Analysis of the slow phases of the in vivo chlorophyll fluorescence induction curve. Changes in the redox state of Photosystem II electron acceptors and fluorescence emission from Photosystems I and II

An analysis of the photo-induced decline in the in vivo chlorophyll a fluorescence emission (Kautsky phenomenon) from the bean leaf is presented. The redox state of PS II electron acceptors and the fluorescence emission from PS I and PS II were monitored during quenching of fluorescence from the maximum level at P to the steady state level at T. Simultaneous measurement of the kinetics of fluorescence emission associated with PS I and PS II indicated that the ratio of PS I/PS II emission changed in an antiparallel fashion to PS II emission throughout the induction curve. Estimation of the redox state of PS II electron acceptors at given points during P to T quenching was made by exposing the leaf to additional excitation irradiation and determining the amount of variable PS II fluorescence generated. An inverse relationship was found between the proportion of PS II electron acceptors in the oxidised state and PS II fluorescence emission. The interrelationships between the redox state of PS II electron acceptors and fluorescence emission from PS I and PS II remained similar when the shape of the induction curve from P to T was modified by increasing the excitation photon flux density. The contributions of photochemical and non-photochemical quenching to the in vivo fluorescence decline from P to T are discussed.     

Re-examination of the acid hydrolysis of 5,6-anhydro-1,2-O-isopropylidene-β-l-idofuranose

The neutral polysaccharide isolated from the fruits of Cordia dichotoma was separated into two fractions, I and II, by gel filtration using Sephadex G-100. The major fraction, II, was found to contain d-glucose and l-arabinose in the molar ratio of 21:4. The results of complete hydrolysis with acid, permethylation studies, periodate oxidation, and Smith degradation suggested it to be an arabinoglucan, and the backbone of the polysaccharide to be composed of (1→6)-linked d-gluco-pyranosyl and (1→2)-linked l-arabinofuranosyl residues. The average degree of polymerization, determined through end-group analysis, was found to be ～144.     

Novel inhibitors of the αvβ3 integrin—lead identification strategy

A novel approach to inhibition of the αvβ3 integrin is described, which uses compounds designed to generate nM potency without using the arginine binding site.     

Identification of the uvrB gene product

We have constructed plasmids carrying various restriction fragments of the biouvrB region of the Escherichia coli chromosome. By analyzing the proteins synthesized in maxicells from the uvrB⁺ plasmid pDR1494, and its derivatives containing γδ sequences inserted in the uvrB gene, we have determined that the uvrB gene is about two kilobase-pairs in length, that it is transcribed clockwise on the standard E. coli genetic map, and that it codes for a single polypeptide of M_r = 84,000. The number of uvrB polypeptides in a normal cell is estimated to be about 140.We have also found that the uvrB gene is cut by EcoRI near its promoter.     

Endoglucanase A from Cellulomonas fimi in which the hinge sequence of human IgA1 is substituted for the linker connecting its two domains is hydrolyzed by IgA proteases from Neisseria gonorrhoeae

The hinge in IgA1 and the linker in endoglucanase A (CenA) are quite similar. The IgA1 hinge is 18 amino acids long and contains only proline, threonine and serine. The linker in CenA is 27 amino acids long and contains only proline, threonine and a single serine. IgA proteases from Neisseria gonorrhoeae cleave Pro-Ser and Pro-Thr bonds within the IgA1 hinge sequence, but they do not attack CenA. When the linker sequence of CenA is replaced with the hinge sequence of IgA1, the hybrid polypeptide is susceptible to the N. gonorrhoeae proteases. It is cleaved within the hinge sequence at the same sites as IgA1.
