Evidence is presented for two distinct CA2+ pools in amoebae of Dictyostelium discoideum. One pool, presumably mitochondrial, was sensitive to the mitochondrial inhibitors oligomycin and dinitrophenol and showed an affinity for Ca2+ in the micro M concentration range. The other Ca2+ pool, which was insensitive to these inhibitors, was of lower capacity but had higher affinity (in the nM range). Inositol 1,4,5-trisphosphate (5 micro M) added to saponin-permeabilized amoebae induced a rapid release of Ca2+ from the latter pool but had no effect on the presumed mitochondrial pool. Controls using addition of inositol 1,4-bisphosphate (the hydrolytic product of IP3) induced no such CA2+ release. The results provide strong support for the involvement of IP3 in signal transmission during chemotaxis of D. discoideum.  相似文献

Point counts are commonly used to assess changes in bird abundance, including analytical approaches such as distance sampling that estimate density. Point‐count methods have come under increasing scrutiny because effects of detection probability and field error are difficult to quantify. For seven forest songbirds, we compared fixed‐radii counts (50 m and 100 m) and density estimates obtained from distance sampling to known numbers of birds determined by territory mapping. We applied point‐count analytic approaches to a typical forest management question and compared results to those obtained by territory mapping. We used a before–after control impact (BACI) analysis with a data set collected across seven study areas in the central Appalachians from 2006 to 2010. Using a 50‐m fixed radius, variance in error was at least 1.5 times that of the other methods, whereas a 100‐m fixed radius underestimated actual density by >3 territories per 10 ha for the most abundant species. Distance sampling improved accuracy and precision compared to fixed‐radius counts, although estimates were affected by birds counted outside 10‐ha units. In the BACI analysis, territory mapping detected an overall treatment effect for five of the seven species, and effects were generally consistent each year. In contrast, all point‐count methods failed to detect two treatment effects due to variance and error in annual estimates. Overall, our results highlight the need for adequate sample sizes to reduce variance, and skilled observers to reduce the level of error in point‐count data. Ultimately, the advantages and disadvantages of different survey methods should be considered in the context of overall study design and objectives, allowing for trade‐offs among effort, accuracy, and power to detect treatment effects.  相似文献

HIV-1 Rev and the Rev response element (RRE) enable a critical step in the viral replication cycle by facilitating the nuclear export of intron-containing mRNAs, yet their activities have rarely been analyzed in natural infections. This study characterized their genetic and functional variation in a small cohort of HIV-infected individuals. Multiple Rev and RRE sequences were obtained using single-genome sequencing (SGS) of plasma samples collected within 6 months after seroconversion and at a later time. This allowed the identification of cognate sequences that were linked in vivo in the same viral genome and acted together as a functional unit. Phylogenetic analyses of these sequences indicated that 4/5 infections were founded by a single transmission event. Rev and RRE variants from each time point were subjected to functional analysis as both cognate pairs and as individual components. While a range of Rev-RRE activities were seen, the activity of cognate pairs from a single time point clustered to a discrete level, which was termed the set point. In 3/5 patients, this set point changed significantly over the time period studied. In all patients, RRE activity was more sensitive to sequence variation than Rev activity and acted as the primary driver of the cognate set point. Selected patient RREs were also shown to have differences in Rev multimerization using gel shift binding assays. Thus, rather than acting as a simple on-off switch or maintaining a constant level of activity throughout infection, the Rev-RRE system can fluctuate, presumably to control replication.  相似文献

Sarracenia purpurea L. (northern pitcher plant) is an insectivorous plant with extrafloral nectar that attracts insects to a water-filled pitfall trap. We identified and quantified the amino acids in extrafloral nectar produced by pitchers of S. purpurea. Nectar samples were collected from 32 pitchers using a wick-sampling technique. Samples were analyzed for amino acids with reverse-phase high-performance liquid chromatography with phenylisothiocyanate derivatization. Detectable amounts of amino acids were found in each of the 32 nectar samples tested. Mean number of amino acids in a nectar sample was 9 (SD = 2.2). No amino acid was detected in all 32 samples. Mean amount of amino acids in a nectar sample (i.e., amount per wick) was 351.4 ng (SD = 113.2). Nine amino acids occurred in 20 of the 32 samples (aspartic acid, cysteine, glutamic acid, glycine, histidine, hydroxyproline, methionine, serine, valine) averaging 263.4 ng (SD = 94.9), and accounting for ~75% of the total amino acid content. Nectar production may constitute a significant cost of carnivory since the nectar contains amino acids. However, some insects prefer nectar with amino acids and presence of amino acids may increase visitation and capture of insect prey.  相似文献

