Six peaks of small GTP-binding proteins (G proteins) were separated by column chromatographies from the cytosol fraction of the differentiated HL-60 cells: two peaks of rho p21, one peak of smg/rap1 p21, two peaks of rac1 p21, and one peak of an unidentified small G protein with a Mr of about 20,000 (20 KG). smg GDS, previously thought to be a stimulatory GDP/GTP exchange protein for smg p21, Ki-ras p21, and rho p21, but not for Ha-ras p21 or smg p25A, was also active on rac1 p21. rho GDI, previously thought to be an inhibitory GDP/GTP exchange protein specific for rho p21, was also active on rac1 p21. These results indicate that both smg GDS and rho GDI are active on multiple small G proteins.  相似文献

The chemical synthesis of four phosphonate-containing phosphatidylserine analogs namely, L-serine (±)-[2,3-bis(hexadecyloxy) and 2,3-bis(Palmitoyloxy)-propyl] phosphonates, and L-serine (±)-[3,4-bis(hexadecyloxy and 3,4-bis(palmitoyloxy)-butyl]phosphonates is described. (±)-2,3-Bis(hexadecyloxy) and 2,3-bis(palmitoyloxy)-propylphosphonic acids and (±)-3,4-bis (hexadecyloxy)butylphosphonic acid were prepared by reaction of tris(trimethylsilyl) phosphite on the corresponding haloalkane. Condensation of the above phosphonic acids or (±)-3,4-bis (palmitoyloxy)butylphosphonic acid with N-carboxy-L-serine dibenzyl ester in the presence of trichloroacetonitrile or triisopropylbenzenesulfonyl chloride yielded the protected serine intermediates, which on hydrogenolysis gave the desired L-serine analogs. By a similar route, 1,2-dihexadecyl-rac-glycero-3-phosphoric acid was converted to 1,2-dihexadecyl-rac-glycerophospho-L-serine [L-serine (±)-2,3-bis(hexadecyloxy)propyl hydrogen phosphate(ester)].  相似文献

Reflex peripheral vasoconstriction induced by activation of cardiopulmonary baroreceptors in response to reduced central venous pressure (CVP) is a basic mechanism for elevating systemic vascular resistance and defending arterial blood pressure during orthostatically-induced reductions in cardiac filling and output. The sensitivity of the cardiopulmonary baroreflex response [defined as the slope of the relationship between changes in forearm vascular resistance (FVR) and CVP] and the resultant vasoconstriction are closely and inversely associated with the amount of circulating blood volume. Thus, a high-gain FVR response will be elicited by a hypovolemic state. Exposure to microgravity during spaceflight results in reduced plasma volume. It is therefore reasonable to expect that the FVR response to cardiopulmonary baroreceptor unloading would be accentuated following adaptation to microgravity. Such data could provide better insight about the physiological mechanisms underlying alterations in blood pressure control following spaceflight. We therefore exposed eleven men to 6 degrees head-down bedrest for 7 days and measured specific hemodynamic responses to low levels of the lower body negative pressure to determine if there are alterations in cardiopulmonary baroreceptor stimulus-FVR reflex response relationship during prolonged exposure to an analog of microgravity.  相似文献

Carob pod extract and its tannin and sugar fractions were compared with gallotannic acid and sucrose for their effect on the cellulolytic, proteolytic, protein biosynthetic, and deaminative activities of rumen microorganisms. The inhibitory effects of carob pod extract upon the cellulolysis and deamination were correlated mainly with its sugar, rather than its tannin components. On the other hand, proteolytic activity and protein biosynthesis were more significantly affected by the tannin fraction. In contrast to the tannin fraction of carob pod extract, gallotannic acid inhibited cellulolytic activity. The harmful effect of a low concentration of tannins on protein biosynthesis could be prevented by the addition of carbohydrates to the reaction mixture. At high tannin concentration (40 μg/ml), however, the addition of carbohydrates did not prevent the inhibition.  相似文献

Cytotoxic and antiretroviral activity of cycloSal-d4TMP derivatives were tested in a new AZT-resistant H9 cell subline (H9rAZT250). The results showed, that cycloSal-d4TMP derivatives overcame resistance of HIV-1 to d4T in H9rAZT250 cells, which exert decreased thymidine kinase (TK) gene expression.  相似文献

smg p25A/rab3A p25 is a member of the small GTP-binding protein superfamily which is implicated in intracellular vesicle transport. smg p25A has a cDNA-predicted C-terminal structure of Cys-Ala-Cys. The protein purified from bovine brain membranes is geranylgeranylated at both the two cysteine residues and carboxyl-methylated at the C-terminal cysteine residue. Two types of prenyltransferase for small GTP-binding proteins have thus far been reported: ras p21 farnesyltransferase (ras p21 FT) and rhoA p21 geranylgeranyltransferase (rhoA p21 GGT). Neither of them geranylgeranylated smg p25A having a C-terminal Cys-Ala-Cys structure. In this paper, a smg p25A GGT was partially purified from bovine brain cytosol and separated from the ras p21 FT and rhoA p21 GGT by column chromatographies. smg p25A GGT transferred the geranylgeranyl moiety from geranylgeranyl pyrophosphate to both the two cysteine residues in the C-terminal Cys-Ala-Cys structure of smg p25A. smg p25A GGT did not use farnesyl pyrophosphate as a substrate and was also inactive on c-Ha-ras p21 and rhoA p21 with either farnesyl pyrophosphate or geranylgeranyl pyrophosphate as a substrate. These results indicate that there are at least three types of prenyltransferase for small GTP-binding proteins in mammalian tissues.  相似文献

苹果蠹蛾Cydia pomonella(L.)是世界著名的严重危害苹果的重要入侵害虫,常造成20%~30%的产量损失,现已传入新疆、甘肃、内蒙古、宁夏、黑龙江和吉林等省区,对我国全国苹果安全生产构成严重威胁。2009年农业部科教司批准设立公益性行业(农业)专项"入侵生物苹果蠹蛾监测与防控技术研究",由中国科学院动物研究所主持,全国农业技术推广服务中心、西北农林科技大学、中国农科院植保所和郑州果树所、中国农业大学、河北农业大学以及相关省区植保部门等16单位参加。本文对该项目执行3年中在苹果蠹蛾关键生物学特性、实用监测和防治技术方面取得的主要进展进行了总结,详细情况可通过苹果蠹蛾信息网(www.codlingmoth.net)获得。  相似文献
