Human 125I-labelled HDL3 is degraded by isolated rat intestinal mucosal cells. In our experimental conditions, lipoprotein degradation occurred by two different mechanisms. In one, lipoprotein was degraded within the cell, following binding and internalisation. In the other, degradation occurred in the medium, which seemed to contain protease activity released from cells during incubation. Though lipoprotein-deficient serum apparently interfered with degradation in the medium, bovine serum albumin had no such effect. The lysosomal inhibitor, chloroquine, reduced degradation by 60% without inhibiting HDL binding. Intestinal cell extracts contained at least two different proteases, with pH optima of 4.5 and 8.0, respectively. Comparing HDL and LDL degradation on a molar basis, more HDL particles were degraded by the cell-free extracts at pH 4.5. This degradation was activated by dithiothreitol and was inhibited by iodoacetic acid. From these observations we conclude that HDL3 is taken up by the rat intestinal mucosal cell through a specific binding site and subsequently degraded by a thiol-dependent protease in the lysosome.  相似文献

Despite of the economic importance of the olive fly (Bactrocera oleae) and the large amount of biological and ecological studies on the insect, the factors driving its population dynamics (i.e., population persistence and regulation) had not been analytically investigated until the present study. Specifically, our study investigated the autoregressive process of the olive fly populations, and the joint role of intrinsic and extrinsic factors molding the population dynamics of the insect. Accounting for endogenous dynamics and the influences of exogenous factors such as olive grove temperature, the North Atlantic Oscillation and the presence of potential host fruit, we modeled olive fly populations in five locations in the Eastern Mediterranean region. Our models indicate that the rate of population change is mainly shaped by first and higher order non-monotonic, endogenous dynamics (i.e., density-dependent population feedback). The olive grove temperature was the main exogenous driver, while the North Atlantic Oscillation and fruit availability acted as significant exogenous factors in one of the five populations. Seasonal influences were also relevant for three of the populations. In spite of exogenous effects, the rate of population change was fairly stable along time. We propose that a special reproductive mechanism, such as reproductive quiescence, allows populations of monophagous fruit flies such as the olive fly to remain stable. Further, we discuss how weather factors could impinge constraints on the population dynamics at the local level. Particularly, local temperature dynamics could provide forecasting cues for management guidelines. Jointly, our results advocate for establishing monitoring programs and for a major focus of research on the relationship between life history traits and populations dynamics.  相似文献

Certain strains of mice display an increased frequency of fetal resorption, but little is known about the effector mechanisms involved. We have examined the events associated with lipopolysaccharide (LPS)-induced fetal resorption in mice. Administration of 25 micrograms LPS on Day 12 of gestation resulted in the appearance of tumour necrosis factor-alpha (TNF-alpha) in the amniotic fluid and fetal resorption. Levels of LPS-induced TNF-alpha were reduced by 90% after pretreatment with the TNF-alpha-suppressing drug pentoxifylline (PXF). Treatment of pregnant mice during early gestation with 0.1 micrograms LPS resulted in fetoplacental resorption which was maximal when the LPS was given on Day 8. Resorption induced by 0.1 micrograms LPS on Day 8 of gestation was significantly reduced by pretreatment with PXF. Infiltration of asialo-GM1-positive cells was observed in the decidual-ectoplacental cone area of embryonic units from LPS-treated mice. In addition, treatment with anti-AGM1 antiserum prevented the LPS-induced resorption. Our results suggest that TNF-alpha and asialo-GM1-positive cells are involved in LPS-induced fetal resorption.  相似文献

Summary Foraging efficiency was studied by measuring the rate of tuna fish bait discovery by ants in unshaded and two types of shaded coffee systems. We also investigated the effect of weed biomass upon ant foraging efficiency. We found that the rate of discovery was faster in the coffee system with no shade than in systems with shade trees. The rate of discovery in the two types of shade systems (monospecific and polyspecific shade) was similar. Differences in the foraging rate between systems seem to be related to the composition of the ground ant community in each of the systems, and to cumulative factors such as plant diversity, microclimate and interspecific competition. No correlation was found between weed biomass and ant foraging efficiency. The results of this study support the idea of manipulating agroecosystem plant and structural diversity in order to enhance pest regulation by ants.  相似文献

Increasing body weight appears to alter lipid metabolism in adipose tissue. We have measured the content of lipoprotein lipase and the uptake of chylomicron triglyceride fatty acids in epididymal fat pads of rats of different weights. In order that the results might be expressed in terms of cell numbers, the relationship between the weights of fat pads and the numbers and volumes of fat cells isolated from them was determined. Highly significant correlations were found between fat pad weight and both the number and the volume of the individual adipocytes. In rats weighing from 140 to 350 g, the increase in the size of fat pads was attributable almost equally to increases in cell size and in cell number. Lipoprotein lipase activity was measured in acetone powders of whole fat pads and of isolated fat cell preparations. With both, lipoprotein lipase activity per cell diminished significantly as the weight of fat tissue increased, i.e., larger fat cells contained less enzyme per cell than smaller cells. The uptake of triglyceride fatty acid radioactivity was measured after incubation of fat pads with radiolabeled rat lymph chylomicrons in flasks containing either buffer alone or with added glucose or glucose plus insulin. The addition of glucose and insulin led to a mean increase of 70% in the uptake of radioactivity, but larger adipocytes were stimulated less than smaller cells. This resulted in a significant negative correlation between the weights of fat pads and the uptake of radioactivity. Enlargement of fat cells also led to a diminution in their capacity to esterify fatty acids.  相似文献

