Collagen crosslinking and cartilage glycosaminoglycan composition in normal and scoliotic chickens

The amounts of lysine-derived crosslinks in collagens from tendon, cartilage, intervertebral disc, and bone and changes in the composition of sternal cartilage glycosaminoglycans were estimated in two lines of chickens, a control-isogenic line and a line that develops scoliosis. In the scoliotic line, scoliosis first appears at 3-4 weeks and progressively increases in severity and incidence so that 90% of the birds express the lesion by week 10. We have reported previously that cartilage, tendon, and bone collagens from scoliotic birds are more soluble than corresponding collagens from normal birds. Herein, collagen crosslinking and altered proteoglycan metabolism are examined as possible mechanisms for the differences in collagen solubility. At 1 week of age there were fewer reducible crosslinking amino acids (hydroxylysinonorleucine, dihydroxylysinonorleucine, and lysinonorleucine) in collagens from sternal cartilage and tendon in the scoliotic line than in the isogenic line. However, by week 3 and at weeks 5 or 7 values were similar in both groups. The amounts of hydroxypyridinium in vertebral bone and intervertebral disc collagen were also similar in both groups of birds. Consequently, differences in collagen crosslinking do not appear to be a persistent developmental defect underlying the expression of scoliosis in the model. However, differences were observed in cartilage proteoglycans and glycosaminoglycans from the scoliotic line that were not present in cartilage from the isogenic line. The average molecular weight of the uronide-containing glycosaminoglycans was 30% less in the scoliotic line than in the isogenic line, i.e., 12,000 compared to 18,000. The size distribution of cartilage proteoglycans from the scoliotic line also differed from that of proteoglycans from the isogenic line.(ABSTRACT TRUNCATED AT 250 WORDS)     

Morphology of defense glands of the opilionids (daddy longlegs) Leiobunum vittatum and L. flavum (Arachnida: Opiliones: Palpatores: Phalangiidae)

Opilionid defense glands consist of 0.5 × 0.9-mm sacs attached to the underside of low tubercles located on the dorsal side of the cephalothorax, posterior to the first pair of legs. Each gland opens via an elongated slit, located in the posterior floor of a crater that is situated at the summit of the tubercle. The center of the sac, called the reservoir, is lined by a cuticle consisting of epicuticle and endocuticle which is continuous through the slit with the exoskeleton. The layers of cuticle vary in thickness with different locations in the gland. A hemocoelomic (basement) membrane, 0.5–1, μ thick, forms the boundary between glandular cells and hemocoel. The gland has a nonsecretory portion consisting only of cuticle-supporting cells and a secretory portion consisting of secretory and cuticle-supporting cells. The cuticle lining the reservoir in the secretory area is broached by many cuticle-lined ductules, each of which drains an isolated intercellular space called the intercalated cistern. This in turn drains microvilli-lined canaliculi located between and extending into secretory cells. The cisterns are devoid of microvilli. Secretory cell cytoplasm contains a Golgi apparatus, many free ribosomes, rough endoplasmic reticulum (RER), two types of granules (speckled and dense), and mitochondria. Speckled granules are partially filled with fairly large particles and are found in association with the Golgi apparatus. They also surround canaliculi into which they empty. Dense granules are packed with very small particles, have a gray homogeneous appearance, and are scattered throughout the cytoplasm. Mitochondria containing matrix granules tend to scatter throughout the cytoplasm but are concentrated around canaliculi.     

Sexual behaviour of group-living adolescent chimpanzees

This paper reports a study of the heterosexual behavior of three male and four female captive adolescent chimpanzees living a semi-natural life style in a large field enclosure. Observations made with binoculars from an over-head deck were balanced over the daylight hours and the seven weekdays. We recorded 213 copulations in 741.25 hours of observation between February 14 and July 21, 1972. Analysis of the observations suggests that group-living chimpanzees in a large field enclosure behave more like free-living chimpanzees than like other captive chimpanzees paired in small cages. In this group, copulations were non-randomly distributed throughout the day, a finding in agreement with results on wild-living chimpanzees but not previously reported for captive chimpanzees. In this group, moreover, copulations were highly concentrated within each female's period of maximum tumescence, as are those of free-living chimpanzees; and individual differences in sexual attractiveness among females were apparently based on a maturational threshold of swelling size, a phenomenon that has also been reported for wild chimpanzees.     

Determination of free and esterified cholesterol in human serum by high-performance liquid chromatography and optical activity detection

A separation and detection scheme is presented for the determination of free, esterified and total cholesterol in human serum. Separation is accomplished by reversed-phase high-performance liquid chromatography and the eluate is monitored by the laser-based optical activity detector. The method is simple, accurate and has the advantage of specificity and selectivity when compared with the many methods commonly used.