Knowledge of the crop-size and vertical profile with respect to sea level of dead leaf blades and other shoot parts of saltmarsh grasses is a prerequisite for projections of seawater impact upon saltmarsh grass-shoot decomposers. We measured seasonal changes in quantities of leaf blade and leaf sheath+stem of smooth cordgrass in southern temperate saltmarshes of three types: low- (LD), mid- (MD), and high-drainage (HD) marshes (i.e., with low to high densities of drainage creeks). In each type of marsh, we took samples in the three characteristic subsites, short-shoot, intermediate-height, and tall-shoot. Wholly dead (brown) and living (some green) shoots were counted and their canopy heights measured, and the mass of all shoots was measured in 10-cm height increments (above sediment), with separation of living and dead parts. Detached material was also quantified, separated into that trapped in the canopy, and that prostrate on the sediment. Extent of marsh drainage did not affect the density of wholly dead shoots, which ranged from an average of 51 m−2 in autumn to 103 m−2 in spring, and 57 m−2 in tall subsites to 89–96 m−2 in short and intermediate subsites. The top of the dead-shoot canopy was about 20 cm taller than the living-shoot canopy in winter/spring, but was below it ( ≈50 cm) in summer/autumn. The mass of standing-dead leaf blades was 4- to 5-fold greater in winter than in summer, and it was about 2.5-fold greater at tall than at short sites. Detached litter trapped among shoots above the sediment averaged 26% of total dead-shoot mass, but litter lying on the sediment averaged only 6% of total dead-shoot mass. Since export of macrodetritus from marshgrass stands is inconsequential, the small prostrate-litter mass implies that breakdown of shoots to small particles takes place within the vegetated marsh, and that standing decay predisposes fallen material to rapid disintegration. Comparison of heights of usual tidal contact on shoots at the short- and tall-shoot subsites showed that projected impacts of seawater upon leaf-blade decay would be greater at short-shoot subsites (high marsh) than at tall-shoot subsites (low marsh). This was because the vertical profile of dead-blade mass resulted in more of the short-shoot dead-blade mass residing within the vertical span most regularly contacted by flooding tides.  相似文献

Hirschsprung disease is a congenital malformation affecting 1 in 5000 live births. The absence of parasympathetic neuronal ganglia (Meissner, Auerbach) in the hindgut results in poor coordination of peristaltic movement, and a varying degree of constipation. Four different genes have been implicated in the pathogenesis of Hirschsprung disease: the RET tyrosine kinase receptor gene; one of its ligands, the glial cell line-derived neurotrophic factor (GDNF) gene; the endothelin receptor B (EDNRB) gene; and its ligand, endothelin-3 (EDN3). Recently, combinations of mutations in two of these genes (RET and GDNF) have been reported in Hirschsprung patients. We report a family with missense mutations in both the RET gene (R982C) and the EDNRB gene (G57S). In this family, three out of five members have the two mutations, but only one, a boy, has the Hirschsprung disease phenotype. A total of 235 accessions of Tripsacum dactyloides (L.) L. representing natural populations from 88 locations distributed throughout ten states of the United States were collected in the wild and studied morphologically and cytologically. Multivariate analysis of variance (MANOVA) was applied to 17 morphological characters recorded for 868 specimens from the 88 locations, which were further combined into 25 groups to facilitate analysis. Multivariate F ratios for six comparisons involving different groups were highly significant, indicating that the populations were significantly different on the basis of all 17 characters. Non-significant MANOVA F ratios for two comparisons between sympatric diploid and polyploid accessions showed them to be morphologically similar. Discriminant function (DF) analysis was employed to determine which characters contributed towards group discrimination. No one variable was found to distinguish between groups; rather, a combination of characters was necessary, and the combination changed with each comparison. Accessions were mainly either diploid (2n = 36) or tetraploid (2n = 72), with two triploids (2n = 54) reported from separate locations in Texas. Cytological data revealed that diploids and tetraploids occupy a more sympatric distribution than previously supposed. Morphological analysis suggested that diploids could not always be distinguished from polyploids on the basis of morphology alone, unless the appropriate characters were used. Information from cytological and morphological data showed Tripsacum dactyloides to be composed of many populations covering a wide range of variability, with each population possessing a characteristic combination of morphological attributes. It was concluded that T. dactyloides can be most meaningfully analyzed in terms of particular geographic regions, rather than attempting to elucidate natural relationships by correlating data from the entire United States.  相似文献

In Salmonella typhimurium the two enzymes of proline catabolism, proline oxidase and Delta(1)-pyrroline-5-carboxylic acid dehydrogenase, are subject to catabolite repression when the cells are grown in the presence of glucose. Mutants partially relieved of catabolite repression (PutR) for the proline catabolic enzymes have been isolated by selection on agar plates containing glucose and proline. The specificity of the catabolite repression-insensitive character for the enzymes of proline utilization has been confirmed by an analysis of other unrelated catabolic enzymes. Histidase and amylomaltase of the mutant strains are equally as sensitive to glucose repression as are the enzymes from the wild type. All four PutR mutants exhibit higher induced and higher basal levels of proline oxidase as compared with the corresponding wild-type levels. The mutations of three strains tested are cotransducible with constitutive, pleiotrophic-negative and structural gene mutations of the put region. Three-factor crosses indicate that two putR mutations are located at one end of the cluster of put mutations.  相似文献
