When fed to starved adults of Anthonomus grandis, several pentoses and hexoses were metabolized to the corresponding polyols (sugar alcohols). Xylitol, galactitol, arabitol, ribitol, rhamnitol, mannitol, and sorbitol were metabolites of d-xylose, d-galactose and lactose, d-arabinose, d-ribose, l-rhamnose, d-mannose, and d-glucose and d-fructose, respectively. l-Sorbose was not metabolized to a polyol. Large quantities of xylitol and galactitol and intermediate amounts of arabitol, ribitol, and rhamnitol accumulated while only small amounts or traces of mannitol and sorbitol were detected. The limited accumulation of sorbitol in the glucose- and fructose-fed weevils probably was caused by the rapid metabolism of sorbitol to glucose, fructose, trehalose, and glycogen. Each of the ingested sugars, the corresponding polyols, and trehalose were present in the weevil haemolymph. Most of the polyols had never before been detected as metabolites in an insect.  相似文献

From 1981 through 1986, plasma or serum samples were obtained from 322 wild turkeys (Meleagris gallopavo) from Georgia (n = 111), Kentucky (n = 21), Louisiana (n = 22), North Carolina (n = 118), Tennessee (n = 19), Missouri (n = 24) and Iowa (n = 7). These samples were tested for antibodies to Mycoplasma gallisepticum (MG) and in most instances, M. synoviae (MS), M. meleagridis (MM), and avian influenza (AI) virus. All 322 turkeys were seronegative for MG by the rapid plate agglutination (RPA) test. All of a subsample (n = 147) also were negative (titer less than or equal to 1:40) for MG by the hemagglutination inhibition (HI) test. Five of 253 turkeys (2%) were seropositive (+4 reaction) for MS by the RPA test; however, HI tests for MS on these five turkeys were negative as were attempts to isolate MS from trachea and homogenized lung tissue. Three of 253 turkeys (1%) were seropositive (+1 to +3 reactions) for MM by the RPA test. None of 210 turkeys had antibodies to AI by the agar gel precipitation test. These data suggest that populations of native eastern wild turkeys are not important in the epizootiology of MG, MS, MM, or AI.  相似文献

In situ gelation of injectable polypeptide-based materials is attractive for minimally invasive in vivo implantation of biomaterials and tissue engineering scaffolds. We demonstrate that chemically cross-linked elastin-like polypeptide (ELP) hydrogels can be rapidly formed in aqueous solution by reacting lysine-containing ELPs with an organophosphorous cross-linker, beta-[tris(hydroxymethyl)phosphino]propionic acid (THPP) under physiological conditions. The mechanical properties of the cross-linked ELP hydrogels were largely modulated by the molar concentration of lysine residues in the ELP and the pH at which the cross-linking reaction was carried out. Fibroblasts embedded in ELP hydrogels survived the cross-linking process and were viable after in vitro culture for 3 days. DNA quantification of ELP hydrogels with encapsulated fibroblasts indicated that there was no significant difference in DNA content between day 0 and day 3 when ELP hydrogels were formed with an equimolar ratio of THPP and lysine residues of the ELPs. These results suggest that THPP cross-linking may be a biocompatible strategy for the in situ formation of cross-linked hydrogels.  相似文献

Tubulin is an important molecular target in cancer chemotherapy. Antimitotic agents able to bind to the protein are currently under study, commonly used in the clinic to treat a variety of cancers and/or exploited as probes to investigate the protein’s structure and function. Here we report the binding modes for a series of colchicinoids, combretastatin A4 and chalcones established from docking studies carried out on the structure of tubulin in complex with colchicine. The proposed models, in agreement with published biochemical data, show that combretastatin A4 binds to the colchicine site of β-tubulin and that chalcones assume an orientation similar to that of podophyllotoxin. The models can be used to design a new class of podophyllotoxin mimics, the α-aryl chalcones, capable of binding to the colchicine-binding site of β-tubulin with higher affinity.  相似文献