As part of the development of a sterile insect technique (SIT) application for the Ethiopian fruit fly, Dacus ciliatus, we studied the mating behaviour of a laboratory‐adapted strain (a 4‐year‐old colony kept for more than 40 generations) and a wild population. Effects of laboratory rearing and irradiation were assessed by carrying out mating compatibility and male mating competitiveness tests using a 1:1 ratio between irradiated (120 Gy) laboratory males and non‐irradiated wild males. Mating behaviour was studied on host and non‐host plants under field cage conditions. To assess the effect of mass rearing upon male performance, we repeated the mating competitiveness test using non‐irradiated laboratory insects. The findings indicated a high degree of compatibility among the two populations and satisfactory competitiveness of the irradiated laboratory males (ca. 35%). The competitiveness of non‐irradiated laboratory males was also ca. 35%, suggesting that no adverse effects resulted from their irradiation. Mating occurred only at twilight and mainly on the underside of leaves of non‐host plants (lemon trees). Findings are discussed in view of their implications for a future application of SIT against this fruit fly pest.  相似文献

During the last decades, the economic importance of tephritid fruit flies (FF) has increased worldwide because of recurrent invasions and expansions into new areas, and reduced control capabilities of current control systems. Efficient monitoring systems, thus, are required to provide fast information to act promptly. With this aim in mind, we developed two electronic trap (e‐trap) versions for adult FF: one with specific volatiles for male and female adult Ceratitis capitata, and the second, based on the attraction of adult FF to yellow colour, targeting Dacus ciliatus, Rhagoletis cerasi and Bactrocera oleae. In the case of B. oleae, the female pheromone and ammonium bicarbonate were added as synergists. In the two versions, attracted FF were retained in the trap on glued surfaces. Real‐time images of the surfaces were automatically taken and transmitted to a server. We tested the two e‐trap versions in insect‐proof cages, where flies were released and recaptured, and in commercial orchards throughout the Mediterranean: C. capitata in peach orchards in Italy; R. cerasi in cherry orchards in Greece; B. oleae in olive orchards in Spain and in Greece; and D. ciliatus in melons in plastic tunnels in Israel. The e‐trap showed excellent abilities to transmit real‐time images of trapped FF and a high specificity for trapping different FF species. The ability of the entomologist to correctly classify FF from images in the office was >88%. In addition, average number of flies/trap in e‐trap grids did not differ from numbers reported on grids of conventional traps that were operating simultaneously. The e‐traps developed and tested in this study provide the basis for the real‐time monitoring of FF were no olfactory attractants are available, and for the surveillance of alien FF incursions where generic, but not specific, olfactory attractants exists.  相似文献

To elucidate the mechanisms underlying the plasma triacylglycerol-lowering effects of certain fish oils, livers from male rats fed either a standard commercial diet (controls) or diets supplemented with 15% (w/w) fish or safflower oils were perfused with undiluted rat blood. Rates of hepatic lipogenesis, measured by the incorporation of 3H2O into fatty acids, followed the order: control greater than safflower oil greater than fish oil. Secretion of newly synthesized fatty acids in very-low-density lipoproteins was also inhibited by the feeding of both oil-supplemented diets with the greater suppression being seen in livers from animals fed fish oil. The hepatic release of very-low-density lipoprotein triacylglycerol mass was also significantly depressed in animals fed the fish oil-supplemented diet but not in those fed safflower oil. Ketogenesis did not differ between livers from rats fed the control and safflower oil diets but was significantly raised in the fish oil group. Increased ketogenesis with fish oil was paralleled by a decrease in the sensitivity of carnitine palmitoyl transferase of isolated mitochondria to inhibition by malonyl-CoA. The inhibitory effect of malonyl-CoA in the safflower oil group was intermediate between that in the fish oil and control groups. Activities of glycerophosphate acyltransferase with either palmitoyl-CoA or oleyl-CoA were increased by feeding oil-supplemented diets. Activity with palmitoyl-CoA that was suppressible by N-ethylmaleimide was also considerably diminished in both groups. The results indicate that the lowering of plasma triacylglycerols by fish oil reflects: (a) diminished lipogenesis; (b) increased fatty acid oxidation possibly in peroxisomes; and (c) diminished secretion of triacylglycerols by the liver.  相似文献

The effects of one acaricide (cyhexatin), two fungicides (penconazole and sulfur), and six insecticides (azinphos-methyl, chlorpyrifos, imidacloprid, pirimicarb, triazamate and vamidothion) on the adult stage of the parasitoidAphelinus mali (Haldeman), a parasite of the woolly apple aphidEriosoma lanigerum (Hausmann), were investigated under laboratory conditions. Chlorpyrifos (an organophosphorus insecticide-OP) was found to be highly toxic to the adult wasps. Vamidothion (OP) was more toxic to the parasitoid than azinphos-methyl (OP). On the other hand, both chlorpyrifos and azinphos-methyl were found to be harmless to the immature stages of the parasitoid in a test conducted under semi-field conditions. Of the other insecticides, imidacloprid was more toxic to the adult parasitoid than pirimicarb and triazamate. Neither cyhexatin nor penconazole had a considerable toxic effect upon the parasitoid. In contrast, sulfur was found to be moderately toxic to the parasitoid under laboratory conditions, as well as in a field survey in which we followed populations ofE. lanigerum andA. mali parasitization in sulfur-treated plots, and in a plot treated withAmpelomyces quisqualis Ces., a fungus antagonist to powdery mildew.
