Background  

Lyme disease, caused by Borrelia burgdorferi, affects a large number of people in both the USA and Europe. The mouse is a natural host for this spirochete and is widely used as a model system to study Lyme pathogenesis mechanisms. Since disease manifestations often depend upon the spirochete burden in a particular tissue, it is critical to accurately measure the bacterial number in infected tissues. The current methods either lack sensitivity and specificity (SYBR Green), or require independent analysis of samples in parallel to quantitate host and bacterial DNA (TaqMan). We have developed a novel molecular beacon-based convenient multiplex real-time quantitative PCR assay to identify and detect small numbers of B. burgdorferi in infected mouse tissues.  相似文献

Background

Historically, counting influenza recorded in administrative health outcome databases has been considered insufficient to estimate influenza attributable morbidity and mortality in populations. We used database record linkage to evaluate whether modern databases have similar limitations.

Methods

Person-level records were linked across databases of laboratory notified influenza, emergency department (ED) presentations, hospital admissions and death registrations, from the population (∼6.9 million) of New South Wales (NSW), Australia, 2005 to 2008.

Results

There were 2568 virologically diagnosed influenza infections notified. Among those, 25% of 40 who died, 49% of 1451 with a hospital admission and 7% of 1742 with an ED presentation had influenza recorded on the respective database record. Compared with persons aged ≥65 years and residents of regional and remote areas, respectively, children and residents of major cities were more likely to have influenza coded on their admission record. Compared with older persons and admitted patients, respectively, working age persons and non-admitted persons were more likely to have influenza coded on their ED record. On both ED and admission records, persons with influenza type A infection were more likely than those with type B infection to have influenza coded. Among death registrations, hospital admissions and ED presentations with influenza recorded as a cause of illness, 15%, 28% and 1.4%, respectively, also had laboratory notified influenza. Time trends in counts of influenza recorded on the ED, admission and death databases reflected the trend in counts of virologically diagnosed influenza.

Conclusions

A minority of the death, hospital admission and ED records for persons with a virologically diagnosed influenza infection identified influenza as a cause of illness. Few database records with influenza recorded as a cause had laboratory confirmation. The databases have limited value for estimating incidence of influenza outcomes, but can be used for monitoring variation in incidence over time.  相似文献

Current information on the structure and function of motile tubular vacuoles in Pisolithus tinctorius and other fungi is reviewed. The use of fluorochromes to label the vacuole lumen is evaluated and observations on the structure and motility of vacuoles in P. tinctorius are differentiated from possible artifacts. The styryl dyes FM4-64 and MDY-64, used in yeast to demonstrate endocytosis, show little or no labeling of internal membranes in undamaged P. tinctorius cells. This agrees with our data showing that other probes for endocytosis such as Lucifer yellow CH are not taken up by hyphal tip cells. Overall, the observations do not support endocytosis in hyphal tips. It has been suggested that tubular vacuole systems carry out longitudinal transport, and evidence in favor of this hypothesis is evaluated. New data are presented to show that many of the large vacuoles in subapical cells are attached to the plasma membrane and are relatively immobile, while video sequences show movement of fluorochrome in pulses along a series of several large vacuoles, all interconnected via tubules. Tubular vacuoles from thick sections of hyphae processed under anhydrous conditions are shown by X-ray microanalysis to contain relatively high levels of P and K, as seen previously in the larger vacuoles. This paper presents the approach and methods used, and the outputs generated, in the derivation of numeric biological objectives for the protection of rivers and streams across Victoria, Australia. Biological objectives have been designed for inclusion in State government legislation for the protection of the State’s waterways. The objectives were developed using data from extensive macroinvertebrate sampling of multiple reference sites across the State. Regional variations are incorporated into the objective-setting process by using an a priori, macroinvertebrate-based regionalisation of the State. The objectives consist of four separate components, including a measure of diversity, two biotic indices and a measure of community composition. The objectives are designed for use across broad regions and any attempt to extend the system to subregional levels would require a careful consideration of the influence of scale upon the grouping of reference sites.  相似文献

Response of epilithic diatom assemblages to urbanization influences   总被引:2,自引:1,他引:1  
Urbanization has long been recognized to alter the hydrology, water quality and channel form of waterways. Recently, consideration of urban impacts on waterways has expanded to include assessment of the aquatic biota, generally focusing upon the macroinvertebrate fauna. This study compares the impacts of urbanization on the structure of stream benthic diatom communities in 16 first- and second-order streams in the east of Melbourne, Australia. Relationships between the physical elements of urbanization, water quality and diatom communities were examined using multivariate analyses with compositional similarity, and univariate analyses with selected diatom species and indices,. There was a strong negative correlation between urban density and the diatom indices of water quality. The element of urbanization most strongly related to the measured decline in the diatom community was drainage connection. Electrical conductivity was the most influential water quality variable. It is hypothesized that, in addition to increasing electrical conductivity, drainage connection may be impacting the diatom community by increasing the delivery of phosphorus during small storm events. The study suggests that reducing directly piped drainage connection using infiltration and retention is a logical step in the mitigation of urban impacts upon receiving streams. A corollary part of the project indicated strong potential for using overseas diatom indices in studies on water quality in southeastern Australia.  相似文献
