* The 'in planta' visualization of F-actin in all cells and in all developmental stages of a plant is a challenging problem. By using the soybean heat inducible Gmhsp17.3B promoter instead of a constitutive promoter, we have been able to label all cells in various developmental stages of the moss Physcomitrella patens, through a precise temperature tuning of the expression of green fluorescent protein (GFP)-talin. * A short moderate heat treatment was sufficient to induce proper labeling of the actin cytoskeleton and to allow the visualization of time-dependent organization of F-actin structures without impairment of cell viability. * In growing moss cells, dense converging arrays of F-actin structures were present at the growing tips of protonema cell, and at the localization of branching. Protonema and leaf cells contained a network of thick actin cables; during de-differentiation of leaf cells into new protonema filaments, the thick bundled actin network disappeared, and a new highly polarized F-actin network formed. * The controlled expression of GFP-talin through an inducible promoter improves significantly the 'in planta' imaging of actin.  相似文献

The mitochondrial tRNA(Leu(UUR)) gene (MTTL) is a hot spot for pathogenic mutations that are associated with mitochondrial diseases with various clinical features. Among these mutations, the A3243G mutation was associated with various types of mitochondrial multisystem disorders, such as MIDD, MELAS, MERRF, PEO, hypertrophic cardiomyopathy, and a subtype of Leigh syndrome. We screened 128 Tunisian patients for the A3243G mutation in the mitochondrial tRNA(Leu(UUR)) gene. This screening was carried out using PCR-RFLP with the restriction endonuclease ApaI. None of the 128 patients or the 100 controls tested were found to carry the mitochondrial A3243G mutation in the tRNA(Leu(UUR)) gene in homoplasmic or heteroplasmic form. After direct sequencing of the entire mitochondrial tRNA(Leu(UUR)) gene and a part of the mitochondrial NADH dehydrogenase 1, we found neither mutations nor polymorphisms in the MTTL1 gene in the tested patients and controls, and we confirmed the absence of the A3243G mutation in this gene. We also found a T3396C transition in the ND1 gene in one family with NSHL which was absent in the other patients and in 100 controls. Neither polymorphisms nor other mutations were found in the mitochondrial tRNA(Leu(UUR)) gene in the tested patients.  相似文献

Background

Curable, non-viral pathogens account for a significant burden of sexually transmitted infections (STIs), and there is established evidence that STIs increase both HIV acquisition and transmission. We investigated the prevalence, trends, and factors associated with Chlamydia trachomatis, Neisseria gonorrhoeae, Trichomonas vaginalis and Treponema pallidum, and the performance of syndromic management, among a cohort of women working in bars, hotels, and other food and recreational facilities near large-scale mines in northwestern Tanzania.

Methods

HIV-negative women aged 18–44 years (N = 966) were enrolled and followed for 12 months in a microbicides feasibility study. We collected sociodemographic and behavioural data, performed clinical examinations, and tested for STIs, at enrolment and 3-monthly. Risk factors for STIs were investigated using logistic regression models with random effects. Sensitivity, specificity and predictive values of syndromic management were calculated.

Results

At enrolment, the prevalences of C. trachomatis, N. gonorrhoeae, T. vaginalis, and high-titre active syphilis were 111/956 (12%), 42/955 (4%), 184/945 (19%) and 46/965 (5%), respectively. There were significant decreases over time for C. trachomatis and T. vaginalis (OR trend per month: 0.94 [95% CI 0.91, 0.97]; and 0.95 [0.93, 0.98], respectively; both p<0.001). The majority of these infections were not diagnosed by the corresponding syndrome; therefore, most participants were not treated at the diagnosis visit. Syndromic management was poorly predictive of laboratory-diagnosed infections. We identified a number of risk factors for STIs, including low educational level, some sexual behaviours, and ever having been pregnant.

Conclusions

This analysis demonstrates that the prevalences of curable STIs are high among women who work in food and recreational facilities in northwestern Tanzania. Most of these infections are missed by syndromic management. Accurate and affordable rapid-point-of-care tests and innovative interventions are needed to reduce the burden of STIs in this population which is at increased risk for HIV.  相似文献

Over the last decade, cholera outbreaks in parts of Kenya have become common. Although a number of recent studies describe the epidemiology of cholera in Kenya, there is pauCity of information concerning the diversity and occurrence of mobile genetic elements in Vibrio cholerae strains implicated in these outbreaks. A total of 65 Vibrio cholerae O1 El Tor serotype Inaba isolated between 1994 and 2007 from various outbreaks in Kenya were investigated for mobile genetic elements including integrons, transposons, the integrating conjugative elements (ICEs), conjugative plasmids and for their genotypic relatedness.  相似文献

Drought can alter stem apex temperature and plant phenological development and it can then have an effect on the duration of the durum wheat stages. Thermal responses of plants to water stress are tentatively analysed as regards microclimate conditions by applying three different water treatments. North Africa's megafauna has developed behavioural adaptations to reduce energetic and fitness costs under harsh arid land conditions. Animal behaviour and activity patterns are difficult to study in the wild, but remote camera traps provide a solution to collecting data without the presence of a researcher influencing outcomes. Here, we report results from a study comprising over 20,382 camera-trap days during a 34-month period in Dghoumes National Park, Tunisia. We aimed to evaluate temporal activity patterns and their overlap and explore opportunities for niche partitioning. Our focal species were the reintroduced scimitar-horned oryx, north-African ostrich, dorcas gazelles and extant regional top-order predators, African wolf and red fox. We found differences in activity patterns between seasons across the focal species, with the most noticeable change being an increase in dawn activity from 1% to 33% between winter and summer for the red fox. Consequently, higher summer temperatures limit opportunities for temporal niche partitioning and push focal species towards dawn-time activity peaks resulting in higher intra-guild overlap values at dawn. Arid antelopes have physiological adaptations that enable them to better exploit ecological resources in hotter time periods than the carnivores, and this reduces inter-guild overlap during summer days.  相似文献

In previous reports from this laboratory it has been shown that the extended aeration process for biological treatment of organically laden municipal and/or industrial waste could be successfully employed for concurrent purification and sludge disposal. Also results using a modified process in which autodigestion was aided and controlled by periodic partial hydrolysis of small portions of the recycle sludge showed that operational control was feasible. There was some question regarding the success of such a process if the original waste contained a large portion of inorganic solids. Accordingly, a 1½ year pilot plant study was made using a waste (hydrolyzed trickling filter sludge) of exceptionally high ash content (50–60%). It was found that the ash content of activated sludge grown on this substrate did not continually increase nor did the high ash content of the waste interfere in any way with the efficiency of removal of organic matter. In general it exceeded 90 percent. Also a highly nitrified effluent was produced. A variety of analyses were performed: COD, BOD, TOC, suspended solids, NH3-N, organic-N, NO3-N, etc. Interrelationships between these important monitoring parameters for assessing plant performance offered useful insight into operational control for hydrolytically assisted extended aeration processes.  相似文献
