Acidic proteinase from the trout spawn is 640 fold purified (yield 22%). Purification includes autolysis, acid treatment, ammonium sulphate fractionation, G-100 Sephadex gel-filtration, ion-exchange chromatography on DEAE-cellulose. Molecular mass of the enzyme under study is 70 kDa according to the data of gel-filtration. Acidic proteinase displays its greatest activity towards hemoglobin (pH 4.0, 37 degrees C) and is inhibited completely by EDTA, by 50%--by Pb2+ and soya inhibitor of trypsin and 2.8 times activated by Zn2+. Enzyme activity is not affected by dithiotreitol, iodine acetate, phenylmethylsulphonylfluoride parachloromercurybenzoate, Hg2+, Na+, Co2+, Ca2+.  相似文献

Glutathione S-transferase (GST) was isolated from the Esox lucius liver and purified to the homogenous state by SDS-PAGE and isoelectrofocusing. It was found to be a homodimer with a subunit molecular weight of 25235.36 Da (HPLC-MS/MS data) and pI of about 6.4. Its substrate specificity, thermal stability, some kinetic characteristics, and optimum pH were studied. The enzyme was identified as Alpha class GST.  相似文献

We studied the growth performance and lipid composition of liver, muscles and visceral fat in rainbow trout Parasalmo mykiss (Walbaum, 1792) and the diets used in its cultivation during the spring-summer season. The differences in dietary lipid composition determined modifications in the biochemical composition of tissues of rainbow trout and had an effect on the weight and growth rate of the fish. Trophic and seasonal factors also had their influence on the studied parameters. The degree of influence depended on the functional characteristics of tissues and organs of rainbow trout.  相似文献

The activity of the enzymes of the energy and carbohydrate metabolisms (cytochrome-c oxidase, L-lactate dehydrogenase, aldolase, and glycerol-1-phosphate dehydrogenase) have been studied in White Sea herring (the 1+, 2+, and 3+ age groups) sampled in Onega Bay, Dvina Bay, and Kandalaksha Bay of the White Sea. The bays differ in the hydrological regime, ecological and feeding conditions. The individual variability of the enzyme activity was the largest in the herring of the age 1+. The flexibility of the intensity and vector of the basic metabolic reactions probably supports the energy homeostasis, preconditions the switching to the most effective way of using the resources, and regulates the synthesis of the structural and storage molecules, as well as vectors the adaptation strategy of herring specimens of each age group to the hydrological regime, environment, and feeding conditions of the particular bay, corresponding to their age-related characteristics.  相似文献

The activity and the cofactor specificity of xylose reductase and xylitol dehydrogenase were studied in extracts of yeasts from the genera Candida, Kluyveromyces, Pachysolen, Pichia, and Torulopsis grown under microaerobic conditions. It was found that xylitol dehydrogenase in all of the yeast species studied is specific for NAD+; xylose reductase in the xylitol-producing species C. didensiae, C. intermediae, C. parapsilosis, C. silvanorum, C. tropicalis, Kl. fragilis, Kl. marxianus, P. guillermondii, and T. molishiama is specific for NADPH; and xylose reductase in the ethanol-producing species P. stipitis, C. shehatae, and Pa. tannophilus is specific for both NADPH and NADH.  相似文献

The differentiating effect of DMSO on K562 cells was studied against the background of pretreatment of the cells by the modulators of activities of protein kinase C and Ca signaling, phorbol 12-myristate 13-acetate, and ionophore A23187. The 2-hour pretreatment of K562 cells with A23187 (1 M), rather than phorbol 12-myristate 13-acetate (0.1 M), led to inhibition of the differentiating effect of DMSO (0.1%) during the subsequent four days of incubation. When the cells were pretreated jointly with A23187 and phorbol 12-myristate 13-acetate, the DMSO-induced erythroid differentiation was restored. Analysis of the DNA-degrading effect of the reagents used on K562 cells by fluorescent dyes under the same conditions suggests that this activity is induced in the presence of DMSO in the cells pretreated with a combination of phorbol 12-myristate 13-acetate and A23187 or without pretreatment.  相似文献

The young of the year mykiss Parasalmo mykiss and coho salmon Oncorhynchus kisutch which perform downstream migration and which stay in the coastal zone during the period of primary dispersion represent different phenotypic groups. They differ in weight, length, and lipid status. Such differentiation is a factor controlling the formation of intrapopulation diversity and of the life strategy of fish. It may determine the timing of smoltification in the future and, further on, of the return for spawning. There were found some analogies and differences in characteristics of lipid metabolism in the investigated species. The content of total lipids decreases by autumn in underyearlings of both species. Furthermore, changes in the content of particular lipid fractions are multidirectional and various scaled, seemingly being related to physiological preparation for the wintering.  相似文献

In comparative-evolutionary aspect, the experimental data are considered about activity, biochemical properties, and peculiarities of structural organization of proteins of the calpain family in some invertebrates and fishes. Peculiarities of calpain-like proteins of invertebrates--the predecessors of calpains of higher animals are revealed. By the example of the studied taxons, there is traced complication of the structural organization and mechanisms of control of the calpain activities, which reflects stages of molecular evolution of the protein family.  相似文献
