INTRODUCTION: The lipid composition of spermatozoa plays an important role for successful fertilization. PATIENTS AND METHODS: In the present study, we analyzed the fatty acid (FA) composition of spermatozoa of normozoospermic, asthenozoospermic, oligozoospermic and oligoasthenozoospermic men. RESULTS: Spermatozoa from asthenozoospermic (P<0.01), oligozoospermic (P<0.05) and oligoasthenozoospermic men (P<0.05) had lower levels of docosahexaenoic acid (22:6w3, DHA) than those from normozoospermic men. In oligozoospermic and asthenozoospermic men, spermatozoa 18:0 content was higher than that of normozoospermics (P<0.01 and P<0.001, respectively). 18:1w9 was higher in oligoasthenozoospermic and oligozoospermic samples when compared with normozoospermic samples (P<0.05 for both). While from the point of view of total w6 FAs there was no significant difference among the groups, the w6/w3 ratio was significantly higher in asthenozoospermic samples than in normozoospermic samples (P<0.05). Monounsaturated fatty acids (MFA) were higher in oligozoospermic samples (P<0.05) than in normozoospermic samples, polyunsaturated fatty acids (PUFA) were lower in asthenozoospermic (P<0.01), oligoasthenozoospermic (P<0.05) and oligozoospermic samples (P<0.05) than in normozoospermic samples. Saturated fatty acids (SFA) were significantly higher in asthenozoospermic (P<0.01) and oligozoospermic samples (P<0.05) compared with normozoospermic samples. In correlation analysis, there were significant positive correlations between DHA with sperm motility (r=0.53), sperm concentration (r=0.36) and normal sperm morphology (r=0.30). In addition, there were significant correlations between PUFA with sperm motility (r=0.50), sperm concentration (r=0.35), and normal sperm morphology (r=0.28), and between w6/w3 with sperm motility (r=-0.47), sperm concentration (r=-0.27), and normal sperm morphology(r=-0.24). DISCUSSION: These suggest that decreased DHA and PUFA, and increased w6/w3 in spermatozoa may be related to infertility in oligo- and/or asthenozoospermic men.  相似文献

Cyanobacteria, red algae, and cryptomonad algae utilize phycobilin chromophores that are attached to phycobiliproteins to harvest solar energy. Heme oxygenase (HO) in these organisms catalyzes the first step in phycobilin formation through the conversion of heme to biliverdin IXα, CO, and iron. The Synechocystis sp. PCC 6803 genome contains two open reading frames, ho1 (sll1184) and ho2 (sll1875), whose products have in vitro HO activity. We report that HO2, the protein encoded by ho2, was induced in the cells growing under a microaerobic atmosphere [0.2% (v/v) O2], whereas HO1 was constitutively expressed under both aerobic and microaerobic atmospheres. Light intensity did not have an effect on the expression of both the HOs. Cells, in which ho2 was disrupted, were unable to grow microaerobically at a light intensity of 40 μmol m−2 s−1, but did grow microaerobically at 10 μmol m−2 s−1 light intensity. These cells grew normally aerobically at both light intensities. Comparative analysis of complete cyanobacterial genomes revealed that possession of two HOs is common in cyanobacteria. In phylogenetic analysis of their amino acid sequences, cyanobacterial HO1 and HO2 homologs formed distinct clades. HO sequences of cyanobacteria that have only one isoform were most similar to HO1 sequences. We propose that HO2 might be the more ancient HO homolog that functioned under low O2 tension, whereas the derived HO1 can better accommodate increased O2 tension in the environment.  相似文献