The International Study of Asthma and Allergies in Childhood (ISAAC) Phase One showed large worldwide variations in the prevalence of symptoms of asthma, rhinoconjunctivitis and eczema, up to 10 to 20 fold between countries. Ecological analyses were undertaken with ISAAC Phase One data to explore factors that may have contributed to these variations, and are summarised and reviewed here.In ISAAC Phase One the prevalence of symptoms in the past 12 months of asthma, rhinoconjunctivitis and eczema were estimated from studies in 463,801 children aged 13 - 14 years in 155 centres in 56 countries, and in 257,800 children aged 6-7 years in 91 centres in 38 countries. Ecological analyses were undertaken between symptom prevalence and the following: Gross National Product per capita (GNP), food intake, immunisation rates, tuberculosis notifications, climatic factors, tobacco consumption, pollen, antibiotic sales, paracetamol sales, and outdoor air pollution.Symptom prevalence of all three conditions was positively associated with GNP, trans fatty acids, paracetamol, and women smoking, and inversely associated with food of plant origin, pollen, immunisations, tuberculosis notifications, air pollution, and men smoking. The magnitude of these associations was small, but consistent in direction between conditions. There were mixed associations of climate and antibiotic sales with symptom prevalence.The potential causality of these associations warrant further investigation. Factors which prevent the development of these conditions, or where there is an absence of a positive correlation at a population level may be as important from the policy viewpoint as a focus on the positive risk factors. Interventions based on small associations may have the potential for a large public health benefit.  相似文献

Biodendrimer-based hydrogel scaffolds for cartilage tissue repair   总被引:2,自引:0,他引:2  
Photo-crosslinkable dendritic macromolecules are attractive materials for the preparation of cartilage tissue engineering scaffolds that may be optimized for in situ formation of hydrated, mechanically stable, and well-integrated hydrogel scaffolds supporting chondrocytes and chondrogenesis. We designed and synthesized a novel hydrogel scaffold for cartilage repair, based on a multivalent and water-soluble tri-block copolymer consisting of a poly(ethylene glycol) core and methacrylated poly(glycerol succinic acid) dendrimer terminal blocks. The terminal methacrylates allow mild and biocompatible photo-crosslinking with a visible light, facilitating in vivo filling of irregularly shaped defects with the dendrimer-based scaffold. The multivalent dendrimer constituents allow high crosslink densities that inhibit swelling after crosslinking while simultaneously introducing biodegradation sites. The mechanical properties and water content of the hydrogel can easily be tuned by changing the biodendrimer concentration. In vitro chondrocyte encapsulation studies demonstrate significant synthesis of neocartilaginous material, containing proteoglycans and type II collagen.  相似文献

Based on the symmetrical bidentate structure of the NS5A inhibitor BMS-790052, a series of new monodentate molecules were designed. The synthesis of 36 new non-dimeric NS5A inhibitors is reported along with their ability to block HCV replication in an HCV 1b replicon system. Intercropping switchgrass (Panicum virgatum L.) with loblolly pine (Pinus taeda L.) has been proposed for producing bioenergy feedstock in the southeastern United States. This study investigated switchgrass growth and pine–switchgrass interactions at two established experimental fields (7‐year‐old Lenoir site and 5‐year‐old Carteret site) located on the coastal plain of eastern United States. Position effects (edge and center of switchgrass alley in intercropping plots) and treatment effects (intercropping vs. grass‐only) on aboveground switchgrass growth were evaluated. Interspecific interactions with respect to capturing resources (light, soil water, and nitrogen) were investigated by measuring photosynthetically active radiation (PAR) above grass canopy, soil moisture, and soil mineral nitrogen contents. Switchgrass growth was significantly (P = 0.001) affected by treatments in Lenoir and by position (P < 0.0001) in both study sites. Relative to the center, PAR above grass canopy at edge in both sites was about 48% less during the growing season. Soil water content during the growing season at the edge of grass alley was significantly (P = 0.0001) lower by 23% than at the center in Lenoir, while no significant (P = 0.42) difference was observed in Carteret, in spite of more grass growth at center at both sites. Soil mineral nitrogen content at the center of intercropping plots in Lenoir (no fertilization during 2015) was significantly (P < 0.07) lower than at the edge during the peak of growing season (June, July, and August), but not during early and late parts of growing season (May, September, and November). Position effects on soil water and mineral nitrogen were less evident under conditions with higher external inputs (rainfall and fertilization) and lower plant uptake during nongrowing seasons. Results from this study contributed to a better understanding of above‐ and belowground pine–switchgrass interactions which is necessary to properly manage this new cultivation system for bioenergy production in the southeastern United States.  相似文献
