rapid transition between cortical alveoli and lipid droplet oogenic phases, coinciding with significantly lower plasma 17β-oestradiol in age 2+ females (p < 0.05). Mean mature oocyte diameter (2.44 mm), post-fertilisation viability (30.80%) and hatchability (0.99%) of eggs from age 2+ females were significantly reduced relative to age 3+ adults (2.81 mm, 84.89% and 23.58%, respectively). Ovaries of pubertal Murray cod exhibited both vitellogenic and ovulatory capacities, yet functional abnormalities during secondary oocyte growth are likely to have contributed to poor egg fertility and consequently, evaluations of age-at-first maturity based on the presence of advanced ovarian stages may overestimate the reproductive potential of younger broodstock populations.  相似文献

Lipopolysaccharides, the ubiquitous part of the outer membrane of Gram-negative bacteria, and their derivatives are recognised by plants to trigger or potentiate particular defence responses such as induction of genes encoding pathogenesis-related proteins. The molecular mechanisms of LPS perception that underpin these effects in plants are, however, unknown. Here, lipid A from Halomonas magadiensis, which is an antagonist of lipid A action in human cells, was used to investigate lipid A action in plants. Our findings offer an insight into the different structural requirements for direct induction and potentiation of plant defences by lipid A.  相似文献

Are setts significant determinants of badger socio‐spatial organisation, and do suitable sett sites represent a limited resource, potentially affecting badger distributions? The factors determining diurnal resting den, or sett, location and selection by Eurasian badgers Meles meles L. were investigated in Wytham Woods, Oxfordshire. 279 sett sites were located. The habitat parameters that were associated with the siting of these setts were analysed and associations were sought between sett location and character and the body condition and body weight of resident badgers Habitat characteristics in the vicinity of setts were significantly different from randomly selected points. Badgers preferentially selected sites with sandy, well‐drained soils, situated on NW‐facing, convex and moderately inclined slopes at moderate altitude. There was no evidence that sett morphology (number of entrances, sett area, number of hinterland latrines) was affected by the surrounding sett site habitat characteristics. Mean body weight was significantly higher for badgers occupying territories with setts in sandy soils, situated on NW‐facing slopes, than in territories with less optimal sett characteristics. Contrary to the hypothesis that the availability of sett sites was limiting, and therefore that sett dispersion dictates the spatial and social organisation of their populations, the badgers were clearly able to excavate new setts. On our measures, these new setts were not inferior to old established ones, despite occupying subsequently exploited sites; the badgers utilising these new setts had neither lighter body weights nor poorer body condition scores. During the period of our study badgers have manifestly been able to dig numerous new setts; as satisfactory sites still remain available, this indicates that suitable sett sites have not yet become a limiting resource. There was no relationship between sett age and the characteristics of the site in which it was dug, as suitable sites were not limiting. Significantly, population expansion during the decade 1987–1997 was not constrained by lack of setts, rather the main proliferation in setts occurred after the population size had peaked in 1996. Some implications for the management and conservation of the Eurasian badger are considered.  相似文献

Calmodulin (CaM) is the primary transducer of calcium fluxes in eukaryotic cells. Its two domains allosterically regulate myriad target proteins through calcium-linked association and conformational change. Many of these proteins have a basic amphipathic alpha-helix (BAA) motif that binds one or both CaM domains. Previously, we demonstrated domain-specific binding of melittin, a model BAA peptide, to Paramecium CaM (PCaM): C-domain mutations altered the interaction with melittin, whereas N-domain mutations had no discernable effect. Here, we report on the use of fluorescence and NMR spectroscopy to measure the domain-specific association of melittin with calcium-saturated ((Ca(2+))(4)-PCaM) or calcium-depleted (apo) PCaM, which has enabled us to determine the free energies of calcium binding to the PCaM-melittin complex, and to estimate interdomain cooperativity. Under apo conditions, melittin associated with each PCaM domain fragment (PCaM(1-80) and PCaM(76-148)), as well as with the C-domain of full-length PCaM (PCaM(1-148)). In the presence of calcium, all of these interactions were again observed, in addition to which an association with the N-domain of (Ca(2+))(4)-PCaM(1-148) occurred. This new association was made possible by the fact that melittin changed the calcium-binding preferences for the domains from sequential (C > N) to concomitant, decreasing the median ligand activity of calcium toward the N-domain 10-fold more than that observed for the C-domain. This selectivity may be explained by a free energy of cooperativity of -3 kcal/mol between the N- and C-domains. This study demonstrates multiple domain-selective differences in the interactions between melittin and PCaM. Our findings support a model that may apply more generally to ion channels that associate with the C-domain of CaM under low (resting) calcium conditions, but rearrange when calcium binding triggers an association of the N- domain with the channel.  相似文献

Human parainfluenza virus type 3 (HPIV3) is a major respiratory pathogen in humans. Failure to induce immunological memory associated with HPIV3 infection has been attributed to inhibition of lymphocyte proliferation. We demonstrate that the inability of mixed lymphocytes to respond to virally infected antigen-presenting cells is due to an interleukin-2-dependent, nonapoptotic mechanism involving natural killer (NK) cells and their influence is exerted in a contact-dependent manner. These results suggest a novel regulatory mechanism for NK cells during HPIV3 infection, offering an explanation for viral persistence and poor memory responses.  相似文献

TRIM5alpha is a potent barrier to cross-species retroviral transmission, and TRIM5alphas from different species have divergent antiretroviral specificities. Multiple TRIM5 alleles circulate within rhesus macaque populations. Here we show that they too have different antiretroviral specificities, highlighting how TRIM5 genotypes contribute to protection in an individual or a population.  相似文献

The binding problem—question of how information between the modules of the linguistic system is integrated during language processing—is as yet unresolved. The remarkable speed of language processing and comprehension (Pulvermüller et al. 2009) suggests that at least coarse semantic information (e.g. noun animacy) and syntactically-relevant information (e.g. verbal template) are integrated rapidly to allow for coarse comprehension. This EEG study investigated syntax–semantics interface processing during word-by-word sentence reading. As alpha-band neural activity serves as an inhibition mechanism for local networks, we used topographical distribution of alpha power to help identify the timecourse of the binding process. We manipulated the syntactic parameter of verbal event structure, and semantic parameter of noun animacy in reduced relative clauses (RRCs, e.g. “The witness/mansion seized/protected by the agent was in danger”), to investigate the neural bases of interaction between syntactic and semantic networks during sentence processing. The word-by-word stimulus presentation method in the present experiment required manipulation of both syntactic structure and semantic features in the working memory. The results demonstrated a gradient distribution of early components (biphasic posterior P1–N2 and anterior N1–P2) over function words “by” and “the”, and the verb, corresponding to facilitation or conflict resulting from the syntactic (telicity) and semantic (animacy) cues in the preceding portion of the sentence. This was followed by assimilation of power distribution in the α band at the second noun. The flattened distribution of α power during the mental manipulation with high demand on working memory—thematic role re-assignment—demonstrates a state of α equilibrium with strong functional coupling between posterior and anterior regions. These results demonstrate that the processing of semantic and syntactic features during sentence comprehension proceeds in highly integrated fashion using gating of attentional resources to facilitate rapid comprehension, with attentional suppression of global alpha power to facilitate interaction of local networks.  相似文献