The aim of this study was to investigate whether nicotine affects 6-phosphogluconate dehydrogenase (6PGD) enzyme activity in some rat tissues, and to see the modulatory effects of vitamin E on this effect in vivo. In addition, the effects of nicotine and vitamin E on 6PGD activity were also tested in vitro. The groups were: nicotine [0.5 mg/kg/day, intraperitoneal (i.p.)]; nicotine + vitamin E [75 mg/kg/day, intragastric (i.g.)]; and control group (receiving only vehicles). There were eight rats per group and supplementation period was 3 weeks. The results of in vivo study showed that nicotine activated the muscle, lungs, and testicular 6PGD enzyme activity but had no effect on heart and liver 6PGD activity. Also, nicotine + vitamin E activated the muscle, testicle, and liver 6PGD enzyme activity, while this combination had no effect on heart, and lungs in vivo. When nicotine is administered with vitamin E the increase in 6PGD enzyme activity in muscle and testicles were lower. On the other hand the increase in 6PGD enzyme activity was eliminated by vitamin E in lungs, while 6PGD enzyme activity was increased by vitamin E, which was not affected by nicotine only. In vitro results correlated well with in vivo experimental results. Our results suggest that vitamin E may favourably increase 6PGD enzyme activity in liver in nicotine treated rats, while it has negligible effects on this enzyme activity in other tissues.  相似文献

Novel chiral mono and diamide derivatives of calix[4]arene have been prepared from the aminolysis reaction of 5,11,17,23-tetra-tert-butyl-25,27-diethoxycarbonyl-methoxy-26,28-dihydroxycalix[4]arene 1 and 25,27-diethoxycarbonyl-methoxy-26,28-dihydroxycalix[4]arene 2 with chiral (S)-(-)-1-phenylethylamine (PEA) and (1S,2S)-(+)-2-amino-1-(4-nitrophenyl)-1,3-propanediol, respectively. Spectrophotometric titrations have been performed in CHCl(3) at 20-30 degrees C in order to obtain the binding constants (K) and the thermodynamic quantities (DeltaH and DeltaS) for the stoichiometric 1:1 inclusion complexation of various chiral amines with these new host compounds. Preliminary experiments were undertaken to confirm the complexation properties of receptors 9 and 13 with PEA by (1)H NMR in CDCl(3) at room temperature. The molecular recognition abilities and enantioselectivities for guests (R and S)-alpha-PEA and (R and S)-cyclohexylethylamine (CHEA) are discussed from a thermodynamic point of view.  相似文献

The aim of this research was to link vegetation characteristics, such as spatial and temporal distribution, and environmental variables, with land cover information derived from remotely sensed satellite images of the Eastern Mediterranean coastal wetlands of Turkey. The research method was based on (i) recording land cover characteristics by means of a vegetation indicator, and (ii) classifying and mapping coastal wetlands utilizing a Landsat Thematic Mapper (TM) image of Çukurova Deltas in Turkey. Vegetation characteristics of various habitats, such as sand dunes, salt marshes, salty plains and afforestation areas, were identified by field surveys. A Landsat TM image of 4 July 1993 was pre-processed and then classified using the Maximum Likelihood (ML) algorithm and Artificial Neural Networks (ANN). As a result of this supervised classification, the land cover types were classified with a largest accuracy of 90.2% by ANN. The classified satellite sensor imagery was linked to vegetation and bird census data, which were available through literature in a Geographical Information System (GIS) environment to determine the spatial distribution of plant and bird biodiversity in this coastal wetland. The resulting data provide an important baseline for further investigations such as monitoring, change detections and designing conservation policies in this coastal ecosystem.  相似文献

The mismatch discrimination potential of probes in fluorescence in situ hybridization can be defined as the difference between the melting formamide points of perfect complementary and mismatched duplexes (Delta[FA](m)). Using a combined experimental and theoretical approach, Delta[FA](m) was determined for a set of 35 mismatched probes targeting seven locations in the 16S rRNA of Escherichia coli. The mismatches were created by changing single nucleotides on the probes, while maintaining the target unmodified. Estimated Delta[FA](m) values were used to systematically evaluate four predictors of mismatch stability: weighted mismatch (WM) scores from the software arb, published statistical summary of microarray hybridizations, free energy of mismatch stability (DeltaDeltaG degrees (1)) and theoretical Delta[FA](m) estimations obtained with a thermodynamic model. Based on the predictors' ability to explain variability in Delta[FA](m) and to discriminate weak mismatches from strong ones, DeltaDeltaG degrees (1) and WM scores from arb (with an updated set of relative strength parameters) were demonstrated to be adequate estimators of mismatch stability, with DeltaDeltaG degrees (1) offering the benefit of capturing the variability associated with nearest-neighbour effects and being compatible with thermodynamic models of in situ hybridization. The use of DeltaDeltaG degrees (1) and WM in probe design was illustrated as a tool that complements experimental design approaches.  相似文献

Three beta-cyclodextrin (polymers 1-3) and a starch-based (polymer 4) polymers were synthesized using hexamethylene diisocyanate (HMDI) as a cross-linking agent in dry dimethylformamide and used as a sorbent for the removal of some selected azo dyes from aqueous solutions. The cross-linked polymers were characterized by Fourier transform infrared spectroscopy, thermogravimetric and differential scanning calorimetric analysis. Results of sorption showed that cyclodextrin and starch based polymers can be effectively used as a sorbent for the removal of anionic azo dyes. The Influence of the amide groups and the chemical structure of azo dyes are also studied. Results of sorption experiments showed that these adsorbent exhibited high sorption capacities toward Direct Violent 51 (80% for polymer 1, 69% for polymer 2, 70% for polymer 3 and 78% for polymer 4). The sorption capacity of dyes on the polymers was dependent on the presence of sulfonate groups of the anionic dyes. In order to explain the results an adsorption mechanism mainly physical adsorption and interactions such as hydrogen bonding, ion-exchange due to the nature of the polymer network and the formation of an inclusion complex due to the beta-CD molecules through host-guest interaction is proposed.  相似文献

This communication describes preparation, characterization, and the evaluation of sorption properties of a calix[4]arene-based chitosan polymer (C[4]BCP). C[4]BCP was used to sorption studies of some heavy metal cations (Co(2+), Ni(2+), Cu(2+), Cd(2+)(,) Hg(2+) and Pb(2+)) and dichromate anions (Cr(2)O(7)(2-)/HCr(2)O(7)(-)) as sorbent material. Also the supporting material (chitosan) was used for comparison in these experiments. The results for heavy metal cations showed that C[4]BCP was excellent sorbent and the effect of chitosan was low. In the sorption studies of dichromate anions, C[4]BCP was highly effective sorbent at pH 1.5.  相似文献

The objective of this study was to search for polymorphisms and gene regulatory sequences in the 5' flanking region of the sheep insulin-like growth factor I (IGF-I) gene. PCR-SSCP analysis of the 5' flanking region revealed three banding patterns. Family study indicated that these patterns in mixed breed sheep corresponded with three genotypes (with their frequencies in parentheses) AA (0.70), AB (0.25), and BB (0.05), which arose from a one-locus, two allele (A, B) polymorphism. Genotypic frequencies in 22 purebred Polypay sheep were AA (0.77) and AB (0.23). Calculated frequency of the A allele in Polypays was 0.89. No deviation from Hardy-Weinberg equilibrium was detected in this study. Fragments amplified using DNA from homozygous individuals were sequenced and aligned next to each other. A T to C transition and a G to C transversion were found at positions 179 and 181, respectively, of the amplified PCR product, resulting in recognition sites for Bsp143II and HaeI. Analysis of a fragment of 2,162 base pairs upstream of Exon 1, assembled from sheep ESTs and sequence of our amplified PCR products, revealed a promoter sequence approximately 100 bp downstream of the polymorphic sites. The assembled DNA fragment shared 70% sequence homology between sheep and human. These results suggest that sequence of the 5' flanking region of IGF-I gene and location of the IGF-I promoters are similar in human and sheep.  相似文献

Ant responses were tested under both the natural geomagnetic and artificially induced Earth-strength electromagnetic field. Foragers were trained for a month to visit a food source at the north arm accessed through an orientation platform assembly. Under the natural geomagnetic field, when all other orientational cues were eliminated, results indicated significant heterogeneity of ant distribution with the majority seeking geomagnetic north in darkness. However, in light, foragers failed to discriminate geomagnetic north. Under shifted artificial electromagnetic field, orientation was predominantly on the artificial magnetic N/S axis with a significant preference for the artificial north in both light and dark conditions.  相似文献
